目的建立液相色谱-串联质谱(LC-MS/MS)平台检测血浆游离变肾上腺素(MN)和去甲变肾上腺素(NMN)诊断嗜铬细胞瘤的临床策略。方法招募255例健康人以建立参考范围。另招募于复旦大学附属中山医院泌尿外科和内分泌科就诊患者,嗜铬细... 目的建立液相色谱-串联质谱(LC-MS/MS)平台检测血浆游离变肾上腺素(MN)和去甲变肾上腺素(NMN)诊断嗜铬细胞瘤的临床策略。方法招募255例健康人以建立参考范围。另招募于复旦大学附属中山医院泌尿外科和内分泌科就诊患者,嗜铬细胞瘤组110例、疾病对照组1 262例和健康对照组500例,比较各组间MN和NMN水平。依据参考范围和由受试者工作特征曲线(ROC)得到最佳特异性切点制定诊断策略。结果MN和NMN参考范围分别为〈64.97 pg/ml和〈185.83 pg/ml。嗜铬细胞瘤组MN和NMN水平显著高于疾病对照组(Z=-17.410,P=0.000;Z=-17.278,P=0.000)和健康对照组(Z=-16.434,P=0.000;Z=-16.439,P=0.000)。定义MN和NMN中任意一个超过参考范围上限为诊断策略A,MN和NMN中任意一个指标超过特异性诊断切点为诊断策略B,用于排除和辅助诊断嗜铬细胞瘤。结论建立LC-MS/MS平台检测MN和NMN的参考范围,为临床提供嗜铬细胞瘤的诊断策略。 展开更多

目的 :探讨嗜铬细胞瘤患者血浆间甲肾上腺素类物质(metanephrines,MNs)水平与其血压及糖脂代谢情况间的相关性。方法:纳入2005年6月至2013年9月我院内分泌科经病理证实的193例嗜铬细胞瘤患者,所有患者均接受血浆MNs及代谢指标检测。采用... 目的 :探讨嗜铬细胞瘤患者血浆间甲肾上腺素类物质(metanephrines,MNs)水平与其血压及糖脂代谢情况间的相关性。方法:纳入2005年6月至2013年9月我院内分泌科经病理证实的193例嗜铬细胞瘤患者,所有患者均接受血浆MNs及代谢指标检测。采用Pearson相关分析、多元线性回归和多元logistic回归分析MNs与血压、血糖及血脂等代谢指标间的相关性。结果:嗜铬细胞瘤患者中合并高血压者占83.4%;伴有明显的糖脂代谢异常,合并糖尿病及血脂异常者分别占35.8%和23.8%。Pearson相关分析显示,间甲肾上腺素(metanephrine,MN)与空腹血糖及糖负荷2 h血糖呈显著正相关(P<0.05);去甲变肾上腺素(normetanephrine,NMN)与收缩压、舒张压、空腹血糖、糖负荷2 h血糖、总胆固醇、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇及游离脂肪酸呈显著正相关(P<0.05)。多元线性回归分析表明,在校正年龄、性别、体质量指数、吸烟史及饮酒史后,MN与空腹血糖及游离脂肪酸仍显著正相关(P<0.05),NMN与收缩压、舒张压、空腹血糖、糖负荷2 h血糖、总胆固醇、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇及游离脂肪酸显著正相关(P<0.05)。多元logistic回归分析证实,MN和NMN水平升高的患者糖尿病患病风险增加,且NMN水平升高患者的高血压患病风险增加。结论:嗜铬细胞瘤患者血浆MN、NMN水平与血压及糖脂代谢调控密切相关,且NMN对代谢的影响更为显著。 展开更多

Chemical inducement and DDRT-PCR (differential display reverse transcription PCR) are adopted to investigate the effect of epinephrine (EPI) on the settlement and metamorphosis of Manila clam larvae. Chemical inducement shows that EPI has an effect to some extent on the metamorphosis of Manila clam larvae at all concentrations and in all treatments designed. The most significant result of inducement is obtained at the concentration of 10^-6 tool L^-1 and for 4 h. DDRT-PCR using six primer pairs shows that the gene expression pattern is quite different between EPI treatment and the control. Three hundred and forty-three amplification bands are obtained in total, among which, 67 (19.53%) are differentially appeared. Therefore, EPI has an effect on the gene expression of the eye spot larval Manila clam. It can be hypothesized that EPI is a settlement and metamorphosis inducer for Manila clam. EPI may lead to larvae settlement and metamorphosis by binding to the receptors on the membrane and then changing the gene expression of larvae cells. 展开更多

Low larval survival,poor settlement,and abnormal metamorphosis are major problems in seed production of donkey-ear abalone Haliotis asinina.We examined the effects of chemical cues including epinephrine,nor-epinephrine,and serotonin on larval survival,settlement,and metamorphosis in order to determine the possibility of using these chemicals to induce the problems.The results show that epinephrine could enhance metamorphosis rate at 10-6 mol/L only but higher concentrations(10-3-10-4 mol/L);and nor-epinephrine could inhibit the performance significantly,and serotonin could increase significantly the performance at a wide-range concentration(10-3-10-6 mol/L).Treatment with serotonin at 10-5 mol/L for 72 hours resulted in the highest settlement rate(42.2%) and survival rate(49.3%),while at 10-4 mol/L for 72 hours resulted in the highest metamorphosis rate(38.8%).Therefore,serotonin may be used as a fast metamorphosis inducer in abalone culture. 展开更多

Using the high-resolution Terahertz Time-domain spectroscopy (THz-TDS) and the standard sample pellet technique, the far-infrared vibrational spectra of clenbuterol hydrochloride (CH), a 2 -adrenergic agonist for decreasing fat deposition and enhancing protein accretion, were measured in temperature range of 77-295 K. Between 0.2 and 3.6 THz (6.6-120.0 cm-1 ), seven highly resolved spectral features, strong line-narrowing and a frequency blue-shift were observed with cooling. However, ractopamine hydrochloride, with some structural and pharmacological similarities to clenbuterol hydrochloride, showed no spectral features, indicating high sensitivity and strong specificity of THz-TDS. These results could be used for the rapid and nondestructive CH residual detection in food safety control. 展开更多

Objective The current study was aimed to investigate the correlations between immobility time in the forced swimming test (FST, a behavioral indicator of stress level) and hippocampal monoamine levels (markers of depression), plasma adrenalin level (a peripheral marker of stress) as well as fluoro-jade C staining (a marker of neurodegeneration). Methods Male Sprague-Dawley rats were subjected to acute, sub-chronic (7 d) or chronic (14 d) FSTs and immobility time was recorded. Levels of noradrenalin, serotonin and dopamine in the hippocampus, and adrenalin level in the plasma were quantified by high-performance liquid chromatography with electrochemical detection. Brain sections from rats after chronic forced swimming or rotenone treatment (3 mg/kg subcutaneously for 4 d) were stained with fluoro-jade C. Results The rats subjected to swimming stress (acute, sub-chronic and chronic) showed long immobility times [(214 ± 5), (220 ± 4) and (231 ± 7) s, respectively], indicating that the animals were under stress. However, the rats did not exhibit significant declines in hippocampal monoamine levels, and the plasma adrenalin level was not significantly increased compared to that in unstressed rats. The rats that underwent chronic swimming stress did not manifest fluoro-jade C staining in brain sections, while degenerating neurons were evident after rotenone treatment. Conclusion The immobility time in the FST does not correlate with markers of depression (monoamine levels) and internal stress (adrenalin levels and neurodegeneration), hence this parameter may not be a true indicator of stress level. 展开更多
