[Method] The conventional method and medium dilution method were used for bacterial, mold and yeast counting in sample recovery test. [Result] The medium dilution method (1:10 test solution, 0.5 ml/plate) could effectively eliminate the inhibition effect of the Bupi Qiangli Ointment, and the recovery of Staphylococcus aureus was greater than 70% in the 3 batches of samples; and the conventional method exhibited the recoveries of E. coil, Bacillus subtilis, Candida albicans and Aspergillus greater than 70% in the 3 batches of samples. [Conclusion] Due to Bupi Qiangli Ointment has strongly antibacterial effect on Staphylococcus au- reus, the medium dilution method was used for bacterial counting, and the conventional method was used for mold and yeast counting; and the conventional method was used for controlled bacterium examination of E. coll.展开更多

METHODS: Thyl populations were analyzed by fluorescence activated c...AIM: To investigate whether Thyl recognizes oval cells in the fetal liver and to characterize the cultured Thy1 selected cells from E14 rat livers. METHODS: Thyl populations were analyzed by fluorescence activated cell sorter analysis. Thyl positive cells were isolated using magnetic beads. Hepatic markers were detected by Western blotting, immunocytochemistry and RT-PCR. RESULTS: The percentage of Thyl-positive cells decreased during early development of fetal rat liver (E13-E16). E14 fetal livers contained 7.8% Thy1 positive cells, of which 61% were positive for α-fetoprotein (AFP) and 25% expressed albumin. The Thy1+ population expressed oval cell markers c-Kit and CXCR4, liver enriched-transcription factors HNF1α and HNF6, hepatocytic markers albumin, AFP and cytokeratin 18, and biliary marker cytokeratin 19. Thy1- selected cells formed only mesenchymal colonies when plated on collagen and in serum-containing media. Thyl selected cells were able to form hepatic colonies positive for HNF1α, HNF6, albumin, AFP, cytokeratin 18, cytokeratin 19 and glycogen, when grown on STO feeder layers in serum free-media. It effectively reduced nitrogen, phosphorus, chemical oxygen demand and biochemical oxygen demand in the water by 67.3%, 73.2%, 38.1% and 15.5%, respectively, during May to September 1998 when the water eutrophication was developing. This could control water eutrophication and produce pearls, shellfish meat and shells. This is an economical and effective way to control water eutrophication by using the ultra strong filtering capability of freshwater pearl mussels.展开更多
A gel microbead (GMD) cultivation method was employed to cultivate microorganisms from an amphioxus breeding zone in Qingdao, P. R. China. In th...A gel microbead (GMD) cultivation method was employed to cultivate microorganisms from an amphioxus breeding zone in Qingdao, P. R. China. The culture results were compared with those by standard plating method. In the GMD-based method, the microcolony-forming GMDs were sorted by fluorescence-activated cell sorting (FACS). To further get pure cultures, a subsequent enrichment culture and a streaking purification procedure were conducted on marine R2A medium. Eighty bacterial strains isolated by the GMD-based method were randomly selected for sequencing. These isolates belonged to Alphaproteobacteria (33%), Gammaproteobacteria (44%), Bacteroidetes (11%), Actinobacteria (5%), Firmicutes (5%), Epsilonproteobacteria (1%), and Verrucomicrobia (1%), the last two groups being usually difficult to culture. The 16S rRNA gene sequences revealed a diverse community with 91.1%-100% of the bacterial rRNAs similarities. Thirteen strains were sharing 16S rRNA gene sequence which was less than 97% similar to any other rRNA genes currently deposited in TYP16S database. Seventy isolates derived from the standard plating method fell into 4 different taxonomic groups: Alphaproteobacteria (9%), Gammaproteobacteria (81%), Bacteroidetes (7%) and Firmicutes (3%) with a 16S rRNA gene sequence similarities between 95.8%-100%, in which only 3 strains were sharing 16S rRNA gene sequence of less than 97%. The results indicated that the GMD-based method with subenrichment culture yielded more taxonomic groups and more novel microbial strains, including members of previously rarely cultured groups, when compared with the standard plating method, and that this method markedly improved the bacterial cultivability.展开更多

The plantlets exposed to vibration stimuli at all those frequencies have a larger total number and a larger total length of roots and a smaller permeability of root plasma-membrane, compared with those cultivated in an environment without vibration stress. Vibration at respectively 1 Hz, 2 Hz, 3 Hz and 4 Hz enhances root activity and the 3 Hz vibration is the most favorable. There is an obvious negative correlation between root activity and permeability of root plasma-membrane. The effects may be explained by the likelihood that mechanical Vibration at an appropriate frequency facilitates roots' absorbing water and minerals which are indispensable to inducing and synthesizing in roots some active substances favorable to growth. Nevertheless, overstress damages the integrity of root plasm-membrane, increases the permeability, and results in the disability of protecting root cells.展开更多

In this study, we determined the effects of various nitrogen sources, different nitrogen concentrations in the culture medium, and two culture methods on the growth of the green alga, Enteromorpha prolifera. The relationship between the specific growth rate of E. prolifera and NO3-N concentration was consistent with that estimated using the Monod equation (R2=0.9713, P〈0.01). In the N O3-N medium, the maximum specific growth rate was calculated to be 0.1634/d and the semi-saturation constant was calculated to be 16.86 μmol/L. Our results show that E. prolijkra can effectively utilize NH4^+-N, NO3^-N, and NO^2-N and urea-N in the range of 5 to 50 μmol/L. NH4^+-N was preferentially assimilated by E. prolifera, and urea-N was favorable for long-term growth.展开更多

Investigation into the optimal culture conditions needs a large number of experiments So it is desirable to minimize the number of experiments and maximize the information gained from them. The orthogonal design of experiments and mathematical statistical method are considered as effective methods to optimize the culture condition of magnetotactic bacteria WMol for high magnetic cells concentration. The effects of the four factors, such as pH value of medium, oxygen concentration of gas phase in the serum bottle, C:C (mtartaric acid: m=succinic acid) ratio and NaNO3 concentration, are simultaneously investigated by only sixteen experiments through the orthogonal design L16(44) method. The optimal culture condition is obtained. At the optimal culture condition ( pH 7.0, an oxygen concentration 4.0%, C:C (mtartaric acid: m=succinic acid) ratio 1:2 and NaNO3 100 mg 1^-1), the magnetic cells concentration is promoted tO 6.5×10^7 cells ml^-1, approximately 8.3% higher than that under the initial conditions. The pH value of medium is a very important factor for magnetic cells concentration. It can be Proved that the orthogonal design of experiment is of 90% confidence. Ferric iron uptake follows MichaelisoMenten kinetics with a Km of 2.5 pM and a Vmax of 0.83 min^-1.展开更多

Sterilized for 6 rain with 0.1% (w/v) mercuric chloride solution is an essential sterilization method for stems. June and July proved to be better for material taking, followed by May. The callus formation rate of balsam pears reached the highest when medium added with IBA 1.0mg/L and BA 2.0 mg/L, while the optimum medium for adventitious bud induction was MS added with IBA 0.5mg/L and BA 4.0 mg/L. It ensures interdisciplinarity, multi...GeoQuest Project is the design of a new teaching and learning technology based on Computer Class Role Playing Games (RPG) to teach Earth Science in a STEAM educational approach. It ensures interdisciplinarity, multilinguai approach (CLIL compliance) and interactive activities in classrooms. This educational game uses students' personal tablets and smartphones to interact with the game, with a system more comprehensible and funny for young people compared to traditional methodologies. Students are bringing "in situation" by the adventure, so teachers can get a rigorous assessment of their skills. The Game includes several adventure paths, suitable for different age students. They are focused on scientific topics, humanities, hands-on activities in interdisciplinary way. If used conditions do not allow practical or laboratorial activities provided by the game, players can see some videos to understand the activities. Experimental results were outstanding: the RPG appreciation is in agreement with the experiments outcome. This work experience gets excellent feedbacks by teachers, in terms of efficacy of this new teaching methodology and of results achieving.展开更多

Pre- and post-test on creativity using Torrance's Test of Creativity (TTCT) and Entrepreneurial Self-efficacy questionnaires were used to determine the level of both creativity and entrepreneurial self-efficacy The findings of the study show that a significant leap in how teaching and learning takes place is necessary if we are to properly prepare students for the 21st century knowledge economy and that active participation, confidence and ideas generation taking place under teamwork and informal settings are a way forward and to harness the two constructs.展开更多

The extraction of low concentration proteins from large volumes of culture media,...Secreted proteins are important sources for early detection and diagnosis of disease, and as such have received considerable attention. The extraction of low concentration proteins from large volumes of culture media, which are rich in salts and other compounds that interfere with most proteomics techniques, presents a problem for secretome studies. Ultrafiltration, precipitation, and dialysis are three major extraction methods that can be used to overcome this problem. The present study for the first time, compared the merits and shortcomings of these three methods, without bias. Centrifugal ultrafiltration provided the best extraction efficiency, and precipitation provided the highest number of identifiable proteins. The three methods yielded closely related, but different, information on the secretome; thus, they should be considered complementary or, at least, supplementary methods. Three hundred and sixty unique proteins were identified, including 211 potential secreted proteins. Compared with previous studies, this study also identified 42 new secreted proteins. The present study not only offers a reference for the selection of secretome extraction methods, but also expands the secretome database for the investigation of hepatocellular carcinoma.展开更多

In this paper, the reading-discussion-writing approach which guarantees the innovative construction of writing is proposed to train college students' critical thinking ability, and specific suggestions on the practice are articulated such as designing diversified listening and speaking lead-in activities to activate or supplement the relative notion and background knowledge, using variety of challenging reading-discussion-writing tasks, and conducting mutual evaluation, etc., to let students deeply understand the text through discussion and promote their language competence and writing ability as well as their reflective thinking ability.

The overall objective of military talent training is to cultivate the comprehensive-quality military personnel that meet the requirements of military modernization and preparations for military struggle. In this paper, we construct the military talent training objectives model, and on this basis, we propose the innovative military talent training mode, from the perspective of curriculum content system and talent quality assurance system.

文摘Secreted proteins are important sources for early detection and diagnosis of disease, and as such have received considerable attention. The extraction of low concentration proteins from large volumes of culture media, which are rich in salts and other compounds that interfere with most proteomics techniques, presents a problem for secretome studies. Ultrafiltration, precipitation, and dialysis are three major extraction methods that can be used to overcome this problem. The present study for the first time, compared the merits and shortcomings of these three methods, without bias. Centrifugal ultrafiltration provided the best extraction efficiency, and precipitation provided the highest number of identifiable proteins. The three methods yielded closely related, but different, information on the secretome; thus, they should be considered complementary or, at least, supplementary methods. Three hundred and sixty unique proteins were identified, including 211 potential secreted proteins. Compared with previous studies, this study also identified 42 new secreted proteins. The present study not only offers a reference for the selection of secretome extraction methods, but also expands the secretome database for the investigation of hepatocellular carcinoma.
