In pBinMoBc, cry 1Ac3 gene, which encodes the Bt toxin, is under the control of chimeric OM promoter. In pBinoBc, it is under control of CaMV 35S promoter. After co_cultivation with Agrobacterium tumefimpfaciens LBA4404 (containing pBinMoBc or pBinoBc), kanamycin_resistant selection, somatic embryos were induced and regenerated plants were obtained. Then the regenerated plantlets were grafted to untransformed stocks in greenhouse to produce descendants. The integration of cry 1Ac3 gene and its expression in T 2 generation of transgenic cotton plants were confirmed by Southern hybridization and Western blotting. The analyses of insect bioassay indicated that the transgenic plants of both constructions have significant resistance to the larvae of cotton bollworm ( Heliothis armigera ) and that cry 1Ac3 gene driven by chimeric OM promoter could endue T 2 generation cotton with high pest_resistant ability, implicating that it has a profound application in genetic engineering to breed new pest_resistant cotton varieties.展开更多

Fluridone was sprayed on potato leaves at rates of l, 5, 10, 20 and 30 μM/ha. Each rate was tested for single and sequential application at 20 d and 35 d after potato emergence. Results indicated that fluridone at all the tested rates (single or sequential applications) significantly reduced Phelipanche infestation and shoot number compared to the control. Fluridone at 10 μM/ha reduced Phelipanche infestation by 99% when applied twice (20 d and 35 d after potato emergence). All tested rates except for fluridone at 30μM/ha (single or two applications) were selective in potato and had no negative effect on potato growth and yield as compared to the control.展开更多

However, fewer studies concerned the effects of transgenic corn cultivation on the soil Collembola in black soil region in China. In Jilin black soil region, a six treatments study including three corn varieties (transgenic corn, control and local variety) and two sampling locations (in rizhosphere and in between plants) were deployed to study the effect of transgenic corn cultivation and sampling location on densities, species richness and community structure of Collembola. We found that root biomass was marginally decreased in transgenic corn and control variety when compared to local variety. Collembola significantly affected by sampling location treatments. Densities and species richness of collembolans significantly decreased in "between plants" than in "rizhosphere". Densities of Collembola were impacted by corn varieties. In the whole growing season, densities of Collembola significantly decreased in control than in local variety. Collembolans in transgenic variety only slightly decreased in "jointing stage" than local variety. Collembolans adversely affected in "between plants" when compared to "in rhizosphere" in "seeding stage" but not other stages. These variations of collembolans were in lines with the trend of corn root biomass. It is indicated that Collembola variations could be caused by food resources (root biomass) but not corn varieties (transgenic, control and local).展开更多

The results showed that production of local early-grown Satsuma or- ange orchards could be significantly improved through the application of cultivation techniques such as rational trimming, strengthening soil, fertilizer and water manage- ment, conducting flower and fruit management pertinently, effective and comprehen- sive control of disease and insects. The yield difference between "on" and "off" years was 10%-15%, and the orchards with serious alternate bearing phenomenon were changed into the one with high and stable yield. Meanwhile, the commercial rate of early-grown Satsuma orange fruit enhanced, leading to good economic benefits.展开更多

Contr...A field experiment was conducted using Ceranock bait station, "attract and kill" system to combat Mediterranean fruit fly, Ceratitis capitata, in apricot orchards, Central of lraq during the 2013 field season. Control program was implemented in three orchards of mixed apricot varieties. Ceranock traps were hung in each tree of two separate orchards. Delta and McPhail traps were used to monitor Medfly population density in each treated and control orchards. Results indicated that Medfly activity in apricot orchards started during the last week of April coincided with the beginning of maturity for fruits of early varieties. The total of trapped insects in monitoring traps after one week of Ceranock application were 0, 1 and 31 adults for the 1st, 2nd and the control orchards, respectively. Three weeks later and on, the number of trapped adults started to increase dramatically. At the end of apricot season and after 45 days of Ceranock trap application, the number of trapped insects in monitoring traps reached 110, 111 and 2,349 adults/week, respectively, for the 1st, 2nd and 3rd orchard. The use of Ceranock traps reduced Medfly population density by 86%-97%. The percentage of fruit injuries in early maturing varieties were 0.75%, 1% and 34% for the 1st, 2nd and control orchards, respectively. While, late varieties were 2% and 3% for 1st and 2nd orchards, and 69% for the control. The percentage of reduction in fruit damages reached to 95% and 97%, respectively in the 1st and 2nd orchard. The results of this study demonstrate clearly the efficacy of Ceranock bait station, "attract and kill" system as a control measure for Medfly in apricot orchards.展开更多

Russet Burbank, Shepody, Innovator, FL-1867, and FL-1833 were loaded into the Univers...Ethylene was tested as a sprout inhibitor on potatoes intended for processing over the 2006-2007 and the 2007-2008 storage seasons. Russet Burbank, Shepody, Innovator, FL-1867, and FL-1833 were loaded into the University of Maine potato research facility in October of 2006. Ethylene gas was metered into the storage until a concentration of 0.1 ppm was reached and then held for one week. At weekly intervals, the ethylene concentration was increased to 0.3 ppm, 0.6 ppm, 12 ppm, 5.0 ppm and 102 ppm. Subsequently, the concentration was maintained at 10.0 ppm for the duration of the storage season. Control potatoes received chlorpropham (CIPC) for sprout control. The trial was repeated in the 2007-2008 storage season. The Agtron readings were performed on the processed potatoes at the end of the storage season. There was no difference in the US color grade between the CIPC-treated and ethylene-treated tubers within any tested variety in either storage season. Sprout inhibition of the ethylene-treated potatoes was acceptable for normal storage length of the potato varieties tested. Ethylene may have promise as an alternate to CIPC for sprout control in potato storages.展开更多

The habitat manipulation strategy tested in the study was to plant seedlings ofPaspalum scrobiculatum on one meter strip of land at the border of each rice plot early in the cropping season to conserve P. diplosisae in its alternative host, Orseolia bonzii that infest P. scrobiculatum. The Paspalum seedlings were allowed to be fully established, infested naturally by 0. bonzii and parasitized by P. diplosisae before transplanting rice varieties in each plot. The control plots were planted with only rice seedlings leaving the border empty without any Paspalum. Rice plots with Paspalum at the edge had much lower levels of O. oryzivora infestation than the control plots. The percentage number of gall midge parasitized by P. diplosisae showed that rice plots with Paspalum at the edge had more parasitized galls than the control plots without Paspalum. The role of the Paspalum border was substantiated with the negative correlation results recorded with the infestation indicating significant decrease in the percentage tiller infestation as the percentage parasitism increased. That resulted in higher grain yield in the plots surrounded with Paspalum treatment than in the control. The present study therefore demonstrated that management of P. scrobiculatum on a strip of land around the edge of rice crops could significantly increase parasitoid carry-over into the crop.展开更多

Nativo 300 SC (trifloxystrobin 100 g L^-1 + tebuconazole 200 g L^-1) and Prosaro 250 EC (prothioconazole 125 g L^-1 + tebuconazole 125 g L^-1), in controlling stem rust on susceptible wheat cultivar Duma. AmistarXtra 280 SC (azoxystrobin 200 g L^-1 + cyproconazole 80 g L^-1) and Folicur 250 EC (tebuconazole) were used as checks. The treatment at each site and year included non-treated control and two spray applications of the fungicides at growth stages (GS) 55 (heading) and 65 (flowering). Stem rust severities were scored using the modified Cobb scale at 14-day intervals after application. The data were used to calculate mean rust severity (MRS). Stem rust epidemics were severe at KARI-Njoro in 2008 and the treatment effects on stem rust severities, grain yield and 1,000 kernel weights were significant at both KARI-Njoro and Mau-Narok sites. The fungicide treatments, significantly (P _〈 0.05) reduced stem rust severity, increased grain yield and 1,000 kernel weight of the susceptible wheat cultivar Duma compared to the non-treated control.
