通过外加不同强度的电磁场处理滨梅茎段外植体,以促进其不定芽的增殖。结果表明:对滨梅外植体施加强度为97 kA/m磁场,处理10 min,可明显地促进不定芽的增殖,其增殖倍数是对照的2.3倍。将经过97 kA/m磁处理的外植体接种在培养基MS+ZT(2 m... 通过外加不同强度的电磁场处理滨梅茎段外植体,以促进其不定芽的增殖。结果表明:对滨梅外植体施加强度为97 kA/m磁场,处理10 min,可明显地促进不定芽的增殖,其增殖倍数是对照的2.3倍。将经过97 kA/m磁处理的外植体接种在培养基MS+ZT(2 mg/L)+IBA(0.2 mg/L)+Vc(30 mg/L)上,芽的增殖倍数与幼苗高度、鲜重、干重都优于接种在培养基MS+ZT(2 mg/L)+NAA(0.2 mg/L)+Vc(30 mg/L)和对照的情况,经97 kA/m磁处理的外植体接种于优化生根培养基1/2MS+NAA(0.1 mg/L)+IBA(0.1 mg/L)+牛血清白蛋白(30 mg/L),诱导的不定根平均根长(5.39±0.68)cm,显著高于对照。说明培养基的优化与一定强度的磁场能有效地促进滨梅组培苗的增殖分化与生长。 展开更多

【目的】提高栓皮栎体胚的再生与增殖能力。【方法】将栓皮栎不同发育阶段合子胚诱导的胚性愈伤组织及不同发育时期的体胚,分别接种于含不同组合激素的MS培养基上,在不同光照条件下培养,探讨影响栓皮栎体胚再生与增殖能力保持的因素。... 【目的】提高栓皮栎体胚的再生与增殖能力。【方法】将栓皮栎不同发育阶段合子胚诱导的胚性愈伤组织及不同发育时期的体胚,分别接种于含不同组合激素的MS培养基上,在不同光照条件下培养,探讨影响栓皮栎体胚再生与增殖能力保持的因素。【结果】随着继代次数的增加,胚性愈伤组织再生体胚能力呈下降趋势;6-BA与NAA配合使用可延缓继代过程中胚性愈伤组织再生体胚能力的下降,1.00mg/L 6-BA与0.25mg/L NAA是继代中保持胚性愈伤组织再生体胚能力的最佳培养基激素组合;胚性愈伤组织再生体胚需要自然散射光,强光及全程暗培养均不利于体胚再生;合子胚外植体发育程度影响胚性愈伤组织再生体胚能力,处于心形期及早子叶形期的幼嫩合子胚诱导的胚性愈伤组织,其再生体胚能力强于成熟子叶形合子胚诱导的胚性愈伤组织;体胚发育阶段越老,再生与增殖体胚能力越差,胚性愈伤组织及处于球形期的体胚,再生增殖体胚能力强于其他发育时期的体胚。【结论】选择处于心形期的幼嫩合子胚诱导胚性愈伤组织,然后在含1.00mg/L 6-BA与0.25mg/L NAA的MS培养基上于自然散射光下培养,可有效保持栓皮栎的体胚再生与增殖能力。 展开更多

从人胎肝cDNA文库中分离到一种cDNA ,推测的蛋白氨基酸序列具有明显的丝氨酸苏氨酸激酶结构域,可能编码一种新的丝氨酸苏氨酸激酶(命名为CPK ,cellproliferationassociationkinase) .CPKmRNA全长约3 0kb .RT PCR检测表明CPKmRNA在增殖... 从人胎肝cDNA文库中分离到一种cDNA ,推测的蛋白氨基酸序列具有明显的丝氨酸苏氨酸激酶结构域,可能编码一种新的丝氨酸苏氨酸激酶(命名为CPK ,cellproliferationassociationkinase) .CPKmRNA全长约3 0kb .RT PCR检测表明CPKmRNA在增殖活跃组织或细胞如人胚胎组织、肿瘤细胞株中呈高丰度表达,在成人组织则呈低丰度或不表达.利用PCR技术扩增大鼠同源cDNA ,以此为探针,Northern杂交发现CPK表达于多种成年小鼠组织,以脑组织丰度最高.小鼠2 3肝部分切除可迅速诱导CPKmRNA的表达,在术后2 4~4 8h达高峰,与2 3肝部分切除后细胞增殖期吻合.以小鼠同源cDNA片段为模板,合成RNA探针,原位杂交显示在胚胎发育期CPK低丰度表达在大多数组织,以神经系统组织变化最大,在第8d胚胎神经管内皮细胞中即出现表达,11~13d在端脑、脑膜、间脑、脊神经节表皮细胞、海马、小脑神经胶质细胞等多种神经细胞中表达且丰度较高,16d后这些组织的表达迅速下降到较低水平,表明CPK可能与神经系统的生长发育有一定关联.利用信号通路检测技术,观察到CPK的表达对MAPK ,p38MAPK途径的激活有明显的影响,并可显著增强表皮生长因子对这两条途径的激活,提示该激酶可能参与细胞因子的信号转导. 展开更多

Expression patterns and action analysis of genes associated with inflammatory responses during rat liver regeneration 被引量:4

AIM: To study the relationship between inflammatory response and liver regeneration (LR) at transcriptional level.METHODS: After partial hepatectomy (PH) of rats, the genes associated with inflammatory response were obtained according to the databases, and the gene expression changes during LR were checked by the Rat Genome 230 2.0 army. RESULTS: Two hundred and thirty-nine genes were associated with liver regeneration. The initial and total expressing gene numbers found in initiation phase (0.5-4 h after PH), G0/G1 transition (4-6 h after PH), cell proliferation (6-66 h after PH), cell differentiation and structure-function reconstruction (66-168 h after PH) of liver regeneration were 107, 34, 126, 6 and 107, 92, 233, 145 respectively, showing that the associated genes were mainly triggered at the beginning of liver regeneration, and worked at different phases. According to their expression similarity, these genes were classified into 5 groups: only up-regulated, predominantly up-, only down-, predominantly down-, up- and down-, involving 92, 25, 77, 14 and 31 genes, respectively. The total times of their up- and down-regulated expression were 975 and 494, respectively, demonstrating that the expressions of the majority of genes were increased, and that of a few genes were decreased. Their time relevance was classified into 13 groups, showing that the cellular physiological and biochemical activities were staggered during liver regeneration. According to gene expression patterns, they were classified into 33 types, suggesting that the activities were diverse and complex during liver regeneration. CONCLUSION: Inflammatory response is closelyassociated with liver regeneration, in which 239 LR- associated genes play an important role. 展开更多

Effect of Abscisic Acid on NaCI Stressed Callus Proliferation and Plant Regeneration in Rice

In rice, 2,4-Dichlorphenoxyacetic acid (MS2) has been considered best for callus induction and combination ofNAA, IAA, ABA (MS3) for somatic embryogenesis. Efficient plant regeneration was observed when MS basal salts supplied with 3% sorbitol and 3% maltose as carbon sources without hormones (MSac). During this experiment, effect of sodium chloride (NaCl) and abscisic acid (ABA) was assessed on callus growth, plant regeneration and root induction efficiency of rice (Oryza sativa L.) varieties IR-6 and Basmati-370. Callus proliferation rate was highly decreased in both varieties on MS2b (100 mol.m3 NaCl + 0.5 mg'L1 ABA) than MS2a (100 mol'm3 NaCl) cultures significantly. Proline, glycine-betaine and reducing sugars were increased significantly in MS2a and MS2b callusing cultures. However, total proteins were decreased in MS2a, while slightly increased in MS2b. Maximum plant regeneration (9.42 ±0.54 and 10.67 ±0.50 plantlets.callus1) from somatic embryos was observed on MS4c in IR-6 and Basmati-370, while 1.56 ± 0.06 (IR-6) and 0.95 ±0.05 (Basmati-370) plantlets callus1 were observed on MSab (100 mol·m3 NaCl). No plant regeneration was observed on MS4b (100 mol·m3 NaCl + 0.5 mg·L-1 ABA) medium in both varieties. Inhibition of root induction efficiency was high in MSsb (100 mol·m3 NaCl + 0.5 mg·L-1 ABA) than MS5a (100 mol·m3 NaCl) in the stressed cultures (P 〈 0.05). In this experiment, it was concluded that ABA involved in somatic embryogenesis and elevation of NaCl stress, while it causes inhibition of cell's growth as well as its regeneration into plantlets from somatic embryos. 展开更多

To investigate the effects of Ginse noside Rb 1 on the proliferation of Schwann cells in culture.Methods: Applying MTT assay and Thymidine incorporation assay, the effects of Ginsenoside Rb 1 on the proliferation of Schwann cells isolated from the sciatic nerve of adult rat were studied.Results: Ginsenoside Rb 1 (10 μg/ml) significantly induced Sc hwann cell proliferation, the effect was similar to NGF (50 μg/ml). At high con centrations of Ginsenoside Rb 1 (1 mg/ml), the proliferation of Schwann cells w as significantly inhibited. Conclusions: Ginsenoside Rb 1 at the optimal concentrations is found to be effective in inducing the proliferation of Schwann cells, but at hi gher concentrations the drug is cytotoxic for Schwann cells. 展开更多
