低能N^+注入紫花苜蓿生物学效应初步研究 被引量:10

对N+注入紫花苜蓿Medicago sativa所引起生物学效应从生理生化层面进行了较系统的研究,结果显示:在低剂量范围内(0~2.08×1016N+/cm2),N+注入对紫花苜蓿种子存在当代刺激效应,所研究的几种生理生化指标相对CK都有所提高。剂量为6.24... 对N+注入紫花苜蓿Medicago sativa所引起生物学效应从生理生化层面进行了较系统的研究,结果显示:在低剂量范围内(0~2.08×1016N+/cm2),N+注入对紫花苜蓿种子存在当代刺激效应,所研究的几种生理生化指标相对CK都有所提高。剂量为6.24×1016~8.32×1016N+/cm2的N+注入对紫花苜蓿种子存在反常辐照损伤效应。即随着N+注入剂量增加,各生理生化指标先降,后升,再降。N+注入使紫花苜蓿过氧化物酶同工酶酶谱发生变异。处理组与对照组扩增出的相同谱带亦存在谱带荧光强度有差异的现象。另外,对N+注入束介导大豆基因组DNA转入紫花苜蓿做了初步研究,结果M2总性状突变率达到19.8%,并得到3株叶片粗蛋白含量较高的突变株(粗蛋白含量比对照高约0.5%)、1株高叶绿素含量的突变株(叶绿素含量比对照高33.3%)。 展开更多

[Objective] The aim of this study was to elucidate the quantitative traits of plants mutagenized by ion beam. [Method] The particular variation phenotypes, a- gronomic traits, and protein and wet gluten contents of progenies derived from the same ion beam induced mutant were investigated. [Result] Morphological polymor- phism existed in some individuals. Plant height, spike length and protein content were significantly influenced by ion beam, and effective tiller number and wet gluten content were moderately influenced. Multiple comparisons of all the indices within groups indicated genomic instability among these groups. Coefficient of variation im- plied the differences within group were very low. [Conclusion] Ion beam irradiation displayed characteristics of multi-directivity and non-directiveness. It aroused multiple variations in the same mutant. Instability among progeny indicates cells had different fate even in the same irradiated tissue. It may take several generations for mutants to stabilize particular phenotypes. The effects of ion beam irradiation may be the in- terrelated direct irradiation damage, indirect irradiation damage and late effect, such as bystander effect and adaptive response. 展开更多

Galectins, a family of β-galactoside-binding proteins, participate in both innate immunity and adaptive immunity. This study identified one novel galectin-related protein from half-smooth tongue sole Cynoglossus semilaevis, which was designated as CsGRP. The full-length cDNA of its encoding gene was 785 bp in length with a 528 bp open reading frame encoding a putative protein of 176 amino acids. The deduced CsGRP contained a putative 131-aa galactoside-binding lectin domain and 3 critical residues responsible for carbohydrate binding (R93, W109 and R114). Genomic structural analysis revealed that CsGRP consisted of five exons and four intzons. CsGRP showed 68% similarity with Poecilia latipinna GRP and 67% similarity wih Stegastes partitus GRP. CsGRP showed the highest expression level in liver, although its expression was detected in all tested tissues. When challenged with Vibrio harveyi, the expression of CsGRP was significantly down-regulated in liver (P〈 0.05). In addition, we found that in spleen and kidney of C. semilaevis, the CpG island of CsGRP showed significantly higher (P 〈 0.05) methylation level in disease-resistant family of C. semilaevis (DR-Cs) than in disease-susceptible ones (DS-Cs). Our results suggested that CsGRP may play important roles in the immune response of C. semilaevis. Moreover, DNA methylation patterns provided valuable data for understanding the relationship between epigenetic regulation and immunity, which would assist the animal genetic research and improve the animal breeding in the future. 展开更多

AIM To determine the role of G0/G1 switch gene 2(G0 S2) and its transcriptional regulation in palmitate-induced hepatic lipid accumulation.METHODS Hep G2 cells were treated with palmitate,or palmitate in combination with CCAAT/enhancer binding protein(C/EBP)β si RNA or G0 S2 si RNA. The m RNA expression of C/EBPβ,peroxisome proliferator-activated receptor(PPAR)γ and PPARγ target genes(G0 S2,GPR81,GPR109 A and Adipoq) was examined by q PCR. The protein expression of C/EBPβ,PPARγ,and G0 S2 was determined by Western blotting. Lipid accumulation was detected with Oil Red O staining and quantified by absorbance value of the extracted Oil Red O dye. Lipolysis was evaluated by measuring the amount of glycerol released into the medium. RESULTS Palmitate caused a dose-dependent increase in lipid accumulation and a dose-dependent decrease in lipolysis in Hep G2 cells. In addition,palmitate increased the m RNA expression of C/EBPβ,PPARγ,and PPARγ target genes(G0 S2,GPR81,GPR109 A,and Adipoq) and the protein expression of C/EBPβ,PPARγ,and G0 S2 in a dose-dependent manner. Knockdown of C/EBPβ decreased palmitate-induced PPARγ and its target genes(G0 S2,GPR81,GPR109 A,and Adipoq) m RNA expression and palmitate-induced PPARγ and G0 S2 protein expression in Hep G2 cells. Knockdown of C/EBPβ also attenuated lipid accumulation and augmented lipolysis in palmitate-treated Hep G2 cells. G0 S2 knockdown attenuated lipid accumulation and augmented lipolysis,while G0 S2 knockdown had no effects on the m RNA expression of C/EBPβ,PPARγ,and PPARγ target genes(GPR81,GPR109 A and Adipoq) in palmitate-treated Hep G2 cells. CONCLUSION Palmitate can induce lipid accumulation in Hep G2 cells by activating C/EBPβ-mediated G0 S2 expression. 展开更多

目的探究shRNA-Fascin基因对人结肠癌细胞SW-480侵袭和凋亡影响。方法利用慢病毒转染技术,构建Fascin的shRNA干扰表达载体,RT-qPCR和Western blot等方法检测Fascin基因和凋亡蛋白Caspase-3和Caspase-9的表达。Transwell试剂盒检测人结... 目的探究shRNA-Fascin基因对人结肠癌细胞SW-480侵袭和凋亡影响。方法利用慢病毒转染技术,构建Fascin的shRNA干扰表达载体,RT-qPCR和Western blot等方法检测Fascin基因和凋亡蛋白Caspase-3和Caspase-9的表达。Transwell试剂盒检测人结肠癌细胞的侵袭,以及AV-PI试剂盒检测人结肠癌细胞的凋亡。结果shRNA-Fascin载体慢病毒转染效率为95%以上。实验组SW-480中Caspase-3和Caspase-9的mRNA的相对表达量明显高于空白对照组,差异有统计学意义(P<0.05)。实验组SW-480中Caspase-3和Caspase-9的蛋白相对表达水平明显高于空白对照组,差异有统计学意义(P<0.05)。实验组SW-480细胞侵袭能力明显低于空白对照组,差异有统计学意义(P<0.05)。实验组SW-480细胞的凋亡率明显高于空白对照组,差异有统计学意义(P<0.05)。结论shRNA-Fascin可以明显的抑制人结肠癌细胞SW-480的侵袭,促进人结肠癌细胞SW-480的凋亡。 展开更多
