[Method] Chalconesynthase(CHS) gene was cloned from Morus(Moraceae) in silico. The amino acid sequence, physical and chemical properties, transmembrane structural domain, hydrophobicity/hydrophilicity,subcellular localization, secondary and tertiary structure of protein were predicted and analyzed by bioinformatics tools. [Result] The cDNA sequence of CHS gene was 1 365bp by splicing using the software DNAstar and it contained a complete ORF including 1 170 bp which encoded 389 amino acids. Bioinformatic analysis showed that CHS gene included specific peptide sequence RLMMYQQGCFAGGTVLR of chalcone synthase superfamily, but has no signal peptide, belonging to the non-secretory proteins, located inside of cytoplasm. Its molecular evolution is more conservative.[Conclusion] The results above provided foundation for the further studies of structure and function of CHS protein.展开更多

The winding layer is composed of two monolayers where steel wires are cross helically wound. An analytical procedure is developed to predict the short-term burst pressure of PSP as the monolayer is assumed to be elastic and orthotropic. The 3D anisotropic elasticity and Maximum Stress Failure Criterion are employed in the formulation of the elasticity problem. Good agreement between the theoretical results and the experimental data shows that the proposed approach can well predict the short-term burst pressure of PSP.展开更多

Adsorption experiments were performed by adding the resins to the serum. The results showed that this type of adsorbent could cut down the concentration of triglyceride in serum by 56. 4% (3. 35mg/gresin ) at most, while concentration of the total protein (TP ) decreased only by 10. 9% at least, so this novel adsorbent can be used to cure hypertriglyceridemia by hemoperfusion in the future.展开更多

The dimensional accuracy was selected as the objective function. By investigating the rheological properties of resin film, the compaction behavior of fiber preform and characteristics of RFI process, an optimal mathematical model was established, it was found that the numerical results obtained from the RFICOMP program package have good consistency with the experimental results, and this optimization procedure can be applied to other composites manufacture processes.展开更多

The process conditions were optimized by orthog...Calcium lignosulphonate was used to synthesize a spherical lignosulphonate resin in a cheap and non-toxic disperse medium by reversed phase suspension polymerization. The process conditions were optimized by orthogonal experiments. Under .the optxmal conditxons (T=95 ℃, CHCl= 3 mol·L^-1, mHCHO: mCLS=7%, WCLS=50%), globulation took about 20 min and the product was featured with excellent spherical shape, narrow particle size range, 61.20% of water retention capacity, 0.83 mmol·ml^- 1 of total volume exchange capacity and 3.46 mmol·g^- 1 of total exchange capacity. The results of Scanning Electron Micrograph and Scanning Probe Micrograph indicate that spherical lignosulphonate resin has a rugged surface with porous microstructure in the gel skeleton. The average pore size of dry samples was determined to be 10.46 nm. by the BET method.展开更多

[Method] With total RNA extracted from buckwheat as the template, CHS cD- NA sequence was cloned from buckwheat by using RACE technology and CODEHOP primer design method, the full length gene was obtained by primers which were de- signed for amplification of full-length gene sequence with buckwheat DNA template. Clustalxl.81 and MEGA4 software were used for sequence analysis and construction of phylogenetic tree; NCBI Blastn and Biastp programs were applied for homology analysis of nucleic acid and protein. [Result] Bioinformatics analysis showed that the full length of this gene is 1 906 bp, containing a 463 bp intron sequence and a 1 188 bp coding region, encoding 395 amino acids. Blastn sequence alignment revealed that the CHS gene sequence obtained in this study shared 86% homology with the CHS gene of closely related species. [Conclusion] This study laid the foundation to clarify molecular basis of the synthesis of buckwheat bioflavanoids and explore an effective way to improve the content of buckwheat bioflavanoids.展开更多

Then numeric simulation is employed as experimental method, and the Taguchi method is used to analyze significance of effect of process parameters on the cell size. At last the process parameters are focused on melt temperature, injection time, mold temperature and pretidied volume. The significance order from big to small of the effect of each process parameters on cell size is melt temperature, pre-filled volume, injection time, and mold temperature. On the basis of above research, the effect of each process parameter on cell size is further researched. Appropriate reduction of the melt temperature and increase of the pre-filled volume can optimize the cell size effectively, while the effects of injection time and mold temperature on cell size are less significant.展开更多

However, its formation mechanism has not been explored until now. With the application of experimental equipment designed by the authors named “Constant Velocity Type Dynamic Rheometer of Capillary” or (CVDRC), we were able to analyze in detail the whole extrusion process of a polymer melt. We did this after superimposing a sine vibration of small amplitude parallel to the extruding direction of the polymer melt. Then, we created a calculation model to determine the shear stress at the wall of the capillary using a superimposed vibration. We also determined the calculation steps needed to establish the afore-mentioned shear stress. Through measurement and analysis, the instantaneous entry pressure of the capillary, the pressure gradient, and the shear stress of the polymer melt within the capillary under vibration force field can be calculated.展开更多

It was found by GC/MS analysis that the symmetrical ketone, such as acetone, in the Mannich reaction mainly produces one Mannich base; while the asymmetrical ketone, such as 2-pentanone, gives two Mannich bases. The reactivity depends on the tereo-hinder of both ketones and amines.展开更多
