目的验证HEV编码的MicroRNA-A6对全长HEV瞬转HepG2细胞系中病毒复制的增效作用。方法按照已报道序列合成miR-A6,利用重组PCR、基因克隆以及体外转录技术获得单一高浓度的HEV RNA。HEV RNA与miR-A6按比例采用脉冲电流转染HepG2细胞,转染... 目的验证HEV编码的MicroRNA-A6对全长HEV瞬转HepG2细胞系中病毒复制的增效作用。方法按照已报道序列合成miR-A6,利用重组PCR、基因克隆以及体外转录技术获得单一高浓度的HEV RNA。HEV RNA与miR-A6按比例采用脉冲电流转染HepG2细胞,转染后24、48和96 h后观察细胞上清IgG分泌和HEV RNA表达载量,与不加miR-A6的HepG2细胞进行比较,分析miR-A6对HEV复制的作用,然后加入anti-A6抑制miR-A6表达,分析其对HEV表达和复制的影响。结果 MiR-A6∶HEV RNA质量比为0.5∶1和1∶1,共转染HepG2细胞后48 h,与单转染HEV RNA相比,lg值升高了1.57和2.44。HEV-IgG先于HEV RNA表达。体外实验显示,Ⅰ型和Ⅳ型HEV复制力没有明显差别,都在转染后48 h达到峰值,加入anti-A6抑制miR-A6后HEV-IgG抑制47.12%,HEV RNA抑制43.3%。结论 MiR-A6可以在体外显著增加HEV RNA瞬转HepG2细胞系中HEV病毒的复制力,为后续HEV分子病毒学和表型耐药研究奠定基础。 展开更多

The effect of heat treatment on microstructure and mechanical properties of the Ti-steel explosive-rolling clad plate was elaborated by optical microscopy (OM), scanning electron microscopy (SEM), X-ray diffraction (XRD), transmission electron microscopy (TEM), micro-hardness test and shear test. The composites were subjected to heat treatment at temperature of 650-950 ~C for 60 min. The results show that the heat treatment process results in a great enhancement of diffusion and microstructural transformation. The shear strength decreases as the treatment temperature increases. Heated at 850 ℃ or below, their shear strength decreases slowly as a result of the formation of TiC in the diffusion interaction layer; while at the temperature of 850 ℃ or above, the shear strength decreases obviously, which is the consequence of a large amount of Ti-Fe intermetaUics (Fe2Ti/FeTi) along with some TiC distributing continuously at diffusion reaction layer. 展开更多

A prototype of the master slave telerehabilitation robotic system with force feedback is developed. This system contains a pair of robots with the master being operated by the therapist and the slave following the master to guide the patients to exercise. A slave device with a slave controller is designed to stretch and mobilize the impaired elbow joints accurately and safely. A master device with a master controller is designed to control/monitor the procedure of treatment and assess the outcome of treatment remotely and accurately. By using the twoport network theory and the circuit equivalent impedance models, the position-force control scheme is designed to generate force feedback for the therapist who is to be informed of the interaction force between the subject and the robot arm during exercise. Experiments were conducted with a healthy male. Results show that the therapist can guide the patient to exercise by the master arm and can feel the interaction forces between the impaired arm and the robot. Compared with the traditional therapy, this system is more cost-efficient, more convenient and safer for both the stroke patients and the clinicians. 展开更多

Coxsackievirus A16(CVA16),together with enterovirus type 71(EV71),is responsible for most cases of hand,foot and mouth disease(HFMD) worldwide.Recent findings suggest that the recombination between CVA16 and EV71,and the co-circulation of these two viruses may have contributed to the increase of HFMD cases in China over the past few years.It is therefore important to further understand the virology,epidemiology,virus-host interactions and host pathogenesis of CVA16.In this study,we describe the viral kinetics of CVA16 in human rhabdomyosarcoma(RD) cells by analyzing the cytopathic effect(CPE),viral RNA replication,viral protein expression,viral RNA package and viral particle secretion in RD cells.We show that CVA16 appears to first attach,uncoat and enter into the host cell after adsorption for 1 h.Later on,CVA16 undergoes rapid replication from 3 to 6 h at MOI 1 and until 9 h at MOI 0.1.At MOI 0.1,CVA16 initiates a secondary infection as the virions were secreted before 9 h p.i.CPE was observed after 12 h p.i.,and viral antigen was first detected at 6 h p.i.at MOI 1 and at 9 h p.i.at MOI 0.1.Thus,our study provides important information for further investigation of CVA16 in order to better understand and ultimately control infections with this virus. 展开更多

Based on twin-roll casting, a cast-rolling force model was proposed to predict the rolling force in the bimetal solid-liquid cast-rolling bonding(SLCRB) process. The solid-liquid bonding zone was assumed to be below the kiss point(KP). The deformation resistance of the liquid zone was ignored. Then, the calculation model was derived. A 2D thermal-flow coupled simulation was established to provide a basis for the parameters in the model, and then the rolling forces of the Cu/Al clad strip at different rolling speeds were calculated. Meanwhile, through measurement experiments, the accuracy of the model was verified. The influence of the rolling speed, the substrate strip thickness, and the material on the rolling force was obtained. The results indicate that the rolling force decreases with the increase of the rolling speed and increases with the increase of the thickness and thermal conductivity of the substrate strip. The rolling force is closely related to the KP height. Therefore, the formulation of reasonable process parameters to control the KP height is of great significance to the stability of cast-rolling forming. 展开更多

Reactivation of hepatitis B is defined as the recurrence or an abrupt rise in hepatitis B virus(HBV) replication,often accompanied by an increase in serum transaminase levels,and both events occurring in a patient with a previous inactive hepatitis B infection.This reactivation can occur in situations in which the ratio of HBV replication and immune response is altered.It can happen during the treatment of hemato-oncological malignancies with chemotherapy and in immunosuppression of autoimmune diseases.Clinical manifestations of hepatitis B reactivation are variable and can range from asymptomatic to acute hepatitis,which are sometimes serious and result in acute liver failure with risk of death,and usually occur in the periods between cycles or at the end of chemotherapy.Immunosuppressive drugs such as corticosteroids or azathioprine can induce HBV reactivation in patients carrying hepatitis B virus surface antigen(HBsAg) or anti-HBc,but much less frequently than chemotherapy treatments.The tumor necrosis factorαinhibitors infliximab,etanercept and adalimumab may cause reactivation of hepatitis B,and the overall frequency with infliximab may be similar(50%-66%) to that caused by chemotherapy.Baseline HBV serology is recommended for all patients receiving chemotherapy and immunosuppressive drugs,and HBsAg positive patients should receive anti-HBV prophylaxis to decrease virus reactivation and death rates. 展开更多

The law of microstructure evolution and mechanical properties of hot roll bonded Cu/Mo/Cu clad sheets were systematically investigated and the theoretical prediction model of the coefficient of thermal expansion(CTE)of Cu/Mo/Cu clad sheets was established successfully.The results show that the deformation of Cu and Mo layers was gradually coherent with an increase in rolling reduction and temperature and excellent interface bonding was achieved under the condition of a large rolling reduction.The development of the microstructure and texture through the thickness of Cu and Mo layers was inhomogeneous.This phenomenon can be attributed to the friction between the roller and sheet surface and the uncoordinated deformation between Cu and Mo.The tensile strength of the clad sheets increased with increasing rolling reduction and the elongation was gradually decreased.The CTE of Cu/Mo/Cu clad sheets was related to the volume fraction of Mo.The finite element method can simulate the deformation and stress distribution during the thermal expansion process.The simulation result indicates that the terminal face of the clad sheets was sunken inward. 展开更多

A kind of Al-TiB2/TiC in situ composite with a homogenous microstructure was successfully prepared through in situ reaction of pure Ti and Al-B-C alloy with molten aluminum.In order to improve the distribution of the particles and mechanical properties of the composites,subsequent hot rolling with increasing reduction was carried out.The microstructure evolution of the composites was characterized using field emission scanning electron microscopy(FESEM)and the mechanical properties were studied through tensile tests and microhardness measurement.It is found that both the microstructure uniformity and mechanical properties of the composites are significantly improved with increasing rolling reduction.The ultimate tensile strength and microhardness of the composites with90%rolling reduction reach185.9MPa and HV59.8,respectively,140%and35%higher than those of as-cast ones.Furthermore,the strengthening mechanism of the composite was analyzed based on the fracture morphologies. 展开更多

Duplicate publication can introduce significant bias into a meta-analysis if studies are inadvertently included more than once. Many studies are published in more than one journal to maximize readership and impact of the study findings. Inclusion of multiple publications of the same study within a meta-analysis affords inappropriate weight to the duplicated data if reports of the same study are not linked together. As studies which have positive findings are more likely to be published in multiple journals this leads to a potential overestimate of the benefits of an intervention. Recent advances in immunosuppression strategies following liver transplantation have led to many studies investigating immunosuppressive regimes including immunosuppression monotherapy. In this letter we focus on a recently published meta-analysis by Lan et al investigating studies assessing immunosuppression monotherapy for liver transplantation. The authors claim to have identified fourteen separate randomised studies investigating immunosuppression monotherapy. Seven of the references appear to relate to only three studies which have been subject to duplicate publication. Several similarities can be identified in each of the duplicate publications including similar authorship, identical immunosuppression regimes, identical dates of enrolment and citation of the original publication in the subsequent manuscripts. We discuss the evidence of the duplicate publication inclusion in the meta-analysis. 展开更多

To weaken the nonlinear coupling influence among the variables in the speed and tension system of reversible cold strip mill, a compound control(CC) strategy based on invariance principle was proposed. Firstly, invariance principle was used to realize static decoupling between the speed and tension of reversible cold strip mill. Then, considering the influence caused by the time variation of steel coil radius and rotational inertia of the left and right coilers, as well as the uncertainties, a CC strategy that is composed of extended state observer(ESO) and global sliding mode control(GSMC) with backstepping adaptive was proposed,which further realized dynamic decoupling and coordination control for the speed and tension system. Theoretical analysis shows that the resulting closed-loop system is global bounded stable. Finally, the simulation was carried out on the speed and tension system of a 1422 mm reversible cold strip mill by using the actual data, and through the comparison of the other control strategies, validity of the proposed CC strategy was shown by the results. 展开更多

Flaviviral NS2B is a required cofactor for NS3 serine protease activity and plays an important role in promoting functional NS2B-NS3 protease configuration and maintaining critical interactions with protease catalysis substrates. The residues D80DDG in West Nile virus (WNV) NS2B are important for protease activity. To investigate the effects of D80DDG in NS2B on protease activity and viral replication, the negatively charged region D80DD and the conserved residue G83 of NS2B were mutated (D80DD/E80EE, D80DD/K80KK, D80DD/A80AA, G83F, G83S, G83D, G83K, and G83A), and NS3 D75A was designated as the negative control. The effects of the mutations on NS2B-NS3 activity, viral translation, and viral RNA replication were analyzed using kinetic analysis of site-directed enzymes and a transient replicon assay. All substitutions resulted in significantly decreased enzyme activity and blocked RNA replication. The negative charge of D80DD is not important for maintaining NS2B function, but side chain changes in G83 have dramatic effects on protease activity and RNA replication. These results demonstrate that NS2B is important for viral replication and that D80DD and G83 substitutions prevent replication; they will be useful for understanding the relationship between NS2B and NS3. 展开更多

AI-Cu/MWCNTs (multi-walled carbon nanotubes) nanocomposites were produced via compaction, sintering and hot extrusion process of AI-Cu/MWCNTs powders, which were fabricated by a conventional ball mill process with AI powders and Cu-MWCNTs composite powders which were synthesized by molecular level mixing technique. Also the change of mechanical properties with different content ratio of Cu/MWCNT composite powders in A1 matrix is analyzed. It is found that the addition of the proper Cu/MWCNTs powders which are well distributed in AI matrix leads to high mechanical stiffness. The 2 wt% Cu/MWCNTs reinforced A1 composites which exhibited 3.2 times higher tensile strength and 4.4 times higher yield strength than pure AI. 展开更多

Conventional optical burst switching(OBS)technique adopts purely statistical multiplex mechanismso that the bursts collide with each other very easily.To address this problem,a novel proactive con-tention avoidance scheme is proposed,which assigns dedicated wavelengths to each ingress node,then st-numbering algorithm is used to construct the traffic load balanced spanning trees .In this way,contentioncan be eliminated at ingress nodes,and the amount of bursts that could be accommodated by ingressnodes will be maximized.Further,those unused wavelengths left by traffic load balanced spanning treeare also organized as partial trees to carry bursts,thus the link utilization can be improved effectively.Simulation result shows that our scheme can improve the burst loss performance significantly without thewavelength converters or optical buffers comparing to other popular routing and wavelength assignment(RWA)algorithms. 展开更多

This is particularly so in the case... Delamination in composite structures can be a serious threat to the safety of the structure. Delamination leads to loss of stiffness and strength of laminates under some conditions. This is particularly so in the case of compressively loaded structures as the loss of stiffness may lead to separation of layers, the consequences of which can be catastrophic. Causes of delamination are many. In aerospace applications, this includes manufacturing defects, as well as operationally induced defects such as bird strikes, hits due to runway debris and tool drops. In this work one of the main causes of delamination that is dealt with, is the one that redistribution of the stress state due to some defects all mostly like here is broken single fiber (cutout fiber) in composite system already initiated by one of the above causes. When a laminate is subjected to in-plane tension, the effects of delamination on the stiffness and strength may be characterized by analytical results concerning the onset of delamination growth and its subsequent development. Many of the analytical treatments deal with just free-edge delamination. In mean time the redistribution and the gradient of the stress state in composite system is playing an important role for causing delamination. The main task of this work is to analyze single fiber with and without cutout embedded in matrix. Different FE models were generated, from the results, the redistribution of the stress state around the defected fiber were presented and discussed. Finally concluded remarks were indicated. 展开更多
