[Method] Several DNA extraction methods were used to extract DNA from Wusuli raccoon dog hair, including Chelex-100 method, PCR buffer method, organic phenol-chloroform method and centrifugal col- umn type kit method. The extracted DNA was analyzed by using PCR amplification and electrophoresis to compare these four DNA extraction methods. [Result] Accord- ing to the results of spectrophotometer detection and gel electrophoresis, nucleic acid extracted by Chetex-100 method had proteins and other impurities; nucleic acid ex- tracted by PCR buffer method was low in concentration; however, DNA extracted by organic phenol-chloroform method and centrifugal column type kit was high in con- centration with no impurity band. [Conclusion] This study had laid the strong founda- tion of scientific theory to further explore the efficient and simple method for extracting DNA from Wusuli raccoon dog hair follicle.展开更多

Results: We performed the resection for ciliated hepatic foregut cyst under laparoscopy; the patient recovered well after the procedure. Conclusion: Ciliated hepatic foregut cyst is quite rare clinically, belongs to non-parasitic,solitary and unilocular cystic lesion, is always less than 4cm in diameter, mostly seen in the left lobe, and has the tendency of malignant change. We present a case of a middle age woman with acromegaly who was investigated for nonspecific gastrointestinal complaints. Ultrasonography and subsequent endosonography diagnosed a large (4.5 cm), hyperechoic, sessile polyp with numerous pedicles. An open cholecystectomy was performed and revealed a villous adenoma with several foci of carcinoma in situ. Detailed investigations showed no other tumors of the gastrointestinal tract. After five years of follo up, the patient reports no complaints, and the results of laboratory testing and imaging studies are within the normal range.展开更多

He was diagnosed as esophageal basaloid squamous carcinoma (BSC) and gastric stromal tumor, which were associated with focal proliferation of melanocytes/ pigmentophages and hair follicles in esophageal mucosa. Melanocytic hyperplasia (melanocytosis) has previously been recognized as an occasional reactive lesion, which can accompany esophageal inflammation and invasive squamous carcinoma. The present case is unusual because of its hyperplasia of not only melanocytes but also hair follicles. To our knowledge, this is the first report of esophageal blue nevus and hair follicle coexisting with BSC.展开更多

Cristae, highly-tangled with tubular-type struc-tures, appeared on the mitochondria, and were morpho-logically similar to that of vegetative cells. In the cor-tical ciliatures, such as ciliary shafts, kinetosomes, sur-rounding fibrillar cirral baskets, and attached structures of ciliatures, etc., they are different from those in rest- ing cysts which are degenerated or lost. All the cilia- ture microtubules of ciliary shafts are of the 9+2 pattern,but the microtubule-like structure aggregates at triplet - microtubule centers of many kinetosmes, are still under various stages of differentiation. AIM:To study the inhibition of tumor angiogenesis by 5,2,4'-trihydroxy-6,7,5'-trimethoxyflavone(TTF1) isolated from an extract of herbal medicine Sorbaria sorbifolia.METHODS:Angiogenic activity was assayed usi...AIM:To study the inhibition of tumor angiogenesis by 5,2,4'-trihydroxy-6,7,5'-trimethoxyflavone(TTF1) isolated from an extract of herbal medicine Sorbaria sorbifolia.METHODS:Angiogenic activity was assayed using the chick embryo chorioallantoic membrane(CAM) method.Microvessel density(MVD) was determined by staining tissue sections immunohistochemically for CD34 using the Weidner capillary counting method.The mRNA and protein levels of vascular endothelial growth factor(VEGF),vascular endothelialgrowth factor receptor 2(VEGFR2,Flk-1/KDR),basic fibroblast growth factor(bFGF),cyclo-oxygenase(COX)-2 and hypoxia-inducible factor(HIF)-1α were detected by quantitative real-time polymerase chain reaction and Western blotting analysis.RESULTS:The TTF1 inhibition rates for CAM were 30.8%,38.2% and 47.5% with treatment concentrations of 25,50 and 100 μg/embryo × 5 d,respectively.The inhibitory rates for tumor size were 43.8%,49.4% and 59.6% at TTF1 treatment concentrations of 5,10,and 20 μmol/kg,respectively.The average MVD was 14.2,11.2 and 8.5 at treatment concentrations of 5 μmol/kg,10 μmol/kg and 20 μmol/kg TTF1,respectively.The mRNA and protein levels of VEGF,KDR,bFGF,COX-2 and HIF-1α in mice treated with TTF1 were significantly decreased.CONCLUSION:TTF1 can inhibit tumor angiogenesis,and the mechanism may be associated with the down-regulation of VEGF,KDR,bFGF,HIF-1α and COX-2.展开更多

[ Method] Larynx, trachea, liver and other organs were collected from infectious laryngotracheitis (ILT)-suspected laye...[ Objective] To isolate and identify infectious laryngotracheitis virus (ILTV) from chickens. [ Method] Larynx, trachea, liver and other organs were collected from infectious laryngotracheitis (ILT)-suspected layers. And ILTV TK gene was amplified from these specimens by PCR for initial diagnosis. Virus fluid was isolated and inoculated into SPF chicken embryos via allantoic cavity and chorioallantoic membrane (CAM), respectively. Hyaluronic acid in allantoic fluid was detected, and CAM lesions were observed. The definite diagnosis was performed through animal regression test. [Result] A 1.3 kbp fragment was amplified from larynx and its secretion of the ILT-suspected chickens. And its amino acid sequence had 98.5% homology to that of ILTV TKgene published in GenBank. After the chicken embryos were inoculated with the isolated ILFV fluid, pox spots, giant polynuclear syncytial cells having intranuclear inclusion bodies were observed in CAM. After being challenged by the IL TV fluid, the chickens showed typical respiratory symptoms and pathological changes of ILT. The diameter of cysts was <5 cm in 76 cases, and w...Objective. To evaluate the surgery in simple ovarian cysts.Methods. From Jan.1994 to Dec.1999, 221 women with simple ovarian cysts were admitted into our hospital. The diameter of cysts was <5 cm in 76 cases, and was>5 cm in 145 cases ultrasonically. One hundred and eighty-four patients underwent laparosocopy, and thirty-seven underwent laparotomy.Results. Histological findings showed no malignancy in this series. Simple cysts, paraovarian cysts and corpus luteum were found histologically in 90.8% of cases with cysts <5 cm, and 60% of those with cysts >1 cm respectively, their difference was significant (x2=37.13, P<0.001). The simple cysts, paraovarian cysts and corpus luteum cysts were found in 81.5% of postmenopausal women and 84.0% of pre-menopausal women.Conclusion. Patients with cysts >1 cm are indicated for surgical procedures, while a period of fol-lowup is acceptable for patients with cysts <7 cm, and surgery is advisable if the cyst is persistent during followup. Postmenopausal women with cysts should have operations.展开更多

In this research, we explored the effect of DADAG on angiogenesis in chick chorioallantoic membrane (CAM) model and on the proliferation and migration of human umbilical vein endothelial cells (HUVECs). We also studied the possible mechanism of the anti-angiogenesis effect of DADAG. The results showed that DADAG (100, 500 and 1000μnol/L) inhibited angiogenesis in CAM model dose-dependently. Sulforhodamine B (SRB) assay indicated that DADAG (45, 90, 135, 180 and 225 μmol/L) suppressed HUVECs proliferation in a dose-dependent and time-dependent manner. High Content Screening (HCS, Cellomics) assay, in which the influence of cell proliferation on migration could be excluded, indicated that DADAG (45, 135 and 225 ~xmol/L) directly inhibited the motility ofHUVECs. Immunofiuorescence assay suggested that DADAG inhibited angiogenesis possibly by decreasing vascular endothelial growth factor (VEGF) expression in HUVECs. Our findings reveal that DADAG show anti-angiogenic activity in vivo and in vitro, which is related to the downregulation of VEGF expression in endothelial cells.展开更多

The theory of the mechanism of adhesion by van der W...The study of the adhesion of millions of setae on the toes of geckos has been advanced in recent years with the emergence of new technology and measurement methods. The theory of the mechanism of adhesion by van der Waals forces is now accepted and broadly understood. However, this paper presents limitations of this theory and gives a new hypothesis of the biomechanism of gecko adhesion. The findings are obtained through measurements of the magnitude of the adhesion of setae under three different conditions, to show the close relationship between adhesion and status of the setae. They are reinforced by demonstrating two setal structures, follicle cells and hair, the former making the setae capable of producing bioelectrical charges, which play an important role in attachment and detachment processes. It is shown that the abundant muscular tissues at the base of the setae cells, which are controlled by peripheral nerves, are instrumental in producing the foot movement involved in attachment and detachment. Our study will further uncover the adhesion mechanism of geckos, and provide new ideas for designing and fabricating synthetic setae.展开更多

Hirsutella rhossi...Functional response is a key index in determining the population fluctuation in predation. However, the lack of operable research system limits the studies on functional response of fungal predators. Hirsutella rhossiliensis is a dominant parasite of the soybean cyst nematode, Heterodera glycines. In a soil microcosm bioassay, we determined fungal biomass at different days within 21 days after inoculation, and parasitism rate of H. glycines by the fungus was determined. The functional response of H. rhossiliensis to H. glycines was established and found to be Holling's type Ill, which was influenced by mycelial densities. Meanwhile, we conducted anti-fungal analysis of metabolic fractions extracted from H. rhossiliensis to explain the potential mechanism of the intraspecific competition illustrated by functional response. The result of anti-fungal experiments indicated that the fungal predators had more complicated interaction at population level than expected, which might be regulated by self-inhibition metabolite(s). This study was the first functional response study of fungal predators in microcosm. With the in- creasing recognition of emerging fungal threats to animal, plant, and ecosystem health, the methodologies and hypotheses proposed in this study might inspire further research in fungal ecology.展开更多

Simple cysts, paraovarian cysts and corpus luteum were found histologically in 90.8% of cases with cysts <5 cm, and 60% of those with cysts >1 cm respectively, their difference was significant (x2=37.13, P<0.001). The simple cysts, paraovarian cysts and corpus luteum cysts were found in 81.5% of postmenopausal women and 84.0% of pre-menopausal women.Conclusion. Patients with cysts >1 cm are indicated for surgical procedures, while a period of fol-lowup is acceptable for patients with cysts <7 cm, and surgery is advisable if the cyst is persistent during followup. Postmenopausal women with cysts should have operations.
文摘With the increasing incidence of hair loss and especially among younger population in recent years,the demand for diversified products on anti-hair loss solutions is growing and the market of cosmetic products expanding accordingly.Worldwide attention is being paid to the research and development of anti-hair loss actives,while there is no unified standard for the efficacy evaluation and testing methods of anti-hair loss cosmetics.Therefore,it is critical to establish standardized methodology for assessing the effectiveness of anti-hair loss actives and their cosmetic products.In this review,an overview of the human hair follicle structure,the hair follicle cycle,and the biological pathways and signaling mechanisms involved in hair growth are summarized.This provides the fundamental understanding of human hair growth for reviewing anti-hair loss actives and cosmetic hair loss products,and respective methods and techniques applied for evaluating their efficacy.Additionally,changes in the normal hair cycle,and corresponding cosmetic strategies to slow down and reduce hair loss have been provided.This review aims to provide recommendations for evaluating the effectiveness of anti-hair loss actives on the mechanisms involved in the hair follicle growth cycle,and so provide criteria for new technology development and scientific research for cosmetic products.
文摘1,2,5,6-Dianhydro-3,4-diacetyl-galactitol (DADAG), an alkylating sugar alcohol derivative, has been shown effective against tumor growth. In this research, we explored the effect of DADAG on angiogenesis in chick chorioallantoic membrane (CAM) model and on the proliferation and migration of human umbilical vein endothelial cells (HUVECs). We also studied the possible mechanism of the anti-angiogenesis effect of DADAG. The results showed that DADAG (100, 500 and 1000μnol/L) inhibited angiogenesis in CAM model dose-dependently. Sulforhodamine B (SRB) assay indicated that DADAG (45, 90, 135, 180 and 225 μmol/L) suppressed HUVECs proliferation in a dose-dependent and time-dependent manner. High Content Screening (HCS, Cellomics) assay, in which the influence of cell proliferation on migration could be excluded, indicated that DADAG (45, 135 and 225 ~xmol/L) directly inhibited the motility ofHUVECs. Immunofiuorescence assay suggested that DADAG inhibited angiogenesis possibly by decreasing vascular endothelial growth factor (VEGF) expression in HUVECs. Our findings reveal that DADAG show anti-angiogenic activity in vivo and in vitro, which is related to the downregulation of VEGF expression in endothelial cells.

文摘The study of the adhesion of millions of setae on the toes of geckos has been advanced in recent years with the emergence of new technology and measurement methods. The theory of the mechanism of adhesion by van der Waals forces is now accepted and broadly understood. However, this paper presents limitations of this theory and gives a new hypothesis of the biomechanism of gecko adhesion. The findings are obtained through measurements of the magnitude of the adhesion of setae under three different conditions, to show the close relationship between adhesion and status of the setae. They are reinforced by demonstrating two setal structures, follicle cells and hair, the former making the setae capable of producing bioelectrical charges, which play an important role in attachment and detachment processes. It is shown that the abundant muscular tissues at the base of the setae cells, which are controlled by peripheral nerves, are instrumental in producing the foot movement involved in attachment and detachment. Our study will further uncover the adhesion mechanism of geckos, and provide new ideas for designing and fabricating synthetic setae.
文摘We investigated the anti-angiogenic effects of the water extract of HangAmDan (WEHAD),which is a crude extract of nine Korean medicinal substances of animal and plant origin.In human umbilical vein endothelial cells,WEHAD significantly inhibited bFGF-induced proliferation,adhesion,migration,and capillary tube formation.We used an antibody array to perform an analysis of signaling proteins,which showed up-regulated expression of various proteins including RAD51,RAD52,and p73,and down-regulated expression of pFAK.Blood vessel formation in a chick chorioallantoic membrane (CAM) treated with WEHAD was markedly reduced in length compared with a PBS-treated control group.These results suggest that inhibition of angiogenesis by WEHAD may be the mechanism of action for the anti-cancer effects of HAD.

文摘Functional response is a key index in determining the population fluctuation in predation. However, the lack of operable research system limits the studies on functional response of fungal predators. Hirsutella rhossiliensis is a dominant parasite of the soybean cyst nematode, Heterodera glycines. In a soil microcosm bioassay, we determined fungal biomass at different days within 21 days after inoculation, and parasitism rate of H. glycines by the fungus was determined. The functional response of H. rhossiliensis to H. glycines was established and found to be Holling's type Ill, which was influenced by mycelial densities. Meanwhile, we conducted anti-fungal analysis of metabolic fractions extracted from H. rhossiliensis to explain the potential mechanism of the intraspecific competition illustrated by functional response. The result of anti-fungal experiments indicated that the fungal predators had more complicated interaction at population level than expected, which might be regulated by self-inhibition metabolite(s). This study was the first functional response study of fungal predators in microcosm. With the in- creasing recognition of emerging fungal threats to animal, plant, and ecosystem health, the methodologies and hypotheses proposed in this study might inspire further research in fungal ecology.
