[ Method] A strain of S. hygroscopicus was screened from the soil of Hainan Island. After natural screening and consecutive ultraviolet induced mutation twice, S6-7 strain was obtained as the original strain then treated by UV irradiation and streptomycin resistance screening, and finally rescreened through shake-flask fermentation. [Result] 7 better strains were selected by primary screening from 62 single colonies which were picked out randomly. After 3 generations of consecutive cultivation on slant media and rescreening, 5 strains presented obvious forward mutation. The forward mutation rate reached 8.06%, and the largest production increasing rate came up to 25.11%. [Conclusion] By combining streptomycin resistance screening and conventional ultraviolet induced mutation, both the antibiotic-producing capacity and forward mutation screening efficiency of the original strain were greatly enhanced. 展开更多

Effects of Nitrate on the Growth of Lateral Root and Nitrogen Absorption in Rice 被引量:14

The development of lateral roots (LR) is controlled by both genetic factors and nutrient status in environment. To investigate the effects of nitrate (NO3-) on rice lateral root growth and nitrogen (N) uptake efficiency under upland condition, three treatments, including root-split culture and whole plant culture in N sufficient and deficient conditions, were used in a vermiculite culture experiment. Root-split treatment showed that the growth of lateral roots was stimulated by localized nitrate supply. However, in whole plant culture, elongation of lateral roots was induced by NO3- deficiency. The effects of NO3- on rice lateral root growth were genotype-dependent. Similar N concentration, soluble sugar concentration and N content in shoot were observed in both root-split treatment and whole plant culture under NO3- sufficient condition, suggesting that the nitrogen requirement for rice normal growth could be satisfied with only half of roots supplied with NO3-. In the root-split treatment, N uptake was positively correlated with the average of lateral root length (ALRL) in NO3--supplied side, suggesting that the ALRL is important for rice root N uptake in the environment where the nitrogen nutrient is limiting factor. No significant correlation was observed between N uptake and ALRL in whole plant culture under N sufficient condition, which implies that the length of lateral roots may not be the main factor to determine tire rice root N uptake in nutrient-rich zone. Morphological and metabolic evidence in this study provided some prospects for genetic improvement of root system characters to improve the efficiency of nutrient absorption in rice. 展开更多

The sources and status quo of heavy met- al pollution in agricultural water resources at home and aboard, and the principles of microbial remediation of heavy metals pollution in agricultural water resources were reviewed in this paper, as well as the several common microbial remediation technology of heavy metals in agricultural water resources. Stability of Defibrase in various pH buffer solutions was investigated. Enzyme-linked immuno-sorbent assay (ELISA) and coagulating time method were used to assess antigenic stability and coagulating stability, respectively. The change of antigenic activities and coagulating activities of Defibrase in the same buffer solutions (pH 6, 7 and 8, with the exception of pH 3.6) showed similar tendency to decline with the time. Concentrated Defi-brase was relatively stable at neutral pH 6~7, more than 95% of its initial activities (100BUmL-1) was kept after a 10-day storage at 40 oC, whereas in pH 3.6 and pH 9 buffer solutions, diluted Defibrase was very labile. Addition of Triton X-100 or bovine serum albumin could effectively prevent loss of Defibrase by minimizing adsorption of De-fibrase to plastic surface (P<0.005). Concentration of Defibrase could also affect its stability in aqueous solutions. 展开更多

A comparative study of soil respiration was conducted between in a semi-arid steppe community and in a wet meadow community in the Xilin River Basin of Nei Mongol. The seasonal pattern, the climatic controls, and the correlations of soil respiration with plant biomass components, were examined for each community. The main results are reported as follows: (1) The seasonal changes in soil respiration in the two communities had similar dynamic patterns (both being of two peaks), ranging from 312.8 to 1738.9 mg C(.)m(-2).s(-1) and from 354.6 to 2235.6 Mg C.m(-2).s(-1) in the growing season for the steppe plot and the meadow plot respectively. The soil respiration rate of the meadow plot was distinctly higher than that of the steppe plot, with the daily averages being 1349.6 mg C-m(-2).s(-1) and 785.9 mg C-m(-2).s(-1) respectively. (2) The correlation between soil respiration rate and soil moisture was much more significant than with temperature for the steppe community, and being on the contrary for the meadow community, reflecting the different effects of the two climatic factors in different habitats. Based on these regressive relations, the total CO2 efflux rate in the growing season in 2001 was estimated as 142.4 g C/m(2) in the steppe plot, and 236.1 g C/m(2) in the meadow plot. (3) There was no evident relation between the total canopy biomass and CO2 evolution rate, but a significant power function relation between the live canopy biomass and CO2 evolution rate in the meadow plot was detected. In the steppe plot, there existed only a weak relation between soil respiration and either live or total canopy biomass. 展开更多

One influenza H3N2 virus, A/swine/Shandong/3/2005 (Sw/SD/3/2005), was isolated from pigs with respiratory disease on a farm in eastern China. Genetic analysis revealed that Sw/SD/3/2005 was a triple-reassortant virus with a PB2 gene from human-like H1N1, NS from classical swine H1N1, and the remaining genes from human-like H3N2 virus. These findings further support the concept that swine can serve as reservoir or mixing vessels of influenza virus strains and maintain genetic and antigenic stability of viruses. Furthermore, we have successfully established a reverse genetics system based on eight plasmids and rescued Sw/SD/3/2005 through cell transfection. HI tests and RT-PCR confirmed that the rescued virus maintained the biological properties of the wild type Sw/SD/3/2005. The successful establishment of the reverse genetics system of Sw/SD/3/2005 will enable us to conduct extensive studies of the molecular evolution of H3N2 influenza viruses in swine. 展开更多

The adsorption and desorption of algae Cladophora fascicularis and their relation with initial Cd2+ concentration, initial pH, and co-existing ions were studied. Adsorption equilibrium and biosorption kinetics were established from batch experiments. The adsorption equilibrium was adequately described by the Langmuir isotherm, and biosorption kinetics was in pseudo-second order model. The experiment on co-existing ions showed that the biosorption capacity of biomass decreased with an increasing concentration of competing ions. Desorption experiments indicated that EDTA was efficient desorbent for recovery from Cd2+. With high capacities of metal biosorption and desorption, the biomass of Cladophora fascicularis is promising as a cost-effective biosorbent for the removal of Cd2+ from wastewater. 展开更多

A previously developed model was modified to derive three phytoplankton size classes (micro-, nano-, and pico-phytoplankton) from the overall chlorophyll-a concentration, assuming that each class has a specific absorption coefficient. The modified model performed well using in-situ data from the northern South China Sea, and the results were reliable and accurate. The relative errors of the size-fractioned chlorophyll-a concentration for each size class were: micro-:21%, nano-:41%, pico-:26%, and nano+pico:23%. The model was then applied on ocean color remote sensing data to examine the distribution and variation of phytoplankton size classes in northern South China Sea on a large scale. 展开更多

Heavy metal pollution from industrial wastewater is a worldwide environmental issue. Biosorption of heavy metals by using biosorbents derived from various types of biomass has been shown to be effective for the uptake of heavy metal ions. In this study, biosorbents derived from the biomass of a group of marine macroalgae were used for the removal and recovery of heavy metal ions from aqueous solutions. Results indicated that the biosorbents have high uptake capacities and affinities for a number of heavy metal ions. The uptake capacities of the biosorbents were in the range of 1.0 to 1.5mmol·g-1 for divalent heavy metal ions. The kinetics of the uptake process was fast and the process can be used in both batch and fixed-bed operations. It appears that the biosorption process by using biosorbents from marine macroalgae can be an efficient and cost effective technology for the treatment of heavy metal containing wastewater. 展开更多

An important filamentous industrial fungus,Rhizopus cohnii(R.cohnii),was used as an efficient biosorbent for removing cadmium from wastewater.The sorption conditions,such as pH,the dose of biomass and the initial concentration of cadmium were examined.Two kinds of adsorption models were applied to simulate the biosorption data.The uptake of cadmium was higher in weak acid condition than in strong acid condition.Nearly no sorption of cadmium occurred when the pH value was lower than 2.0. Biosorption isothermal data could be well simulated by both Langmuir and Freundlich models.Langmuir simulation of the biosorption showed that the maximum uptake of cadmium was 40.5 mg/g(0.36 mmol/g)in the optimal conditions,which was higher than many other adsorbents,including biosorbents and activated carbon.In addition,the reusability results showed that after five times of sorption and desorption process,the sorption capacity of R.cohnii could still maintain nearly 80%,confirming its practical application in cadmium treatment.Fourier transform infrared spectrum revealed that carboxyl,amino and hydroxyl groups on biosorbent R.cohnii surface were responsible for the biosorption of cadmium. 展开更多

The objective of the present study is to characterize the production of hydrogen with a sorptionenhanced steam-methane reaction process using Ca(OH)2 as the CO2 adsorbent. Theoretical equilibrium compositions at different operation conditions were calculated using an iterative method. It was found that with Ca(OH)2 as the CO2 sorbent, the concentration of CO2 adsorption was reduced in the product stream, that gave rise to higher methane conversion and higher H2 concentration. An experimental setup was built to test the theoretical calculation. The effects of sorbents and the particle size of Ca(OH)2 on the concentration of CO2 and H2 were investigated in detail. Results showed that the reactor packed with catalyst and Ca(OH)2 particles produced H2 concentration of 94%. It was nearly 96% of the theoretical equilibrium limit, much higher than H2 equilibrium concentration of 67.5% without CO2 sorption under the same conditions of 500℃, 0.2 MPa pressure and a steam-to-methane ratio 6. In addition, the residual mole fraction of CO2 was less than 0.001. 展开更多

Foaming reduces the working volume and limits the biosynthesis of macrolide immunosuppressant ascomycin(FK520) in the batch fermentation process of Streptomyces hygroscopicus FS-35 in a 7.5 L bioreactor. To find the relation between FK520 production and foaming, effects of 10 fermentation parameters including organic acids and membrane permeability were investigated. The results suggest that acetate accumulation caused by short period oxygen de ficiency and fast consumption of glucose is the reason for increased foaming and declined FK520 production. Therefore, a fed-batch fermentation strategy was developed to reduce the accumulation of acetate. After optimization, the maximum acetate concentration dropped from 320 mg·L-1to 157 mg·L-1, decreased by 50.8%, and the maximum foam height reduced from 5.32 cm to 3.74 cm, decreased by 29.7%, while the maximum FK520 production increased from 375 mg·L-1to 421 mg·L-1, improved by 12%. 展开更多

The Microcystis aeruginosa(MA) was immobilized on sodium alginate and used as biosorbent for removal of Cd(Ⅱ) ions from aqueous solution.The biosorption process is pH dependent,and the optimum biosorption was observed at pH 6.0 with the biosorption capacity of 98.38 mg/g.Among Langmuir,Freundlich and Temkin isotherm models,the Freundlich and the Temkin isotherm fit well with the experimental data.Cd(Ⅱ) ions biosorption follows the pseudo-second-order kinetic model.The rate controlling mechanism study reveals that film diffusion is the rate-limiting step and intraparticle diffusion is also involved in biosorption.Thermodynamic parameters,such as Gibbs free energy(ΔG°),the enthalpy(ΔH°) and entropy(ΔS°) were calculated,and revealed that the biosorption process is spontaneous,exothermic and random.Furthermore,the immobilized MA can be regenerated using 0.1 mol/L HCl solutions. 展开更多

The use of water hyacinth biomass as adsorbent for Cr3+ and Mn2+ ions from aqueous solution by means of batch-adsorption technique was investigated to determine the potential ability of the biomaterial for metal ion removal. The equilibrium isotherm study showed that the maximum monolayer coverage on the biomass surface was 0.933 mg·g-1 and 0.874 mg·g-1 for Mn2+ and Cr3+ ions respectively. The highest percentage of Cr3+ and Mn2+ ions adsorbed by the biomass was 86.4% and 82.6% at the optimum pH of 4.0 and 6.0 respectively. The results also showed that the highest percentage removal 82.5% and 78.3% was obtained at 30 and 20 minutes respectively for Cr3+ and Mn2+ ions. The sorption process was examined by means of the Langmuir model. The adsorption equilibrium data were found to follow the Langmuir isotherm model with high coefficients of determination (R2 = 0.990 and 0.999) for Cr+ and Mn2+ ions respectively. The adsorption capacity of water hyacinth showed that water hyacinth will be useful in recovering chromium (III) and manganese (II) ions from solution and their subsequent removal from industrial effluents. 展开更多

AIM: To investigate non-invasively the incidence of absorption of carbohydrates in diabetic patients during an oral glucose tolerance test (OGTT) and to determine whether malabsorption may be associated with insulin secretion and insulin resistance. METHODS: A standard 75-g OGTT was performed in 82 diabetic patients. The patients received 75 g of anhydrous glucose in 225 mL of water after an overnight fasting and breath samples were collected at baseline and up to 120 rain after ingestion. Breath hydrogen and methane concentrations were measured. Blood glucose and serum insulin concentrations were measured before ingestion and at 30, 60, 90, 120 rain post-ingestion. RESULTS: When carbohydrate malabsorption was defined as subjects with an increase of at least 10 ppm (parts per million) in hydrogen or methane excretion within a 2-h period, 28 (34%) had carbohydrate malabsorption. According to the result of increased breath test, 21 (75%) patients were classified as small bowel bacterial overgrowth and 7 (25%) as glucose malabsorption. Patients with carbohydrate malabsorption were older and had poor glycemic control as compared with those without carbohydrate malabsorption. The HOMA value, the sum of serum insulin during the test and the AinsulinlAglucose ratio were greater in patients with carbohydrate malabsorption. CONCLUSION: Insulin resistance may be overestimated by using these markers if the patient has carbohydrate malabsorption, or that carbohydrate malabsorption may be present prior to the development of insulin resistance. Discharge of heavy metals from metal processing industries is known to have adverse effects on the environment.Bio-sorption of heavy metals by metabolically inactive biomass of microbial organisms is an innovative and alternative technology for removal of these pollutants from aqueous solution.The search of marine actinobacteria with potential heavy metal biosorption ability resulted in the identification of a novel alkalophilic Streptomyces VITSVK5 species.The biosorption property of Streptomyces VITSVK5 spp.was investigated by absorbing heavy metals Cadmium (Cd) and Lead (Pb).Physiochemical characteristics and trace metal concentration analysis of the backwater showed the concentrations of different metals were lead 13±2.1 μg L-1,cadmium 3.1±0.3μg L-1,zinc 8.4±2.6μg L-1 and copper 0.3±0.1μg L-1,whereas mercury was well below the detection limit.The effect of pH and biomass dosage on removal efficiency of heavy metal ions was also investigated.The optimum pH for maximal biosorption was 4.0 for Cd (II) and 5.0 for Pb (II) with 41% and 84% biosorption respectively.The biosorbent dosage was optimized as 3 g L-1 for both the trace metals.Fourier transform infrared absorption spectrum results indicated the chemical interactions of hydrogen atoms in carboxyl (–COOH),hydroxyl (–CHOH) and amine (–NH2) groups of biomass with the metal ions.This could be mainly involved in the biosorption of Cd (II) and Pb (II) onto Streptomyces VITSVK5 spp.The results of our study revealed Streptomyces metabolites could be used to develop a biosorbent for adsorbing metal ions from aqueous environments. 展开更多

Effect of Saline Water on Cadmium Uptake and Remediability of Soil with Brassica Napus

This method in comparison with the other techniques is very simple and low cost. Among the pollutants, heavy metals and specially cadmium (Cd) is very important because of its high movement in soil and high toxicity. Bioavailability of metals in the soil is affected by various factors including saline solution. In fact there are symptoms for increasing Cd uptake due to increasing C1"1 concentration in soil solution. The objective of this study was to investigate the effect of saline irrigation water on cadmium uptake and remediability of soil with Brassica napus (rapeseed) in greenhouse experiment. Saline waters of 0, 1.5 and 2.5 dS/m was applied on soil with different concentrations of Cd (5, 10, 20, 30 and 50 ppm) during the cultivation period of Okapi (fall rapeseed variety). The results of this study showed that there are no significant differences in shoot dry matter exposed to Cd, but root dry matter decreased with increasing Cd concentration (p〈5%). Salinity had a significant effect (p〈l%) on Cd concentration in roots and increased from 26.83 (in control) to 36.46 mg.kg-1 dry wt (in 2.5 dS.m"l treatment). The significant effect (p〈5%) was found in shoots as well and the Cd uptake was increased from 2.14 to 3.25 mg.m^-1dry wt in control and 2.5 dS'm^-1 treatments respectively. Likewise, salinity increased bioaccumulation coefficient, whereas had no significant effect on transport factor. The obtained results showed that Cd metal tend to accumulate more in the roots ofBrassica napus than in the shoots. 展开更多

The biosorption mechanism of Cr (Ⅳ) ions on Synechococcus sp. biosorbent was studied by analyzing the biosorption kinetics as well as speciation change and bond formation during the biosorption process. The kinetics study shows that the adsorption process of Cr (Ⅳ) consists of a very fast stage in the first several minutes, in which more than half of the saturation adsorption is attained, and a slower stage that approximately follows the first order kinetic model, basically Freundlich isotherm models were observed. Comparative studies of FT-LR spectra of K2Cr2O7, free cells of Synechococcus sp., and Cr-bound cells of Synechococcus sp show that the speciation of chromium that binds to the cells ofSynechococcus sp. is Cr (Ⅲ), instead of Cr (Ⅳ), and the carboxylic, alcoholic, amido and amino groups may be involved in the binding of Cr (Ⅲ). Integrative analyses of the surface electric potential, the effect of pH value on adsorption behavior of Cr (Ⅵ), and the results of FT-IR show that the biosorption of Cr (Ⅵ) follows two subsequent steps, biosorption of Cr2O7 ^2- by electrostatical force at the protonated active sites and reduction of Cr2O7^2- to Cr^3+ by the reductive groups on the surface of the biosorbents. 展开更多

The experiments show that this process is suitable for the treatment of sorbic acid manufacturing wastewater. About 98% sorbic acid and 50% CODcr were removed under optimal operating conditions, and 70% of sorbic acid in sorbic acid manufacturing wastewater was reclaimed.
