[Method] On the basis of orthogonal experiment, effects of different culture conditions on recombinant bacteria E. coil BL21-pET-28a(+)-Imp were investigated. Based on the obtained optimal culture condition, effects of different induction conditions on the ex- pression level of Imp protein were explored. The expression level of Imp fusion pro- tein was analyzed by using SDS-PAGE and Gene Tools software. [Result] The re- sults showed that the optimal conditions for culture were at 37℃, pH 7.0, with liq- uid volume of 20% and oscillation speed of 200 r/min, for induction were at 37℃ for 6 h, with initial OD600 of about 1.5 and IPTG final concentration of 0.1 mmol/L. [Conclusion] The expression level of Imp achieved 70.98 mg/L under the optimal conditions. Optimized conditions for expression of Imp fusion protein in E. coil were determined.展开更多

Compared with the feeder layer of MEF cells, medium conditioned by Buffalo rat liver cells (BRL-CM) is able to maintain pluripotency and karyo-typic normality of ES cells only in short term cell propagation. Besides, ES cells grown in BRL-CM are also capable of aggregation with 8-cell embryos of Swiss strain and develop into germ line chimaeras. Modification to the method of aggregating ES cells with early embryos by making a hole in agar layer on the top of MEF feeder cells was shown to be more convenient and efficient than the conventional microdrop method.展开更多

A novel strain of Micrococcus sp.DUT_AHX,which was isolated from the sludge of a nitrobenzene(NB)-manufacturing plant and could utilize NB as the sole carbon source,was identified on the basis of physiological and bio...A novel strain of Micrococcus sp.DUT_AHX,which was isolated from the sludge of a nitrobenzene(NB)-manufacturing plant and could utilize NB as the sole carbon source,was identified on the basis of physiological and biochemical tests and 16S ribosomal DNA(rDNA)sequence analysis.It can grow at the temperature up to 40℃or in the presence of NaCl concentration up to 12 g/L in Luria-Bertani(LB)medium.The optimal degradation conditions are as follows:temperature 37℃,pH 7.0,and shaking speed 150 r/min.The strain involves a partial reductive pathway due to the release of ammonia and can also utilize 2-aminophenol as the sole carbon source.Furthermore,the enzyme activity tests show that crude extracts of NB-grown strain DUT_AHX mainly contain 2-aminophenol 1,6-dioxygenase activity.The exploitation of salt-tolerant bacteria will be a remarkable improvement in NB bioremediation and wastewater treatment at high salinity and high temperature.展开更多

Absolute real-time PCR technology was applied to determine the transcription numbers of the selected test genes in Porphyra yezoensis, which has been regarded as a potential model species in stress responding. According to the results of photosynthesis parameters, we observed that Y(II) and Fv/Fm were significantly affected when stress was imposed on the thalli of Porphyra yezoensis, but underwent almost completely recovered under normal conditions, which were collected for the following experiments. Then three samples, which were treated with different grade stresses combined with salinity, irradiation and temperature, were collected. The transcription numbers of seven constitutive expression genes in above samples were determined after RNA extraction and cDNA synthesis. Finally, a general insight into the selection of internal control genes during stress response was obtained. We found that there were no obvious effects in terms of salinity stress (at salinity 90) on transcription of most genes used in the study. The 18S ribosomal RNA gene had the highest expression level, varying remarkably among different tested groups. RPS8 expression showed a high irregular variance between samples. GAPDH presented comparatively stable expression and could thus be selected as the internal control. EF-la showed stable expression during the series of multiple-stress tests. Our research provided available references for the selection of internal control genes for transcripts determination of P. yezoensis.展开更多

Objective:The purpose of the study is to investigate the technical points,effects and complications of fiducial marker implantation within target areas before the CyberKnife treatment on body malignant solid tumors.Me...Objective:The purpose of the study is to investigate the technical points,effects and complications of fiducial marker implantation within target areas before the CyberKnife treatment on body malignant solid tumors.Methods:Five hundred and four cases of patients with body malignant solid tumors accepted fiducial implantation within target areas under CT guidance before the treatment of CyberKnife.Observe the complications and effect.Results:Among the 504 cases,500 cases successfully accepted the implantation(a success rate of 99.2%).158 patients felt pain at the punctured sites and 3 patients had tachycardia.33 patients had abdominal pain after the surgery due to a small amount of bleeding in the needle passage during liver puncturing process.Among the 19 lung cancer patients who accepted lung paracentesis,1 case had light pneumothorax and 1 case got light haemothorax.Among the 453 patients who accepted liver paracentesis,6 had fiducial migration.Conclusion:The method of fiducial implantation within target areas before treating body malignant solid tumor with CyberKnife is minimally invasive and comparatively secure.展开更多

The proposed converter consists of highly efficient non-isolated cell converters using bidirectional semiconductor power devices, and these cell converters are connected in ISOP (input series and output parallel). The non-isolated ISOP converter achieves high step-down ratio of D/N, operating N cell converters under the duty ratio olD. Availability of the proposed converter has been shown by developing the 48 V-12 V laboratory prototype using two 24 V-12 V cell converters. Design consideration for the 48 V-3 V multicellular converter using four 12 V-3 V cell converters has been also conducted, and the potential to approach the efficiency of 97% has been discussed. The proposed topology is suitable for the POL (point of load) converters in the highly efficient next generation DC distribution system for data centers.展开更多

Theoretically, there are numerous ways for two parenta...Homoploid hybrid speciation (HHS) involves the recombination of two differentiated genomes into a novel, func- tional one without a change in chromosome number. Theoretically, there are numerous ways for two parental genomes to recom- bine. Hence, chance may play a large role in the formation of a hybrid species. If these genome combinations can evolve rapidly following hybridization and sympatric situations are numerous, recurrent homoploid hybrid speciation is a possibility. We argue that three different, but not mutually exclusive, types of contingencies could influence this process. First, many of these "hopeful monsters" of recombinant parent genotypes would likely have low fitness. Only specific combinations of parental genomic con- tributions may produce viable, intra-fertile hybrid species able to accommodate potential constraints arising from intragenomic conflict. Second, ecological conditions (competition, geography of the contact zones or the initial frequency of both parent spe- cies) might favor different outcomes ranging from sympatric coexistence to the formation of hybrid swarms and ultimately hybrid speciation. Finally, history may also play an important role in promoting or constraining recurrent HHS if multiple hybridization events occur sequentially and parental divergence or isolation differs along this continuum. We discuss under which conditions HHS may occur multiple times in parallel and to what extent recombination and selection may fuse the parent genomes in the same or different ways. A series of novel wide bandgap small molecules(IFT-ECA, IFT-M, IFT-TH and IFT-IC) based on the A-D-A structure with indenofluorene core, thiophene bridge, and different electron-deficient end-capping groups, were synt...A series of novel wide bandgap small molecules(IFT-ECA, IFT-M, IFT-TH and IFT-IC) based on the A-D-A structure with indenofluorene core, thiophene bridge, and different electron-deficient end-capping groups, were synthesized and used as non-fullerene acceptors in organic solar cells. The influences of end-capping groups on the device performance were studied.The four materials exhibited different physical and chemical properties due to the variation of end-capping groups, which further affect the exciton dissociation, charge transport, morphology of the bulk-heterojunction films and device performance. Among them, IFT-IC-based device delivered the best power conversion efficiency of 7.16% due to proper nano-scale phase separation morphology and high electron mobility, while the devices based on the other acceptors achieved lower device performance(4.14% for IFT-TH, <1% for IFT-ECA and IFT-M). Our results indicate the importance of choosing suitable electron-withdrawing groups to construct high-performance non-fullerene acceptors based on A-D-A motif.展开更多
