Embryos were derived from artificial fertilization of frogs’ eggs, and the...We investigated the early embryonic and larval development of the concave-eared torrent frogs, Odorrana tormota (Amphibia, Anura, Ranidae). Embryos were derived from artificial fertilization of frogs’ eggs, and the staging of development was based on morphological and physiological characteristics. Two major periods of development were designated: i) early embryonic period, from fertilization to operculum completion stage, lasted for 324 h at water temperature (WT) 18 ?23℃; ii) larval period, from operculum completion stage to tail absorbed stage, took 1207 h at WT 20 ? 24℃. Tadpoles of the concave-eared torrent frogs showed no evidence of abdominal sucker. Absence of this key characteristic supports the view from molecular systematics that concave-eared torrent frog does not belong to the genus Amolops. Two cleavage patterns were observed in embryos at 8-cell and 16-cell stages, with Pattern I2 (latitudinal cleavage at the 8-cell stage, and meridional cleavage at the 16-cell stage with two perpendicular meridional furrows) being the predominant pattern and only 1.5% belonging to Pattern II (meridional cleavage at the 8-cell stage and latitudinal cleavage at the 16-cell stage). The factors affecting cleavage and hatching ratios, developmental speed, and ecological adaptation were discussed.展开更多

METHODS: Incorporation of 59Fe into mucosal and luminal proteins was carri...AIM: To elucidate the sequential transfer of iron amongst ferritin, transferrin and transferrin receptor under various iron status conditions. METHODS: Incorporation of 59Fe into mucosal and luminal proteins was carried out in control WKY rats. The sequential transfer of iron amongst ferritin, transferrin and transferrin receptor was carried out in iron deficient, control and iron overloaded rats. The duodenal proteins were subjected to immunoprecipitation and quantitation by specific ELISA and in situ localization by microautoradiography and immunohistochemistry in tandem duodenal sections. Human duodenal biopsy (n = 36) collected from subjects with differing iron status were also stained for these proteins. RESULTS: Ferritin was identified as the major protein that incorporated iron in a time-dependent manner in the duodenal mucosa. The concentration of mucosal ferritin was significantly higher in the iron excess group compared to control, iron deficient groups (731.5 ± 191.96 vs 308.3 ± 123.36, 731.5 ± 191.96 vs 256.0 ± 1.19, P < 0.005), while that of luminal transferrin which was significantly higher than the mucosal did not differ among the groups (10.9 ± 7.6 vs 0.87 ± 0.79, 11.1 ± 10.3 vs 0.80 ± 1.20, 6.8 ± 4.7 vs 0.61 ± 0.63, P < 0.001). In situ grading of proteins and iron, and their superimposition, suggested the occurrence of a sequential transfer of iron. This was demonstrated to occur through the initial binding of iron to luminal transferrin then to absorptive cell surface transferrin receptors. The staining intensity of these proteins variedaccording to the iron nutrition in humans, with intense staining of transferrin receptor observed in iron deficient subjects. CONCLUSION: It is concluded that the intestine takes up iron through a sequential transfer involving interaction of luminal transferrin, transferrin-transferrin receptor and ferritin.展开更多
AIM:To investigate trefoil factor(TFF) gene copy number,mRNA and protein expression as potential biomarkers in cholangiocarcinoma(CCA).METHODS:TFF mRNA levels,gene copy number and protein expression were determined re...AIM:To investigate trefoil factor(TFF) gene copy number,mRNA and protein expression as potential biomarkers in cholangiocarcinoma(CCA).METHODS:TFF mRNA levels,gene copy number and protein expression were determined respectively by quantitative reverse transcription polymerase chain reaction(PCR),quantitative PCR and immunohistochemistry in bile duct epithelium biopsies collected from individuals with CCA,precancerous bile duct dysplasia and from disease-free controls.The functional impact of recombinant human(rh) TFF2 peptide treatment on proliferation and epidermal growth factor receptor(EGFR) /mitogenactivated protein kinase(MAPK) signaling was assessed in the CCA cell line,KMBC,by viable cell counting and immunoblotting,respectively.RESULTS:TFF1,TFF2 and TFF3 mRNA expression was significantly increased in CCA tissue compared to disease-free controls,and was unrelated to gene copy number.TFF1 immunoreactivity was strongly increased in both dysplasia and CCA,whereas TFF2 immunoreactivity was increased only in CCA compared to diseasefree controls.By contrast,TFF3 immunoreactivity was moderately decreased in dysplasia and further decreased in CCA.Kaplan-Meier analysis found no association of TFF mRNA,protein and copy number with age,gender,histological subtype,and patient survival time.Treatment of KMBC cells with rhTFF2 stimulated proliferation,triggered phosphorylation of EGFR and downstream extracellular signal related kinase(ERK),whereas co-incubation with the EGFR tyrosine kinase inhibitor,PD153035,blocked rhTFF2-dependent proliferation and EGFR/ERK responses.CONCLUSION:TFF mRNA/protein expression is indicative of CCA tumor progression,but not predictive for histological sub-type or survival time.TFF2 is mitogenic in CCA via EGFR/MAPK activation.展开更多

In Saudi Arabia, heavy metals contamination may result from petroleum and mining operations, refining ores, sludge, waste treatment, electrical equipment, paints, alloys, pesticides, batteries and fuel transportation. Microbial processes lead to appreciable and even complete remediation of heavy metals contaminated environments. The chief ways, by which such remediation may be accomplished, include biosorption, bioaugmentation, bioventing, biostimulation, bioaccumulation, biosolubilization, bioreduction, bioprecipitation, mineralization and methylation. Other technologies and methods are fully developed and now are being used in practice, such as heavy metals nanotechnology bioremediation. An area of fungal biotechnology currently in vogue is the use of fungal biornass to absorb metal ions from contaminated solutions. Such biological approaches of metal ions recovery can be used to clean up polluted effluents or to recover precious metal ions from solutions. The present review provides information on fungal bioremediation of heavy metal contamination for use in future studies in Saudi Arabia as well as in the Arabian Gulf Region.展开更多
Objective:The aim of this study was to observe and compare the antiemetic effectiveness and adverse events of magnetotherapy plus 5-hydroxytryptamine (5-HT3) receptor inhibitor granisetron hydrochloride vs granisetron...Objective:The aim of this study was to observe and compare the antiemetic effectiveness and adverse events of magnetotherapy plus 5-hydroxytryptamine (5-HT3) receptor inhibitor granisetron hydrochloride vs granisetron hydrochloride alone in patients with chemotherapy.Methods:Sixty-four patients were randomized to receive either granisetron hydrochloride alone (control group:granisetron hydrochloride 3 mg intervenous infusion before chemotherapy, from the 1st day of chemotherapy course until the day after chemotherapy course completed) or magnetotherapy plus granisetron hydrochloride (treatment group:the same granisetron hydrochloride regimen plus rotatory magnetotherapy of 1 every day after chemotherapy for 5 begin with chemotherapy).Baseline characteristics were similar in both groups.The patients' emesia was evaluated according to WHO's criteria.The density of 5-HT3 was detected by enzyme-linked immunosorbent assay (ELISA).Results:In the treatment of acute vomiting, there was no significant difference between two groups (P>0.05), but in the treatment of tardive vomiting, the effectiveness in treatment group was better than that in control group (P<0.05).The density of 5-HT3 in treatment group and control group were (225.32±57.29) ng/mL vs (213.00 ±53.29) ng/mL before chemotherapy and (273.88 ±75.42) ng/mL vs (313.17±76.36) ng/mL after chemotherapy (P<0.01); the rate of adverse events was 36.36% and 48.39% respectively in treatment group and control group (P>0.05).Conclusion:Magnetotherapy plus granisetron hydrochloride provide better effectiveness than granisetron hydrochloride alone, and both therapies have synergistic effect.The adverse events didn't raised in treatment group.展开更多

Methods A total of 170 mal...Objective To investigate the effect of phenylephrine (an α-adrenergic agonist) on alveolar fluid clearance (AFC) in ventilator-induced lung injury and the possible mechanism involved. Methods A total of 170 male Wistar rats were randomly allocated into 17 groups (n= 10) using ran- dom number tables. Short-term (40 minutes) mechanical ventilation with high tidal volume (HVT) was per- formed to induce lung injury, impair active Na+ transport and lung liquid clearance in the rats. Unventilated rats served as controls. To demonstrate the effect of phenylephrine on AFC, phenylephrine at different con- centrations (1×10^-5, 1 ×10^-6, 1×10^-7, 1×10^-8, and 1×10^-9 mol/L) was injected into the alveolar space of the HVT ventilated rats. To identify the influence of adrenergic antagonists, Na+ channel, and microtubular sys- tem on the effect of phenylephrine, phenylephrine at 1×10^-5mol/L combined with prazosin (an α1-adrener- gic antagonist, 1×10^-4 mol/L), yohimbine (an α2-adrenergic antagonist, 1×10^-4 mol/L), atenolol (a β1- adrenergic antagonist, 1×10^-5 mol/L), ICI- 118551 (an β2-adrenergic antagonist, 1×10^-5 mol/L), amiloride (a Na+ channel blocker, 51×10^-4mol/L), ouabain (a Na+/K+-ATPase blocker, 5~×10^-4mol/L), colchicine (a mi- crotubular disrupting agent, 0.25 mg/100 g body weight), or β-lumicolchicine (an isomer of colchicine, 0.25 mg/100 g body weight) were perfused into the alveolar space of the rats ventilated with HVT for 40 minutes. AFC and total lung water content were measured. Results Basal AFC in control rats was (17.47±2.56)%/hour, which decreased to (9.64± 1.32)%/hour in HVT ventilated rats (P=0.003). The perfusion of phenylephrine at 1 ×10^-8, 1×10^-7, 1×10^-6, and 1×10^-5 mol/L significantly increased the AFC in HVT ventilated rats (all P〈0.05). This effect of phenylephrine on AFC was suppressed by prazosin, atenolol, and ICI-118551 in HVT ventilated rats by 53%, 31%, and 37%, respectively (all P〈0.05). The AFC-stimulating effect of phenylephrine was lowered by 33% and 42% with amiloride and ouabain, respectively (both P〈0.05). Colchicine significantly inhibited the effect of phenylephrine (P=0.031). Conclusion Phenylephrine could increase the AFC in HVT-ventilated rats and accelerate the ab- sorption of pulmonary edema.展开更多

Methods Human B cell li...Objective To investigate the role of TNF receptor-associated factor 2 (TRAF-2) and TRAF6 in CD40-induced nuclear factor-κB (NF-κB) signaling pathway and whether CD40 signaling requires TRAF2. Methods Human B cell lines were transfected with plasmids expressing wild type TRAF2 or dominant negative TRAF2,TRAF2-shRNA,or TRAF6-shRNA. The activation of NF-κB was detected by Western blot,kinase assay,transfactor enzyme-linked immunosorbent assay (ELISA),and fluorescence resonance energy transfer (FRET). Analysis of the role of TRAF-2 and TRAF-6 in CD40-mediated NF-κB activity was examined following stimulation with recombinant CD154. Results TRAF2 induced activity of IκB-kinases (IKKα,IKKi/ε),phosphorylation of IκBα,as well as nuclear translocation and phosphorylation of p65/RelA. In contrast,TRAF6 strongly induced NF-κB activation and nuclear translocation of p65 as well as p50 and c-Rel. Engagement of CD154-induced nuclear translocation of p65 was inhibited by a TRAF6-shRNA,but conversely was enhanced by a TRAF2-shRNA. Examination of direct interactions between CD40 and TRAFs by FRET documented that both TRAF2 and TRAF6 directly interacted with CD40. However,the two TRAFs competed for CD40 binding. Conclusions These results indicate that TRAF2 can signal in human B cells,but it is not essential for CD40-mediated NF-κB activation. Moreover,TRAF2 can compete with TRAF6 for CD40 binding,and thereby limit the capacity of CD40 engagement to induce NF-κB activation.展开更多

There is also evidence that the expression of male sexual ornamentation and associated sperm characteristics depend on an indi- vidual's ability to cope with oxidative stress. Carotenoids are known for their antioxidant properties and carotenoid-based orna- ments might represent honest signals as these pigments can be traded offbetween the investment in sexual ornamentation, sperm function as well as immune response. In this study, we examined the relationship between sexual ornamentation (breeding colo- ration) and sperm characteristics (e.g., velocity and morphology) in the three-spined stickleback Gasterosteus aeuleatus, an ex- ternally fertilising fish species, in which sperm competition commonly occurs. During the breeding season males are sperm limi- ted and develop a conspicuous carotenoid-based coloration, which is under strong pre-copulatory sexual selection due to female mate choice and male-male competition. The results of the present study show that the expression of stickleback male breeding coloration is significantly positively associated with the linearity of sperm movement, whereas sperm morphology (head length to tail length ratio) is significantly negatively related to the trajectory of sperm movement. Moreover, there is some support for the phenotype-linked fertility hypothesis as the intensity of male red breeding coloration is significantly positively correlated with sperm velocity, which is supposed to be an important determinant of fertilisation success in external fertilisers, indicating the honesty of the sexually selected nuptial red coloration.展开更多

Methods: On Octob...Objective: To provide the experience on medical rescue and care for any giant earthquake by analysing the patients treated by the Chinese Medical Rescue Team in the Barakott earthquake of Pakistan. Methods: On October 8, 2005, a giant earthquake (7.8 Richter scale) earthquake occurred in the northwest Pakistan. We belonged to the Chinese Medical Rescue Team and worked in the earthquake-affected town-Barakott from October 28, 2005 to November 17, 2005. The patients were classifted into upper respiratory tract infection, diarrhea, trauma and other diseases. All the data of patients who were injured during the earthquake were analysed. The difference was analyzed by x^2 test. Results: Of the 2 194 patients treated by the Chinese Medical Rescue Team, trauma patients only accounted for 29%, diarrhea patients for 4%, upper respiratory infection patients for 14%, and other types of diseases for 52%.Among the 630 trauma patients, 426 patients were injured during the earthquake. Of the 426 patients, 291 (68%) patients were found having open wounds, 85 (20%) patients mainly complained of soft tissue pain without any wound or fracture, and 76 (18%) patients had fractures with or without open wound. The most frequent site of wound was lower extremity. Head injury in children was 30%, obviously higher than that in the adults. Wound infection was common among the injured patients, with the rates of 72%, 64% and 78% in male, female adult patients and children, respectively. Conclusions: Prevalence of common diseases, trauma, wound infection and fracture are main problems presented at late stage of the giant earthquake. Great attention should be paid to head and lower extremity injuries.展开更多

The Judd-Ofelt intensity parameters ?_t(t=2, 4, 6) were calculated according to the measured absorption spectra and physical-chemical properties of the obtained Na_5Lu_9F_(32) single crystal. The stimulated emission cross-section of the ~3F_4→~3H_6 transition(~1.8 μm) is 0.35×10^(-20) cm^2 for Tm^(3+) doped Na_5Lu_9F_(32) single crystal. The emission spectra under the excitation of 790 nm laser diode(LD) and fluorescence lifetime at 1.8 μm were measured to reveal the fluorescence properties of Tm^(3+) doped Na_5Lu_9F_(32) single crystal. The research results show that the Tm^(3+) doped Na_5Lu_9F_(32) single crystal has larger stimulated emission cross-section compared with other crystals. All these spectral properties suggest that this kind of Tm^(3+)doped Na_5Lu_9F_(32) crystal with high physical-chemical stability and high-efficiency emission at 1.8 μm may be used as potential laser materials for optical devices.展开更多

文摘Objective:The aim of this study was to observe and compare the antiemetic effectiveness and adverse events of magnetotherapy plus 5-hydroxytryptamine (5-HT3) receptor inhibitor granisetron hydrochloride vs granisetron hydrochloride alone in patients with chemotherapy.Methods:Sixty-four patients were randomized to receive either granisetron hydrochloride alone (control group:granisetron hydrochloride 3 mg intervenous infusion before chemotherapy, from the 1st day of chemotherapy course until the day after chemotherapy course completed) or magnetotherapy plus granisetron hydrochloride (treatment group:the same granisetron hydrochloride regimen plus rotatory magnetotherapy of 1 every day after chemotherapy for 5 begin with chemotherapy).Baseline characteristics were similar in both groups.The patients' emesia was evaluated according to WHO's criteria.The density of 5-HT3 was detected by enzyme-linked immunosorbent assay (ELISA).Results:In the treatment of acute vomiting, there was no significant difference between two groups (P>0.05), but in the treatment of tardive vomiting, the effectiveness in treatment group was better than that in control group (P<0.05).The density of 5-HT3 in treatment group and control group were (225.32±57.29) ng/mL vs (213.00 ±53.29) ng/mL before chemotherapy and (273.88 ±75.42) ng/mL vs (313.17±76.36) ng/mL after chemotherapy (P<0.01); the rate of adverse events was 36.36% and 48.39% respectively in treatment group and control group (P>0.05).Conclusion:Magnetotherapy plus granisetron hydrochloride provide better effectiveness than granisetron hydrochloride alone, and both therapies have synergistic effect.The adverse events didn't raised in treatment group.

文摘Objective To investigate the effect of phenylephrine (an α-adrenergic agonist) on alveolar fluid clearance (AFC) in ventilator-induced lung injury and the possible mechanism involved. Methods A total of 170 male Wistar rats were randomly allocated into 17 groups (n= 10) using ran- dom number tables. Short-term (40 minutes) mechanical ventilation with high tidal volume (HVT) was per- formed to induce lung injury, impair active Na+ transport and lung liquid clearance in the rats. Unventilated rats served as controls. To demonstrate the effect of phenylephrine on AFC, phenylephrine at different con- centrations (1×10^-5, 1 ×10^-6, 1×10^-7, 1×10^-8, and 1×10^-9 mol/L) was injected into the alveolar space of the HVT ventilated rats. To identify the influence of adrenergic antagonists, Na+ channel, and microtubular sys- tem on the effect of phenylephrine, phenylephrine at 1×10^-5mol/L combined with prazosin (an α1-adrener- gic antagonist, 1×10^-4 mol/L), yohimbine (an α2-adrenergic antagonist, 1×10^-4 mol/L), atenolol (a β1- adrenergic antagonist, 1×10^-5 mol/L), ICI- 118551 (an β2-adrenergic antagonist, 1×10^-5 mol/L), amiloride (a Na+ channel blocker, 51×10^-4mol/L), ouabain (a Na+/K+-ATPase blocker, 5~×10^-4mol/L), colchicine (a mi- crotubular disrupting agent, 0.25 mg/100 g body weight), or β-lumicolchicine (an isomer of colchicine, 0.25 mg/100 g body weight) were perfused into the alveolar space of the rats ventilated with HVT for 40 minutes. AFC and total lung water content were measured. Results Basal AFC in control rats was (17.47±2.56)%/hour, which decreased to (9.64± 1.32)%/hour in HVT ventilated rats (P=0.003). The perfusion of phenylephrine at 1 ×10^-8, 1×10^-7, 1×10^-6, and 1×10^-5 mol/L significantly increased the AFC in HVT ventilated rats (all P〈0.05). This effect of phenylephrine on AFC was suppressed by prazosin, atenolol, and ICI-118551 in HVT ventilated rats by 53%, 31%, and 37%, respectively (all P〈0.05). The AFC-stimulating effect of phenylephrine was lowered by 33% and 42% with amiloride and ouabain, respectively (both P〈0.05). Colchicine significantly inhibited the effect of phenylephrine (P=0.031). Conclusion Phenylephrine could increase the AFC in HVT-ventilated rats and accelerate the ab- sorption of pulmonary edema.

文摘Objective To investigate the role of TNF receptor-associated factor 2 (TRAF-2) and TRAF6 in CD40-induced nuclear factor-κB (NF-κB) signaling pathway and whether CD40 signaling requires TRAF2. Methods Human B cell lines were transfected with plasmids expressing wild type TRAF2 or dominant negative TRAF2,TRAF2-shRNA,or TRAF6-shRNA. The activation of NF-κB was detected by Western blot,kinase assay,transfactor enzyme-linked immunosorbent assay (ELISA),and fluorescence resonance energy transfer (FRET). Analysis of the role of TRAF-2 and TRAF-6 in CD40-mediated NF-κB activity was examined following stimulation with recombinant CD154. Results TRAF2 induced activity of IκB-kinases (IKKα,IKKi/ε),phosphorylation of IκBα,as well as nuclear translocation and phosphorylation of p65/RelA. In contrast,TRAF6 strongly induced NF-κB activation and nuclear translocation of p65 as well as p50 and c-Rel. Engagement of CD154-induced nuclear translocation of p65 was inhibited by a TRAF6-shRNA,but conversely was enhanced by a TRAF2-shRNA. Examination of direct interactions between CD40 and TRAFs by FRET documented that both TRAF2 and TRAF6 directly interacted with CD40. However,the two TRAFs competed for CD40 binding. Conclusions These results indicate that TRAF2 can signal in human B cells,but it is not essential for CD40-mediated NF-κB activation. Moreover,TRAF2 can compete with TRAF6 for CD40 binding,and thereby limit the capacity of CD40 engagement to induce NF-κB activation.
文摘Recent studies have revealed that sexually selected traits may signal sperm quality and hence male fertilisation ability. There is also evidence that the expression of male sexual ornamentation and associated sperm characteristics depend on an indi- vidual's ability to cope with oxidative stress. Carotenoids are known for their antioxidant properties and carotenoid-based orna- ments might represent honest signals as these pigments can be traded offbetween the investment in sexual ornamentation, sperm function as well as immune response. In this study, we examined the relationship between sexual ornamentation (breeding colo- ration) and sperm characteristics (e.g., velocity and morphology) in the three-spined stickleback Gasterosteus aeuleatus, an ex- ternally fertilising fish species, in which sperm competition commonly occurs. During the breeding season males are sperm limi- ted and develop a conspicuous carotenoid-based coloration, which is under strong pre-copulatory sexual selection due to female mate choice and male-male competition. The results of the present study show that the expression of stickleback male breeding coloration is significantly positively associated with the linearity of sperm movement, whereas sperm morphology (head length to tail length ratio) is significantly negatively related to the trajectory of sperm movement. Moreover, there is some support for the phenotype-linked fertility hypothesis as the intensity of male red breeding coloration is significantly positively correlated with sperm velocity, which is supposed to be an important determinant of fertilisation success in external fertilisers, indicating the honesty of the sexually selected nuptial red coloration.
文摘Objective: To provide the experience on medical rescue and care for any giant earthquake by analysing the patients treated by the Chinese Medical Rescue Team in the Barakott earthquake of Pakistan. Methods: On October 8, 2005, a giant earthquake (7.8 Richter scale) earthquake occurred in the northwest Pakistan. We belonged to the Chinese Medical Rescue Team and worked in the earthquake-affected town-Barakott from October 28, 2005 to November 17, 2005. The patients were classifted into upper respiratory tract infection, diarrhea, trauma and other diseases. All the data of patients who were injured during the earthquake were analysed. The difference was analyzed by x^2 test. Results: Of the 2 194 patients treated by the Chinese Medical Rescue Team, trauma patients only accounted for 29%, diarrhea patients for 4%, upper respiratory infection patients for 14%, and other types of diseases for 52%.Among the 630 trauma patients, 426 patients were injured during the earthquake. Of the 426 patients, 291 (68%) patients were found having open wounds, 85 (20%) patients mainly complained of soft tissue pain without any wound or fracture, and 76 (18%) patients had fractures with or without open wound. The most frequent site of wound was lower extremity. Head injury in children was 30%, obviously higher than that in the adults. Wound infection was common among the injured patients, with the rates of 72%, 64% and 78% in male, female adult patients and children, respectively. Conclusions: Prevalence of common diseases, trauma, wound infection and fracture are main problems presented at late stage of the giant earthquake. Great attention should be paid to head and lower extremity injuries.

文摘Tm^(3+) doped Na_5Lu_9F_(32) single crystal with high optical quality was grown by an improved Bridgman method. The Judd-Ofelt intensity parameters ?_t(t=2, 4, 6) were calculated according to the measured absorption spectra and physical-chemical properties of the obtained Na_5Lu_9F_(32) single crystal. The stimulated emission cross-section of the ~3F_4→~3H_6 transition(~1.8 μm) is 0.35×10^(-20) cm^2 for Tm^(3+) doped Na_5Lu_9F_(32) single crystal. The emission spectra under the excitation of 790 nm laser diode(LD) and fluorescence lifetime at 1.8 μm were measured to reveal the fluorescence properties of Tm^(3+) doped Na_5Lu_9F_(32) single crystal. The research results show that the Tm^(3+) doped Na_5Lu_9F_(32) single crystal has larger stimulated emission cross-section compared with other crystals. All these spectral properties suggest that this kind of Tm^(3+)doped Na_5Lu_9F_(32) crystal with high physical-chemical stability and high-efficiency emission at 1.8 μm may be used as potential laser materials for optical devices.
