目的探讨改良同步化G显带及染色体微阵列分析(chromosomal microarray analysis,CMA)技术在脐血产前诊断中的应用价值。方法选取2020年1月至2021年4月在我院优生遗传科就诊的具有产前诊断指征且错过绒毛、羊水产前诊断的孕妇202例,采集... 目的探讨改良同步化G显带及染色体微阵列分析(chromosomal microarray analysis,CMA)技术在脐血产前诊断中的应用价值。方法选取2020年1月至2021年4月在我院优生遗传科就诊的具有产前诊断指征且错过绒毛、羊水产前诊断的孕妇202例,采集脐血标本,进行常规法及改良同步化G显带染色体核型分析,并同时采用CMA技术对脐血DNA的全基因组拷贝数变异(copy number variant,CNV)进行检测。比较改良同步化、CMA检测与常规法G显带核型条带分辨率、异常检出率。结果改良同步化G显带带纹的分辨达到500~550条带,常规法G显带带纹的分辨仅达到320~400条带。常规法、改良同步化G显带和CMA染色体异常检出率分别为3.47%、4.46%和9.90%。CMA检出的16例CNVs中,改良同步化G显带仅检出2例>5 Mb微缺失;此外,另有3例染色体核型异常但CMA检测为正常。改良同步化G显带与CMA染色体异常检出率比较,差异有统计学意义(χ^(2)=4.495,P=0.034)。结论采用改良同步化染色体G显带核型分辨率可提高至500~550条带,有利于发现更多的染色体结构异常,联合CMA技术在脐血产前染色体病诊断实现优势互补,提高染色体异常检出率,可降低新生儿出生缺陷。 展开更多

目的评价改良同步化染色体培养技术在脐血产前诊断核型分析中的应用。方法选择2011年6月-2012年12月就诊的1 200例有产前诊断指征的孕妇,抽脐血进行产前诊断,用改良同步化染色体培养技术与常规法进行核型分析。结果培养成功率均为100.00... 目的评价改良同步化染色体培养技术在脐血产前诊断核型分析中的应用。方法选择2011年6月-2012年12月就诊的1 200例有产前诊断指征的孕妇,抽脐血进行产前诊断,用改良同步化染色体培养技术与常规法进行核型分析。结果培养成功率均为100.00%。改良同步化染色体培养技术染色体核型达到500~550条带,常规法染色体核型为320~400条带。改良同步化染色体培养技术的染色体长度及分辨率明显提高。对1 200份标本进行核型分析,检出染色体数目、结构异常105例(8.75%),其中染色体结构异常69例(5.75%),染色体数目异常36例(3.00%)。结论改良同步化染色体培养技术操作简单,方法稳定,获得的分裂相比较分散,核型分辨率可提高至500~550条带,应用于脐血产前诊断染色体核型分析有利于发现更多的染色体结构异常,在产前诊断中具有重要的临床价值。 展开更多

The observation data from ground surface meteorological stations is an important basis on which climate change research is carried out, while the homogenization of the data is necessary for improving the quality and homogeneity of the time series. This paper reviews recent advances in the techniques of identifying and adjusting inhomogeneity in climate series. We briefly introduce the results of applying two commonly accepted and well-developed methods (RHtest and MASH) to surface climate observations such as temperature and wind speed in China. We then summarize current progress and problems in this field, and propose ideas for future studies in China. Along with collecting more detailed metadata, more research on homogenization technology should be done in the future. On the basis of comparing and evaluating advantages and disadvantages of different homogenization methods, the homogenized climate data series of the last hundred years should be rebuilt. 展开更多

Cardiac resynchronization therapy (CRT) is associated with a favorable outcome only in patients with left bundle branch block (LBBB) pattern and in patients with a QRS duration 〉 150 ms, in patients with non-LBBB pattern with a QRS duration of 120-150 ms usually is not beneficial. After adjusting for QRS duration, QRS morphology was no longer a determinant of the clinical response to CRT. In contrast to the mainstream view, we hypothesized that the unfavorable CRT outcome in patients with non-LBBB and a QRS duration of 120-150 ms is not due to the QRS morphology itself, but to less dyssynchrony and unfavorable patient characteristics in this subgroup, such as more ischemic etiology and greater prevalence of male patients compared with patients with LBBB pattern. Further, the current CRT technique is devised to eliminate the dyssynchrony present in patients with LBBB pattern and inappropriate to eliminate the dyssynchrony in patients with non-LBBB pattern. We also hypothesized that electrocardiography may also provide information about the presence of interventricular and left intraventricular dyssynchrony and the approximate location of the latest activated left ventricular (LV) region. To this end, we devised new ECG criteria to estimate interventricular and LV intraventricular dyssynchrony and the approximate location of the latest activated LV region. Our preliminary data demonstrated that the latest activated LV region in patients with nonspecific intraventricular conduction disturbance (NICD) pattern might be at a remote site from that present in patients with LBBB pattern, which might necessitate the invention of a novel CRT technique for patients with NICD pattern. The application of the new interventricular and LV intraventricular dyssynchrony ECG criteria and a potential novel CRT technique might decrease the currently high nonresponder rate in patients with NICD pattern. 展开更多

The expression of mRNA for bone morphogenetic protein-1 (BMP-1) was investigated in fetal human cartilage and bone during the developing endochondral ossification by using in situ hybridization. BMP-1 mRNA was localized in the hypertrophic chondrocytes, the cells of perichondrium, osteoblasts and marrow cells. The results suggest that BMP-1 may involve in cartilage and bone formation, and in regulation of calcification of growth plate.As a comparative study, the expression of TGF-β1 protein was investigated in cartilage and bone using immunohistochemical staining, correlated expression of TGF-β1 and BMP-1 was found. The results suggest that BMP-1 and TGF-β1 act synergistically to influence endochrondral ossification. 展开更多

In this paper, it is firstly proved that the oscifiator network with nonlinear coupling is also eventually dissipative under the hypothesis of eventual dissipation of the uncoupled oscillators. And the dynamics of the network is analyzed in its absorbing domain by combining two methods developed recently. Suufficient conditions for synchronization in the oscillator networks with nonlinear coupling are obtained. The two methods are combined effectively and the results embody the respective merits of the two methods. Numerical simulations confirm the validity of the results. 展开更多
