Ten positive porcine foot-and-mouth disease serums and more than two hundreds negative serum were tested, and the results were the same as the background of samples. The sensitivity test and replicate test indicated that this method was stable and sensitive, which was suitable for monitoring Asia 1 type porcine foot-and-mouth disease virus antibody.展开更多

In order to enhance geological body boundary visual effects in images and improve interpretation accuracy using gravity and magnetic field data, we propose an improved small sub-domain filtering method to enhance grav...In order to enhance geological body boundary visual effects in images and improve interpretation accuracy using gravity and magnetic field data, we propose an improved small sub-domain filtering method to enhance gravity anomalies and gravity gradient tensors. We discuss the effect of Gaussian white noise on the improved small sub-domain filtering method, as well as analyze the effect of window size on geological body edge recognition at different extension directions. Model experiments show that the improved small sub-domain filtering method is less affected by noise, filter window size, and geological body edge direction so it can more accurately depict geological body edges than the conventional small sub-domain filtering method. It also shows that deeply buried body edges can be well delineated through increasing the filter window size. In application, the enhanced gravity anomalies and calculated gravity gradient tensors of the Hulin basin show that the improved small sub-domain filtering can recognize more horizontal fault locations than the conventional method.展开更多

But gp85 amino acid sequence comparisons indicated that SDAU09C1 fell into subgroup A; it had homology of 88.8%-90.3% to 6 reference strains of subgroup A, much higher compared to other subgroups including subgroup B. Both the Siberian Crane(Leucogeranus leucogeranus) and Hooded Crane(Grus monacha) have limited population sizes and are considered endangered by domestic Chinese and international agencies.To document the current size...Both the Siberian Crane(Leucogeranus leucogeranus) and Hooded Crane(Grus monacha) have limited population sizes and are considered endangered by domestic Chinese and international agencies.To document the current size of their respective populations and characterize their groups,between October 2012 and April 2013 we undertook fieldwork at four nature reserve areas within the Poyang Lake wetlands.We divided Poyanghu National Nature Reserve(PYH) into the Wucheng(PWC) and Hengfeng areas(PHF),because each are each located in different counties.Our fieldwork showed that the Siberian Crane occurred mainly in PYH(364 in the PHF,158 in the PWC) and the Nanjishan Wetland National Nature Reserve(NJS,with 200 individuals).The Hooded Crane was mainly distributed in PYH(302 in the PHF and 154 in the PWC).Family groups accounted for more than 50% of the total number of groups among both species,with Hooded Cranes forming more family groups than Siberian Cranes.Typically,these groups were formed of two adults with one offspring(Siberian Crane),and two adults with two offspring(Hooded Crane),with the mean family group size of the Siberian Crane and Hooded Crane being respectively 2.65±0.53(n=43) and 3.09±0.86(n=47) individuals per group.The mean collective group size of the Siberian Crane and Hooded Crane included 28.09±24.94(n=23) and 28.94±27.97(n=16) individuals per group,respectively,with the proportion of juveniles among Hooded Cranes being more than double that seen among the Siberian Cranes.展开更多

It has also been confirmed that interindividual variations in IL-10 production are genetically attributed to the polymorphisms of IL-10 gene.Therefore, this study was designed to investigate whether the polymorphisms of IL-10 gene were associated with susceptibility to GC in the Chinese population.METHODS The serum levels of IL-10 were measured by radioimmunoassay. The single nucleotide polymorphisms (SNPs) at positions -1082A/G, -819T/C and -592A/C in the IL-10 gene promoter were analyzed using polymerase chain reactionrestriction fragment length polymorphism (PCR-RFLP).RESULTS 220 patients with gastric cancer and 180 healthy controls were included in this study. The serum levels of IL-10 were signifi cantly higher in GC patients than healthy controls (Z = -19.13, P 〈 0.001). Single SNP analysis showed that the -1082G allele, -1082AG and -819CC genotypes significantly increased in patients with GC (P = 0.029, 0.021, 0.039 respectively). In a logistic regression analysis adjusted for age and sex, the -1082AG genotype was associated with an odds ratio of 1.974 (95% CI,1.14-3.391; P = 0.014), and the -819CC genotype with an odds ratio of 2.496 (95% CI, 1.222-5.102; P = 0.012) for GC. Furthermore,haplotype analysis revealed that at least five haplotypes (ATA,ACC, GCC, ACA and ATC) were existent in this population.Also that the GCC haplotype was associated with a signifi cantly increased risk of GC as compared with the ATA haplotype (OR =1.90; 95% CI, 1.11-3.27; P = 0.02).CONCLUSION The results indicate that the gene haplotype of IL-10 may contribute to the susceptibility to GC in the Chinese population.展开更多

Methods The extracts of Dermatophagoides pteronyssinus were prepared with Coca's solution, lysis buffer of 2-DE, and Trizol reagent, respectively. Bicinchoninic acid (BCA) assay was used to determine the total protein concentration of the samples. The efficiency of different protein extraction methods were evaluated with 2-DE analysis. Results The concentrations of extracted protein by methods of Coca's solution, lysis buffer, and Trizol reagent were 0.63 g/L, 0.90 g/L, and 0.80 g/L, respectively. The 2-DE analysis results showed that some protein spots in low molecular weight (LMW) range could be detected with the Coca's solution method. With the lysis buffer of 2-DE method, more protein spots in LMW range could be detected, while the medium molecular weight (MMW) protein spots were absent. Several MMW protein spots (174-178 kD and 133 kD) and more LMW protein spots were detected withTrizol reagent method. Conclusions Among Coca's solution, lysis buffer of 2-DE, and Trizol reagent, the concentration of extracted protein of Dermatophagoides pteronyssinus by lysis buffer of 2-DE is the highest. However, most protein components of Dermatophagoides pteronyssinus purified mite bodies can be extracted by Trizol reagent, which may generally reflect the whole profile of Dermatophagoides pteronyssinus allergens.展开更多

METHODS: The express...AIM: To investigate the expression of different cytokeratins (CKs) in gastric epithelium of adult patients with chronic gastritis infected with Helicobacter pylori (H pylon) cagA + strains. METHODS: The expression of CK 7, 8, 18, 19 and 20 was studied immunohistochemically in antral gastric biopsies of 84 patients. All the CKs were immunostained in cagA+Hpylori gastritis (57 cases), non-Hpylori gastritis (17 cases) and normal gastric mucosa (10 cases). RESULTS: In cagA+ H pylori gastritis, CK8 was expressed comparably to the normal antral mucosa from surface epithelium to deep glands. Distribution of CK18 and CK 19 was unchanged, i.e. transmucosal, but intensity of the expression was different in foveolar region in comparison to normal gastric mucosa. Cytokeratin 18 immunoreactivity was significantly higher in the foveolar epithelium of H pylori-positive gastritis compared to both Hpylori-negative gastritis and controls. On the contrary, decrease in CK19 immunoreactivity occurred in foveolar epithelium of H pylori-positive gastritis. In both normal and inflamed antral mucosa without Hpyloriinfection, CK20 was expressed stronglyl moderately and homogenously in surface epithelium and upper foveolar region, but in H pylod -induced gastritis significant decrease of expression in foveolar region was noted. Generally, in both normal antral mucosa and H pylori-negative gastritis, expression of CK7 was not observed, while in about half cagA+ H pylori-infected patients, moderate focal CK7 immunoreactivity of the neck and coiled gland areas was registered, especially in areas with more severe inflammatory infiltrate. CONCLUSION: Alterations in expression of CK 7, 18, 19 and 20 together with normal expression of CK8 occur in antral mucosa of H pylori-associated chronic gastritis in adult patients infected with cagA+ strains. Alterations in different cytokeratins expression might contribute to weakening of epithelial tight junctions observed in H pylori-infected gastric mucosa.展开更多

A new sea eagle breeding population arose in the Treboǎ basin area in the early 1980' s. Until this time sea eagles had used former breeding places only for wintering, probably coming from the Baltic. The South Bohemian sea eagle population is very unique: it exists in a densely man-occupied landscape, mainly in areas with very intensive carp breeding in artificial fishponds and was partly artficially (help to wintering birds and reintroduction of some individuals) restored. Parasitic lampreys secrete anticoagulant from their buccal glands and prevent blood coagulation of host fishes. We identified a buccal gland secretory protein-2 (BGSP-2) from a buccal gland cDNA library of Larnpetrajaponica. The full-length BGSP-2 gene was cloned and the recombinant BGSP-2 protein was generated. The role of BGSP-2 on lymphocyte proliferation was studied by examining its effects on human T lymphocytes. We found that lamprey BGSP-2 was able to effectively block the proliferation of T cells in vitro by inducing G1/S cell cycle arrest. Furthermore, it inhibited the proliferation of hmnan T lymphocytes stimulated by phytohemagglutinin (PHA) at a minimum concentration of 0.1μg/ml. Our data suggest that lamprey BGSP-2 is able to block the mitosis of human T lymphocytes at the G1/S point, and has the potential of anti-proliferative effect on PHA-activated T lymphocytes.展开更多

In the bibliography, the most observable genre is Candida and less frequently ...In recent years, an increase in opportunistic fungal colonization in the oral cavity in immunocompetent patients (IC) has been observed. In the bibliography, the most observable genre is Candida and less frequently found are other opportunistic such as Aspergillus, Rhizopus, Cryptococcus, and others. The authors determined the presence of fungi in the oral cavity of IC students, and their relationship with the concentration of secretory IgA in saliva. To this end, we collected 50 samples of oral cavity swabs, which underwent direct examination and culture in Sabouraud dextrose agar with chloramphenicol. For its identification, CHROMagar Candida and API Candida (BioMerieux) were used. We obtained nine positive cultures (7 Candida albicans and 2 Saccharomyses cerevisiae), which represented 18% of the studied population. Throughout radial immunodiffusion (RID plates-PLATE), we determined the concentration of secretory IgA. No relationship was observed between the colonized group and group that was not colinized. The colonization rate found is below the one described in the bibliography (30% to 50%). However, these opportunistic fungi cause transitory colonization with no clinical relevance in IC patients and, its percentage can vary according to the studied population.展开更多

Oligomeric proteins are often stabilized by aromatic clusters located within the subunit interfaces. In the present study, the authors constructed five variants of Ps3aHSD (Pseudomonas sp. B-0831 3a-hydroxysteroid dehydrogenase) in which one or two residues at the dimer interface were replaced with aromatic residues, and examined the effects of introducing aromatic residues in this region on protein thermostability. Under their experimental conditions, all variants formed dimers, similar to wild-type Ps3aHSD. Thermal denaturation experiments indicated that Tm of all variants was 0.2-16.2 °C lower than that of wild-type protein, indicating less stable thanwild-type protein. The results collectively suggest that aromatic residues of natural oligomeric proteins are strictly posted in the interface to facilitate optimal interactions and avoid conformational strain.展开更多
AIM: To investigate whether the changes of gap junction gene connexin messenger RNA in the noncancerous liver tissue of patients with hepatocellular carcinoma (HCC) could play a significant role in its postresection r...AIM: To investigate whether the changes of gap junction gene connexin messenger RNA in the noncancerous liver tissue of patients with hepatocellular carcinoma (HCC) could play a significant role in its postresection recurrence.METHODS: Seventy-nine consecutive patients having undergone curative resection for HCC entered this study.Using a reverse-transcription polymerase chain reaction (RT-PCR)-based assay, connexin (Cx) 26, connexin (Cx)32 and connexin (Cx) 43 mRNAs were determined prospectively in noncancerous liver tissues from these 79 patients and in the liver tissues from 15 controls. The correlations between connexin mRNA expression and the clinicopathological variables and outcomes (tumor recurrence and recurrence related mortality) were studied.RESULTS: Compared with liver tissues of control patients,the expression of Cx 32 mRNA in noncancerous liver tissues was significantly lower (mean: 0.715 vscontrol 1.225,P<0.01), whereas the decreased Cx 26 mRNA (mean:0.700 vs of control 1.205,P>0.05) and increased Cx 43 mRNA (mean: 0.241 vscontrol 0.100, P>0.05) had no statistical significance. We defined the value of Cx 32 mRNA or Cx 26mRNA below 0.800 as a lower value. By multivariate analysis for noncancerous livers, a lower value of Cx 32 mRNA correlated significantly with a risk of HCC recurrence and recurrence-related mortality. The lower value of Cx 26 mRNA did not correlate with recurrence and mortality. The increased value of Cx43 mRNA also did not correlate with postoperative recurrence and recurrence-related mortality. By multivariate analysis, other significant predictors of HCC recurrence included vascular permeation, cellular dedifferentiation, and less encaps-ulation. The other significant parameter of recurrence related mortality was vascular permeation.CONCLUSION: The decreased expression of Cx 32 mRNA in noncancerous liver tissues plays a significant role in the prediction of postoperative recurrence of HCC.展开更多

Conclusions Among Coca's solution, lysis buffer of 2-DE, and Trizol reagent, the concentration of extracted protein of Dermatophagoides pteronyssinus by lysis buffer of 2-DE is the highest. However, most protein components of Dermatophagoides pteronyssinus purified mite bodies can be extracted by Trizol reagent, which may generally reflect the whole profile of Dermatophagoides pteronyssinus allergens.

文摘AIM: To investigate the expression of different cytokeratins (CKs) in gastric epithelium of adult patients with chronic gastritis infected with Helicobacter pylori (H pylon) cagA + strains. METHODS: The expression of CK 7, 8, 18, 19 and 20 was studied immunohistochemically in antral gastric biopsies of 84 patients. All the CKs were immunostained in cagA+Hpylori gastritis (57 cases), non-Hpylori gastritis (17 cases) and normal gastric mucosa (10 cases). RESULTS: In cagA+ H pylori gastritis, CK8 was expressed comparably to the normal antral mucosa from surface epithelium to deep glands. Distribution of CK18 and CK 19 was unchanged, i.e. transmucosal, but intensity of the expression was different in foveolar region in comparison to normal gastric mucosa. Cytokeratin 18 immunoreactivity was significantly higher in the foveolar epithelium of H pylori-positive gastritis compared to both Hpylori-negative gastritis and controls. On the contrary, decrease in CK19 immunoreactivity occurred in foveolar epithelium of H pylori-positive gastritis. In both normal and inflamed antral mucosa without Hpyloriinfection, CK20 was expressed stronglyl moderately and homogenously in surface epithelium and upper foveolar region, but in H pylod -induced gastritis significant decrease of expression in foveolar region was noted. Generally, in both normal antral mucosa and H pylori-negative gastritis, expression of CK7 was not observed, while in about half cagA+ H pylori-infected patients, moderate focal CK7 immunoreactivity of the neck and coiled gland areas was registered, especially in areas with more severe inflammatory infiltrate. CONCLUSION: Alterations in expression of CK 7, 18, 19 and 20 together with normal expression of CK8 occur in antral mucosa of H pylori-associated chronic gastritis in adult patients infected with cagA+ strains. Alterations in different cytokeratins expression might contribute to weakening of epithelial tight junctions observed in H pylori-infected gastric mucosa.
文摘The population density of the white-tailed sea eagle Haliaeetus albicilla is very low in many countries. In last twenty years, the sea eagle population in South Bohemia was restored by strict protection subsidized by reintroduction, The active help consisted of feeding during winter and building of artificial nests. A new sea eagle breeding population arose in the Treboǎ basin area in the early 1980' s. Until this time sea eagles had used former breeding places only for wintering, probably coming from the Baltic. The South Bohemian sea eagle population is very unique: it exists in a densely man-occupied landscape, mainly in areas with very intensive carp breeding in artificial fishponds and was partly artficially (help to wintering birds and reintroduction of some individuals) restored. The experience from South Bohemia may have importance for populations of the sea eagle in other areas of its occurence, primarily in the continental conditions [ Current Zoology 55 (5) : 315 - 318, 2009].

文摘Lamprey is a representative of the agnathans, the most ancient class of vertebrates. Parasitic lampreys secrete anticoagulant from their buccal glands and prevent blood coagulation of host fishes. We identified a buccal gland secretory protein-2 (BGSP-2) from a buccal gland cDNA library of Larnpetrajaponica. The full-length BGSP-2 gene was cloned and the recombinant BGSP-2 protein was generated. The role of BGSP-2 on lymphocyte proliferation was studied by examining its effects on human T lymphocytes. We found that lamprey BGSP-2 was able to effectively block the proliferation of T cells in vitro by inducing G1/S cell cycle arrest. Furthermore, it inhibited the proliferation of hmnan T lymphocytes stimulated by phytohemagglutinin (PHA) at a minimum concentration of 0.1μg/ml. Our data suggest that lamprey BGSP-2 is able to block the mitosis of human T lymphocytes at the G1/S point, and has the potential of anti-proliferative effect on PHA-activated T lymphocytes.
文摘In recent years, an increase in opportunistic fungal colonization in the oral cavity in immunocompetent patients (IC) has been observed. In the bibliography, the most observable genre is Candida and less frequently found are other opportunistic such as Aspergillus, Rhizopus, Cryptococcus, and others. The authors determined the presence of fungi in the oral cavity of IC students, and their relationship with the concentration of secretory IgA in saliva. To this end, we collected 50 samples of oral cavity swabs, which underwent direct examination and culture in Sabouraud dextrose agar with chloramphenicol. For its identification, CHROMagar Candida and API Candida (BioMerieux) were used. We obtained nine positive cultures (7 Candida albicans and 2 Saccharomyses cerevisiae), which represented 18% of the studied population. Throughout radial immunodiffusion (RID plates-PLATE), we determined the concentration of secretory IgA. No relationship was observed between the colonized group and group that was not colinized. The colonization rate found is below the one described in the bibliography (30% to 50%). However, these opportunistic fungi cause transitory colonization with no clinical relevance in IC patients and, its percentage can vary according to the studied population.
文摘The design of variants to enhance conformational stability of proteins is an important aspect of protein engineering. Oligomeric proteins are often stabilized by aromatic clusters located within the subunit interfaces. In the present study, the authors constructed five variants of Ps3aHSD (Pseudomonas sp. B-0831 3a-hydroxysteroid dehydrogenase) in which one or two residues at the dimer interface were replaced with aromatic residues, and examined the effects of introducing aromatic residues in this region on protein thermostability. Under their experimental conditions, all variants formed dimers, similar to wild-type Ps3aHSD. Thermal denaturation experiments indicated that Tm of all variants was 0.2-16.2 °C lower than that of wild-type protein, indicating less stable thanwild-type protein. The results collectively suggest that aromatic residues of natural oligomeric proteins are strictly posted in the interface to facilitate optimal interactions and avoid conformational strain.

文摘AIM: To investigate whether the changes of gap junction gene connexin messenger RNA in the noncancerous liver tissue of patients with hepatocellular carcinoma (HCC) could play a significant role in its postresection recurrence.METHODS: Seventy-nine consecutive patients having undergone curative resection for HCC entered this study.Using a reverse-transcription polymerase chain reaction (RT-PCR)-based assay, connexin (Cx) 26, connexin (Cx)32 and connexin (Cx) 43 mRNAs were determined prospectively in noncancerous liver tissues from these 79 patients and in the liver tissues from 15 controls. The correlations between connexin mRNA expression and the clinicopathological variables and outcomes (tumor recurrence and recurrence related mortality) were studied.RESULTS: Compared with liver tissues of control patients,the expression of Cx 32 mRNA in noncancerous liver tissues was significantly lower (mean: 0.715 vscontrol 1.225,P<0.01), whereas the decreased Cx 26 mRNA (mean:0.700 vs of control 1.205,P>0.05) and increased Cx 43 mRNA (mean: 0.241 vscontrol 0.100, P>0.05) had no statistical significance. We defined the value of Cx 32 mRNA or Cx 26mRNA below 0.800 as a lower value. By multivariate analysis for noncancerous livers, a lower value of Cx 32 mRNA correlated significantly with a risk of HCC recurrence and recurrence-related mortality. The lower value of Cx 26 mRNA did not correlate with recurrence and mortality. The increased value of Cx43 mRNA also did not correlate with postoperative recurrence and recurrence-related mortality. By multivariate analysis, other significant predictors of HCC recurrence included vascular permeation, cellular dedifferentiation, and less encaps-ulation. The other significant parameter of recurrence related mortality was vascular permeation.CONCLUSION: The decreased expression of Cx 32 mRNA in noncancerous liver tissues plays a significant role in the prediction of postoperative recurrence of HCC.
