[Method] Using single factor and orthogo- nal experimental designs, the fermentation medium and culture conditions for B. sub- i tills B-332 were optimized through shaking flask culture. [Result] The optimal culture and inoculation time was 18 h, and the optimized medium formula was as follows: soybean powder 0.60 g/L, sucrose 0.25 g/L, ammonium sulfate 0.07 g/L, trisodium citrate 0.03 g/L, dipotassium hydrogen phosphate 0.003 g/L, magnesium sulfate 0.005 g/L, ferrous sulfate 0.000 5%; the fermentation condition was as follows: tem- perature 30 ℃, shaking bed speed 180 r/min, shaking bottle volume 80 ml/500 ml. In the optimal fermentation conditions, the fermented spore quantity was 1.43×10^11 cfu/ml, which was 34.48 times higher than the spore quantity of 4.03×10^9 cfu/ml in initial condition. The disease,pathogens of which are supposed to be bacteria by most researchers,is highly infectious and can often cause all individuals in the same culture pool to die in a very short time. The 16S rRNA gene phylogenesis of the culturable bacteria from the lesions of diseased individuals was conducted to study the biodiversity of the bacterial communities in the lesions and to identify probable pathogen(s) associated with this kind of disease. S. japonica samples were selected from a hatchery located in the eastern part of Qingdao,China. Bacterial universal primers GM5F and DS907R were used to amplify the 16S rRNA gene of bacteria colonies,and touchdown PCR was performed to amplify the target sequences. The results suggest that γ-proteobacteria(Alteromonadales and Vibrionales) of CFB group,many strains of which have been also determined as pathogens in other marine species,are the predominant bacterial genera of the diseased Apostichopus japonicus individuals.展开更多

A quadratic regression model of biomass to the doses of the above three factors was established. The model fit well and therefore the optimum fermentation condition was obtained. Responses of biomass to the three single factors and to their interactions were discussed. Thus, the highest level of biomass, 21.1 g/L, appeared under the optimized conditions when the initial KH\-2PO\-4, peptone and brown sugar were 1 g/L, 4 g/L and 72 g/L respectively.展开更多

Solid substrate fermentation (SSF) holds tremendous potential for the production of microbial enzyme...The importance of microbial enzymes in pulp and paper manufacturing has grown significantly in the last two decades. Solid substrate fermentation (SSF) holds tremendous potential for the production of microbial enzymes of commercial interest. SSF can be of special interest in those processes where the crude fermented product (whole SSF culture, in situ enzyme) may be used directly as the enzyme source. Xylanase preparations practically free of cellulase activity are especially useful for biobleaching of crude cellulose pulps. Thirty-nine Trichoderma isolates have been screened in SSF for xylanase production on hardwood oxygen-delignified soda-aq pulp as carbon source and enzyme inducer. Xylanase activities varied between 0 and 2200 IU/g dry matter (DM) of initial substrate. In most instances, the simultaneously produced cellulase levels were below 1.0 Filter Paper Unit (FPU) /g DM. The xylanase to cellulase activity ratio varied in the range of 5 to 3500. The three most promising isolates (TUB F-1647, TUB F-1658 and TUB F-1684) yielded xylanase activity of 2040, 1300 and 1500 IU/g DM xylanase, respectively, and 0.64, 0.43 and 0.43 FPU/g DM cellulase with a xylanase to cellulase activity ratio of 3200, 3000 and 3500, respectively. Wild strains F-1647, F-1658 and F-1684 were isolated from tree bark of Maldives, soils of Peru (last two), respectively. Medium optimization experiments to enhance the xylanase yield and to increase the xylanase to cellulase ratio have also been performed.展开更多

Central composite design (CCD) was used to study the interactive effect of culture conditions (temperature, pH, and inoculum) on cellulase activity. Results suggested that temperature and pH all have significant impact on cellulase production. The use of RSM resulted in a 96% increase in the cellulase activity over the control of non-optimized basal medium. Optimum cellulase production of 13 U/mL was obtained at a temperature of 42.24 ℃, pH of 5.25, and inoculum size of 4.95% (v/v) in a fermentation medium containing wheat bran, soybean meal and malt dextrin as major nutritional factors.展开更多

The aim of this study was to investigate the quality of fermented goat milk and cow milk, as well as the viability of LAB in the s...Lactic acid bacteria (LAB) is widely used as culture starters in dairy fermentation. The aim of this study was to investigate the quality of fermented goat milk and cow milk, as well as the viability of LAB in the same products. Fermentations were performed with pasteurized goat milk or cow milk added with skim milk (18% of solids) using three separately different starters; yoghurt starter (a combination of Streptococcus thermophilus FNCC-0040 and Lactobacillus bulgaricus FNCC-0041), single starter of Lactobacillus acidophilus FNCC-0029 and Lactobacillus casei FNCC-0051. The parameters observed were pH, acidity, nutritional quality including protein, fat and lactose content and product's viscosity. Acidity, pH and viability of LAB were also monitored during storage at refrigerated temperature (4 ℃) for 28 days. Results show that the different LAB starters did not affect the pH, acidity, lactose and protein content. Differences on LAB starters affected fat content and viscosity. The highest score of viscosity (30.00 Pa.s ± 7.02 Pa.s) was observed on products fermented by yoghurt starters, followed by products obtained using starter of L. acidophilus (17.7 ±11.4) and L. casei (8.62 ±0.35). Protein content, acidity, pH and viscosity were not significantly different between products obtained from goat milk and cow milk. Fat content in fermented goat milk was higher (5.03% ±0.62%) than in fermented cow milk (3.52% ±0.37%), however, lactose content was higher in fermented cow milk (5.16% ±0.40%) than in fermented goat milk (4.53% ±0.35%). Total LAB concentration in fermented cow milk during storage was 8.03± 0.52 logt0 cfu/mL, while in fermented goat milk was 7.81 loglo cfu/mL ± 0.67 loglo cfu/mL. There was a 10.83% decrease in LAB viability in fermented cow milk and 11.40% in fermented goat milk after 28 days of storage. In conclusion, quality of fermented milk is affected by the starters applied, raw milk source and storage period.展开更多
