Aim:To investigate the hypotensive effect and tolerability and the trough to peak ratio and the side effect of Dilacor(Dila) in essential hypertension.\ Method:After a 2 week placebo run in period,37 patients with dia...Aim:To investigate the hypotensive effect and tolerability and the trough to peak ratio and the side effect of Dilacor(Dila) in essential hypertension.\ Method:After a 2 week placebo run in period,37 patients with diastolic blood pressure (DBP) between 95 mmHg~115 mmHg and systolic blood pressure(SBP)≥200 mmHg were selected and were given Dilacor(180 mg/d ), the Dilacor was added to 240 mg/d if the DBP≥90 mmHg 4 weeks later, and patients were excluded if DBP≥90 mmHg after 4 week dosage addition, the study continued for 24 weeks. Ambulatory blood pressure (ABP) was monitored in 20 cases.\ Results:The DBP significantly decreased from 8th week to 24th week after Dilacor treatment compared with pretreatment, the SBP and DBP of the 24th week decreased by 10.47 mmHg and 6.64 mmHg respectively. The ABPM showed that the total pressure load, the mean SBP and DBP at day and night were lowered apparently, the trough to peak ratios of both SBP and DBP were greater than 50%.\ Conclusion:Dilacor, a long acting calcium antagonist, is an effective and safe hypotensive agent in mild and moderate essential hypertension.展开更多
