目的:探究原发性痛经患者外周血细胞因子和PGF2α的变化及临床意义。方法:选取2011年11月-2013年11月来本科就诊的50例原发性痛经患者作为观察组,选取同期来本院体检的50例健康月经期女性作为对照组。利用酶联免疫吸实验(ELISA)法对两... 目的:探究原发性痛经患者外周血细胞因子和PGF2α的变化及临床意义。方法:选取2011年11月-2013年11月来本科就诊的50例原发性痛经患者作为观察组,选取同期来本院体检的50例健康月经期女性作为对照组。利用酶联免疫吸实验(ELISA)法对两组女性的经期外周血细胞因子和PGF2α的变化情况进行测定,比较两组经前、经中IL-10、IFN-r、PGF2α等指标的变化情况。结果:观察组经前1周内、经中IL-4、IL-5水平无明显变化(P>0.05),而经中IL-10、IFN-r、PGF2α水平高于经前(P<0.05)。对照组经前1周内、经中IL-4、IL-5、IL-10、IFN-r水平均无明显变化(P>0.05),但经中PGF2α水平高于经前(P<0.05)。观察组经中的IL-5、IL-10、IFN-r、PGF2α水平均明显高于对照组经中水平(P<0.05)。结论:原发性痛经发病机制与PGF2α分泌量、外周血细胞因子有关,原发性痛经患者体内的外周血细胞因子和PGF2α水平会出现异常变化,且PGF2α分泌与细胞免疫因子具有一定关系。 展开更多

Objective: To study the effects of andrographolide on immune functions and the immune mechanism in clinical therapy.Methods: The amounts of IFN-α,IFN-γ, TNF-α, IL-8 in the peripheral blood mononuclear cells (PBMC) culture supernatants dealt with by different concentrations of LianBiZhi (LBZ) injection, the effective component of which is andrographolide, were detected by biological activity test or ELISA in vitro. The effects of LBZ injection on macrophage phagocytotic function and natural killer cells cytotoxicity were examined by means of macrophage to phagocytize cock erythrocyte and measurement of lactate dehydrogenase (LDH) activity released from the damaged cells, respectively.Results: The LBZ injection could not only enhance the phagocytosis activity of peritoneal macrophage from guinea pig to phagocytosis cock erythrocytes, but also augment the cytotoxicity mediated by natural killer cells from PBMCs.Conclusion: Andrographolide is an immunostimulant agent which can modulate both antigen specific and nonspecific immune function by means of its natural killer cells, macrophage and cytokines. 展开更多

Objective To observe the impacts of acupuncture on cell-cycl ODK4) and neuronal death in hippocampal neurons in rats with focal cerebra e-related factors (cyclin D1, schemic reperfusion injury Methods Middle cerebral artery occlusion (MCAO) was used to establish the model of cerebral ischemic reperfusion injury. Western blot (WB) and flow cytometry (FCM) were applied to the tests of cell-cycle-related factors and apoptosis respectively. Results In 48 h of reperfusion, the expressions of cell-cycle-related factors (cyclin D1, CDK4) in hippocampal neurons and apoptosis were increased. In acupuncture group, the expressions of cyclin DI and CDK4 and apoptosis were reduced remarkably (P 〈 0.01 ). Conclusion Acupuncture plays the protective role in cerebral ischemic reperfusion injury, which is contributed probably to the modulation of cell-cycle-related factors to inhibit apoptosis. 展开更多

Objective: The aim of this study was to observe the therapeutic effect of cytokine induced killer (CIK) cells in combination with dendritic cells (DCs) on advanced solid carcinoma patients. Methods: Isolated peripheral blood mononuclear cells (PBMCs) from 110 advanced solid tumor patients. Added granulocyte-macrophage colony-stimulating factor (GM-CSF), tumor necrosis factor-a (TNF-a) and interleukin-4 (IL-4) to adherent cells to induce DCs, and sensitized DCs with antigens of autologous tumor cells or extrinsic tumor cell lines. Cultured suspending cells with interferon-y (IFN-y), interleukin-2 (IL-2) and CD3 monoclonal antibody (CD3 McAb) to prepare CIK cells, then co-cultured with DCs. After analyzing the phenotype and checking tumor markers and immune function, the autologous CIK cells and DCs were transfused into the cancer patients. Results: Forty-two patients with measurable nidus, 2 achieved complete remission (CR), 9 partial remission (PR) and 15 stable disease (SD), while 37 patients with immeasurable nidus, 25 had efficient response. The tumor markers and immune function both improved significantly compared with those before treatment. Conclusion: DCs and CIK cells combinational treatment is safe and effective on advanced solid carcinoma and provide a new and efficacious immunity therapeutic methods for the cancer patients. 展开更多

Objective:To investigate the actions of cytokine profile in the immune cells by a seven amino acid peptide mimic from HVR1 of HCV (GQTYTSG,named 7P).Methods:The peripheral blood mononuclear cells (PBMCs) from 17 healthy blood donors were stimulated with 7P,and the cytokine levels in CD4+CD8-,CD4-CD8+ T cells,NK,NKT cells were analyzed by the intracellular cytokine staining.Results:The frequency of cells which secreted interferon-γ (IFN-γ) was found to be significantly increased in all cells,interleukin-10(IL-10) was significantly increased in CD4+CD8-,CD4-CD8+ T cells but decreased in NK,NKT cells,and tumor necrosis factor-α (TNF-α) was decreased in CD4+CD8-,CD4-CD8+ but increased in NK cells.Conclusion:7P could induce a cytokine profile in different immune cells in vitro and there was some difference between the CD4+CD8-,CD4-CD8+ T cells which represented the adaptive immunity cells and NK,NKT cells which represented the innate immunity cells.This kind of variation of cytokine profile might contribute to anti-virus and anti-inflammatory immune reaction. 展开更多

Objective: To assess the effects of penehyclidine hydrochloride on patients with acute lung injury (ALI), to observe the expression of Toll-like receptor 4 (TLR4) on the peripheral monocytes of ALI patients and changes of inflammatory & anti-inflammatory cytokines and to investigate the mechanism of TLR4 in ALI.Methods: Forty-five patients with ALI were randomly divided into penehyclidine hydrochloride treatment group (P group, n=21) and conventional treatment group (control group, C group, n=24). Patients in both groups received conventional treatment, including active treatment of the primary disease, respiratory support, nutritional support and fluid management therapy, while those in P group were given penehyclidine hydrochloride (1 mg, im, q. 12 h) in addition.The TLR4 expression of 20 healthy volunteers were detected.The clinical effect, average length of stay in ICU and hospital,values of PaO2 and PaO2/FiO2, expression of TLR4 on the surface of peripheral blood mononuclear cells and some serum cytokines were evaluated for 48 h.Results: The general conditions of the two groups were improved gradually and PaO2 increased progressively.Compared with 0 h, PaO2 and PaO2/FiO2 at 6, 12, 24 and 48 h after treatment were significantly increased (P<0.05). The improvement in P group was obviously greater than that in C group (P<0.05). The average length of hospitalization showed no difference between the two groups, but penehyclidine hydrochloride significantly decreased the average length of stay in ICU (t=3.485, P<0.01). The expression of TLR4 in two groups were both obviously higher than that of healthy volunteers (P<0.01). It decreased significantly at 24 h (t=2.032, P<0.05) and 48 h (t=3.620, P<0.01)and was lower in P group than in C group. The patients who showed a higher level of TLR4 expression in early stage had a worse prognosis and most of them developed acute respiratory distress syndrome (ARDS). The incidence of ARDS was 23.8% in P group and 29.17% in C group at 24 h.Until148 h, there were other two patients developing ARDS in control group. Serum IL-l, IL-8 and TNF-α expressions reduced after 24 h in both groups. The reduction in P group was more obvious than that in C group (P<0.05). IL-13 increased gradually from 0 h to 24 h, and decreased slightly at 48 h, which showed no difference between two groups (t=1.028, P>0.05).Conclusions: Penehyclidine hydrochloride improves the arterial oxygen pressure, down-regulates the expression of TLR4 and restrains the inflammatory cytokines in the downstream of TLR4 signaling pathway. It prevents the development of ALI and can be considered as an important drug in ALI treatment. 展开更多
