[Method] in vitro anti-bacterial effects of 5 kinds of Chinese herb extracts like Galla Chinensis,Syzygium aromaticum,Salvia miltiorrhiza,Punica granatum L. and Terminalia chebula Retz on Ameromonas hydrophila were studied; furthermore,cure rates of the Chinese herb extracts with better anti-bacterial effects were determined to find out the optimal drug dosage. [Result] Under the same experimental conditions,Galla Chinensis,Punica granatum and Terminalia chebula Retz had relatively strong anti-bacterial effects on Ameromonas hydrophila,among them the anti-bacterial effect of Galla Chinensis was significantly higher than those of the others (P0.05). The optimal treatment dose of Galla Chinensis treating bacterial septicemia caused by Aeromonas hydrophila was that they were treated with medicated bath for 40 min by 0.5 mg/ml Galla Chinensis extract,and the cure rate was 100%. [Conclusion] The research provides a scientific drug basis for the control and prevention of outbreak bacterial diseases of fish.展开更多

The growth of intestinal villi of the fishes that had been fed with the feed mixed with compound probiotics for three months was observed; meanwhile, the content of in- testinal Aeromonas hydrophila was detected by real-time quantitative PCR. |Result] The compound probiotics promoted the development of intestinal villi, and inhibited the growth of A. hydrophila, and the effects were also concentration dependent. However, the compound probiotics did not increase the thickness of the intestinal serous layer, muscular layer and submucosal layer. As a potent pleiotropic cytokine, tumor necrosis factor-alpha (TNF-ct) plays an important role in innate immune responses. The cDNA sequence and genomic structure of the TNF-α gene (AjTNF-c0 in the Japanese eel (...As a potent pleiotropic cytokine, tumor necrosis factor-alpha (TNF-ct) plays an important role in innate immune responses. The cDNA sequence and genomic structure of the TNF-α gene (AjTNF-c0 in the Japanese eel (Anguilla japonica) were identified and characterized. The full-length AjTNF-α cDNA was 1 546 bp, including a 5'-untranslated region (UTR) of 13 bp, a 3'-UTR of 879 bp and an open reading frame of 654 bp encoding a protein of 218 amino acids. The full-length genomic sequence of AjTNF-ct was 2 392 bp and included four exons and three introns. The putative AjTNF-α protein contained TNF family signature motifs, including a protease cleavage site, a transmembrane domain and two conserved cysteine residues. Quantitative real-time reverse transcription PCR analysis revealed AjTNF-α expression in a wide range of tissues, with predominant expression in blood and liver. Lower levels of expression were seen in spleen, gills, kidney, intestine, heart, and skin, with very low levels in muscle. The modulation of AjTNF-ct expression after injection of eels with lipopolysaccharide (LPS), the viral mimic, poly I:C, or Aeromonas hydrophila was assessed in blood, liver, and kidney. In blood, TNF-α mRNA levels increased rapidly and then rapidly decreased after stimulation with LPS, poly I:C or A. hydrophila. However, the response to LPS and A. hydrophila peaked at 6 h while for poly I:C the peak was at 12 h. In liver, after injection with A. hydrophila, an up- and down-regulation of AjTNF-ct expression occurred twice, peaking at 6 h and 24 h, respectively. No remarkable increase of AjTNF-α expression appeared in liver until 72 h after LPS or poly I:C treatment. In kidney, ApiNF-α expression increased significantly only at 72 h post-stimulation with LPS orA. hydrophila. Our results suggest that AjTNF-α plays an important role in fish in the defense against viral and bacterial infection.展开更多

To determine the potential effect of chitosan on the shelf-life of filleted tilapia, this study analyzed the bacterial community diversity in fresh and spoiled tilapia fillets stored at (4 ± 1)℃ and examined the antimicrobial activity of chitosan against relevant bacteria isolates. Results showed that Pseudomonas (20%) and Aeromonas (16%) were abundant in fresh tilapia fillets, whereas Pseudomonas (52%), Aeromonas (32%) and Staphylococcus (12%) were dominant in the spoiled samples. Chitosan showed wide-spectrum antibacterial activity against bacteria isolated from tilapia and 5.0 g L-1 chitosan was selected for application in preservation. We further determined the shelf-life of chitosan-treated, filleted tilapia stored at (4 ± 1)℃ based on microbiological, biochemical and sensory analyses. Results showed that the shelf-life of chitosan-treated, filleted tilapia was extended to 12 d, whereas that of untreated, control samples was 6 d. These indicate that chitosan, as a natural preservative, has great application potential in the shelf-life extension of tilapia fillets.展开更多

Bacteria evaluated were Aeromonas hydrophila, Pseudomonas aeruginosa and Edwardseilla tarda. A. hydrophila was resistant to ampicilin, cephalothin, sulphatriad, cefpodoxime and cefoxitin. Highest sensitivity was recorded to Colistin methane sulphonate and Ceftriaxone. High resistance was observed for P. aeruginosa in Kanamysin, Cephalothin, Sulphatriad, Colistin methane sulphonate, Tobramysin, Nitro Furantoin, Cefpodoxime, Cefoxitin and Aztreonam. Highest sensitivity recorded with Moxifloxacin. High resistance was observed in Co-trimoxamole and higher sensitivity performed for E. tarda by Tetracyclin, Cephotaxime, Ciprofloxacin, Norfloxacin, Ofloxacin, Moxifloxacin, Gatifloxacin and Levofloxacin. Out of that highest sensitivity recorded with Moxifloxacin High sensitivity was recorded for Moxifloxacin, Ciprofloxacin, Gatifloxacin, Levofloxacin.
