The result...Two hydrocarbon degrading bacteria NJ276 and NJ341 were screened from 385 Antarctic marine bacteria and their degrading characteristics were studied. Diesel oil as sole carbon source was used in this study. The results showed that the oil degradation rates of Antarctic psychrophile bacteria NJ276 and NJ341 were 23.47 % and 32.15 %, respectively, after 20 days culturation at 5 ℃, and the rates were 43.95 % and 62.47 % respectively after 20 days culturation at 15 ℃. The oil degradation abilities were enhanced remarkably with the increasing culture temperature. GC - MSs indicated the residual oil contained C15 - C21 7 alkyls after degradation by NJ276, and C16, C17 and C18 3 alkyls after degradation by NJ341. The 16S rDNA gene sequences homology and phylogenetic analysis of the two Antarctic psychrophile bacteria showed that NJ276 belonged to the described genus Pseudoalteromonas and NJ341 belonged to the genus Colwellia.展开更多
The adsorption and leaching of chalcopyrite by two extreme thermophilic archaea(A.brierleyi and S.metallicus)and their mixture were studied.The results revealed that the chalcopyrite leaching rate of S.metallicus was ...The adsorption and leaching of chalcopyrite by two extreme thermophilic archaea(A.brierleyi and S.metallicus)and their mixture were studied.The results revealed that the chalcopyrite leaching rate of S.metallicus was slightly higher than that of A.brierleyi;the mixed system showed the highest rate.Community structure analysis during the leaching process showed that S.metallicus was maintained in a predominant state.However,the proportion of A.brierleyi in the community increased during leaching.Copper concentrations,which increased faster in the mixed system than in the single-organism systems during later stages,was related to the change of A.brierleyi in the community.Langmuir parameter analysis revealed no competitive adsorption between these two thermophilic archaea.Furthermore,qPCR(quantitative polymerase chain reaction)confirmed that adsorption was promoted between A.brierleyi and S.metallicus during mixed leaching.These findings can improve our understanding of the adsorption behaviors of mixed extreme microbial populations on mineral surfaces.展开更多
The effects of introducing M.sedula derivatives having different Cu^2+-resistance on bioleaching capacity of a defined consortium(consisting of A.brierleyi DSM1651 and M.hakonensis HO1-1)were studied in column reactor...The effects of introducing M.sedula derivatives having different Cu^2+-resistance on bioleaching capacity of a defined consortium(consisting of A.brierleyi DSM1651 and M.hakonensis HO1-1)were studied in column reactors at 70℃.Introducing M.sedula copA mutant,a copper sensitive derivative,only had negligible effects on bioleaching.While introducing M.sedula ARS50-2,a Cu^2+resistant strain,substantially consolidated bioleaching process,with 27.77%more copper recovered after 58 d of bioleaching.Addition of M.sedula ARS50-2 likely enhanced the sulfur oxidation capacity of consortium after the 24th day under the Cu^2+stress.The majority of extreme thermoacidophiles were attached on minerals surface as indicated by quantitative PCR(qPCR)data.Successions of microbial community of extremely thermoacidophilic consortia that attached on surface of minerals were different from those in leachate.M.hakonensis HO1-1 was the dominant species attached on minerals surface in each column reactor throughout bioleaching process.The sessile M.sedula ARS50-2 remained as a major species till the 34th day.A.brierleyi DSM1651 was the most abundant planktonic species in leachate of each column reactor.These results highlight that higher Cu^2+-resistance is a beneficial trait for extreme thermoacidophiles to process copper minerals.展开更多
A total of 69 strains of thermophilic bacteria were isolated from water, soil and sediment samples from three Indonesia's hot spring areas (Pantai cermin, Kalianda and Banyu wedang) by using Minimal Synthetic Medi...A total of 69 strains of thermophilic bacteria were isolated from water, soil and sediment samples from three Indonesia's hot spring areas (Pantai cermin, Kalianda and Banyu wedang) by using Minimal Synthetic Medium (MSM). The extreme thermophile Brevibacillus sp. PLI-1 was found to produce extracellular thermophilic alkaline protease with optimal activity at 70℃ and pH 8.0-9.0. The molecular weight of the protease was estimated to be around 56 kD by SDS-PAGE. The maximum activity of the protease was 26.54 U mL-1. The protease activity did not decrease after 30 min and still retained more than 70% of relative activity after 60 min at 70℃ and pH 8.0. The ion Mg2+ was found to promote protease activity at both low and high concentrations, whereas Cu2+ and Zn2+ could almost completely inhibit the activity. Divalent cation chelator EDTA inhibited the enzyme activity by 55.06% ± 0.27%, while the inhibition caused by PMSF, Leupeptin, Pepstain A and Benzamidine were 66.78% ± 3.25%, 52.37% ± 0.25%, 62.47% ± 2.96% and 50.99% ± 0.24%, respectively. Based on these observations, the enzyme activity was conspicuously sensitive to the serine and cysteine protease inhibitors. All these results indicated that the protease isolated from the strain PLI-1 was a thermophilic protease and had a high-temperature stability and a pH stability.展开更多

It is found that adding certain amounts of L-cysteine to the bioleaching system of Ni-Cu sulfide largely enhances the leaching rate. X-ray diffraction (XRD) patterns show the change of bioleached solid residues and the effect of L-cysteine on the surface charges of minerals. Zeta potential and IR spectra of mineral surface show that the interaction between L-cysteine and mineral leads to the formation of metal complex,which is propitious to the bioleaching of Ni-Cu sulfide by Acidianus manzaensis.展开更多

Eigh...Two hundred strains of bacteria from Antarctic sea ice were collected and screened for their ability of producing eieosapentaenoic acid (EPA, 20: 5ω3) by means of Gas Chromatography/Mass Spetmmetry (GC/MS). Eight strains of bacteria containing EPA were investigated, among which the outstanding one was recorded as NJ136. This bacterium was identified as Shewanella by the biochemical characteristics and 16S rRNA sequence. Response surface methodology (RSM) was applied to optimize the medium ingredients. A 24 full factorial central composite design (FFCCD) was employed to determine the naximum EPA production at optimum levels of pH, NaC1, glucose and yeast extract. The predicted optimal combination of media constituents for maximum 14.02 mg/g ( about 1.7-fold increase) EPA production were determined as 30.15%e (m/v) NaC1, 9.98 g/L glucose, 4.42 g/L yeast extract and pH 6.08. The actual experimental results were in agreement with the prediction.展开更多
