Cell Apoptosis Induced by Oligosaccharide in Suspension Cultures of Taxus chinensis 被引量:2

Taxol production of Taxus chinensis (Pilger) Rehd. var. mairei (Lemee et Lévl.) Cheng et L. K. Fu induced by oligosaccharide from Fusarium oxysporum f. vasinfectum (Atkinson) Snyder et Hansen was studied in suspension cultures, and it was found that oligosaccharide triggered cell apoptosis. Under transmission electron microscope the following morphological changes were observed: cell shrinkage, condensation of cytoplasm, nuclear fragmentation, and the increase of high electron density bodies in vacuole in great quantity. In oligosaccharide_treated cells, agarose gel electrophoresis revealed that DNA was digested into oligonucleosomal fragments that were times of 200 bp appearing as DNA ladders. Control cells were in normal physiological state, they were intact, abundant in organelle and with integral nucleus DNA, and the rate of taxol biosynthesis in these cells was very low. After the oligosaccharide to the culture system, the defense system of cells was elicited and the secondary metabolism was strengthened, i.e. phenolics were accumulated in the medium, the activity of polyphenol oxidase (PPO) was increased quickly and secondary wall of cells was thickened. The activity of L _phenylalanine ammonia lyase (PAL), the critical enzyme of the phenylpropanoid pathway, was increased promptly 1 h after elicitation. The rate of taxol production was improved sharply and the maximal taxol concentration at 72 h was six times that of control. Appearance of cell apoptosis was accompanied with the highest concentration of taxol in suspension cultures. 展开更多

[ Method ] The paraffin sections of leaves of tested lily varieties were prepared and then observed under microscope, and the stomatal characteristics and moisture characteristics of tested lily varieties were measured. I Resaltl All the three ornamental lily varieties show isobilateral leaf, single layer of epicuticula and lower epidermis, and no obvious differentiation of palisade tissue and spongy tissue; their stomata distribute in lower epidermis, and the guard cells are dumbbell-shaped; all of these matedais present high moisture. For the leaf sick- ness, midrib sickness and mesophyll tissue sickness, the order was determined to be oriental lily 〉 Lilium/ongiflorum 〉 Asian lily; of the three types of ornamental lily, Ulium Iongiflorum has the largest stomatai aperture and Asian lily has the smallest; focusing the water potential and moisture, the turn was Asian lily 〉 oriental lily 〉 Lilium Iong'fflorum. [Objective] This study was conducted to establish a microbial limit test method for Bupi Qiangli Ointment. [Method] The conventional method and medium dilution method were used for bacterial, mold and yeast counting in sample recovery test. [Result] The medium dilution method (1:10 test solution, 0.5 ml/plate) could effectively eliminate the inhibition effect of the Bupi Qiangli Ointment, and the recovery of Staphylococcus aureus was greater than 70% in the 3 batches of samples; and the conventional method exhibited the recoveries of E. coil, Bacillus subtilis, Candida albicans and Aspergillus greater than 70% in the 3 batches of samples. [Conclusion] Due to Bupi Qiangli Ointment has strongly antibacterial effect on Staphylococcus au- reus, the medium dilution method was used for bacterial counting, and the conventional method was used for mold and yeast counting; and the conventional method was used for controlled bacterium examination of E. coll. 展开更多

AIM: To investigate whether Thyl recognizes oval cells in the fetal liver and to characterize the cultured Thy1 selected cells from E14 rat livers. METHODS: Thyl populations were analyzed by fluorescence activated cell sorter analysis. Thyl positive cells were isolated using magnetic beads. Hepatic markers were detected by Western blotting, immunocytochemistry and RT-PCR. RESULTS: The percentage of Thyl-positive cells decreased during early development of fetal rat liver (E13-E16). E14 fetal livers contained 7.8% Thy1 positive cells, of which 61% were positive for α-fetoprotein (AFP) and 25% expressed albumin. The Thy1+ population expressed oval cell markers c-Kit and CXCR4, liver enriched-transcription factors HNF1α and HNF6, hepatocytic markers albumin, AFP and cytokeratin 18, and biliary marker cytokeratin 19. Thy1- selected cells formed only mesenchymal colonies when plated on collagen and in serum-containing media. Thyl selected cells were able to form hepatic colonies positive for HNF1α, HNF6, albumin, AFP, cytokeratin 18, cytokeratin 19 and glycogen, when grown on STO feeder layers in serum free-media. CONCLUSION: Oval cells positive for Thyl are present in early liver embryonic stages. 展开更多

Our field experiments showed that the cultivation of pearl mussels formed a new biocoene composed of filamentous algae, protozoa, porifera and coelenterate. It effectively reduced nitrogen, phosphorus, chemical oxygen demand and biochemical oxygen demand in the water by 67.3%, 73.2%, 38.1% and 15.5%, respectively, during May to September 1998 when the water eutrophication was developing. This could control water eutrophication and produce pearls, shellfish meat and shells. This is an economical and effective way to control water eutrophication by using the ultra strong filtering capability of freshwater pearl mussels. 展开更多

The root development of Actinidia chinensis planUets was studied in exposure to environmental stress of mechanical vibration at respectively 1 Hz, 2 Hz, 3 Hz, 4 Hz and 5 Hz. The plantlets exposed to vibration stimuli at all those frequencies have a larger total number and a larger total length of roots and a smaller permeability of root plasma-membrane, compared with those cultivated in an environment without vibration stress. Vibration at respectively 1 Hz, 2 Hz, 3 Hz and 4 Hz enhances root activity and the 3 Hz vibration is the most favorable. There is an obvious negative correlation between root activity and permeability of root plasma-membrane. The effects may be explained by the likelihood that mechanical Vibration at an appropriate frequency facilitates roots' absorbing water and minerals which are indispensable to inducing and synthesizing in roots some active substances favorable to growth. Nevertheless, overstress damages the integrity of root plasm-membrane, increases the permeability, and results in the disability of protecting root cells. 展开更多
