The variety is early-maturing, high-yield and good-quality, and has resistances to lodging, blast and bacterial leaf blight, its rice is sweet soft, goluptious and lucidus as well as not coarse when it is cold, so it is a good commodity. To further promote the popularization and cultivation of the variety, maintain its characteristics of high quality and high yield, and prevent commingling and degeneration, the purification and rejuvenation as well as breeding technology of high-quality seeds were proposed after continuous exploration and study.展开更多

[Method] The seeds of L. discolor were germinated in four differ...[Objective] This study was conducted to establish a system for aseptic seeding and rapid propagation of a wild Chinese herbal medicine Ludisia discolor. [Method] The seeds of L. discolor were germinated in four different culture media MS, 1/2MS, Hyponex No.1 (3.0 g/L), and 1/2 Hyponex No.1 (1.5 g/L), and the ger- mination rate was calculated 80 d later to select the optimal medium for seed ger- mination of L. discolor. Then, by culturing the protocorms and buds of L. discolor in the media supplemented with different hormone combinations (6-BA and NAA) or organic substrates (potato juice, banana juice and coconut milk), the optimal medi- um compositions for multiplication and .differentiation, seedling hardening and rooting of L. discolor were respectively determined. [Result] After 80 d of dark culture, 86.67% of the seeds inoculated into the medium 1/2 Hyponex No.1 germinated and developed into protocorms, proving 1/2 Hyponex No.1 was most suitable for seed germination of L. discolor among the four basic media, while MS medium had the worst effects. In culture medium of 1/2 Hyponex No.1+2.0 mg/L 6-BA+100 ml/L co- conut milk supplemented with 0.4 or 0.6 mg/L NAA, a mixed body of protocorms and small buds were induced after being cultured for 45 d, and the multiplication rate was 6.11, which was higher than that in the medium supplied with 0.2 or 0.8 mg/L NAA. By adding 10% potato juice, 10% banana juice or 100 ml/L coconut milk into Hyponex No.1+1.0 mg/L NAA, it was found that 10% banana juice had better effects in hardening and rooting of L. discolor planlets than 10% potato juice and 100 ml/L coconut milk. The small buds grew into plantlets about 80 d later in the medium with banana juice, the rooting rate was 93.33%, and the plantlet height was 7.19 cm. After hardening-culture for 10 d in a greenhouse, the L. discolor plantlets were transplanted into the mixed matrix of peat soil and lava (at a ratio of 3:1), and 91.7% of them survived 60 d later. [Conclusion] A relatively perfect propa- gation system of L. discolor was established by using aseptic seeds as explants in this study, which can be used for the protection of L. discolor germplasm resources, seedling propagation and industrial development.展开更多

The effects of solarisation using clear, UV stabilized, 25μm low density polyethylene mulching combined with soil amendment of chicken manures 12 th^-1, mixed fungicides of Metalaxyl 2 g-Benlate 1.5 g L^-l, Biocontro...The effects of solarisation using clear, UV stabilized, 25μm low density polyethylene mulching combined with soil amendment of chicken manures 12 th^-1, mixed fungicides of Metalaxyl 2 g-Benlate 1.5 g L^-l, Biocontrol agent of Trichoderma harzianum (T.h.) and NPK fertilizer 180 Kg h^-1, on the survived micro and macrocondia of Fusarium proliferatum (Matsushita) Nirenberg were ascertained during summer 2008. Mulched treatments within 45 and 60 days significantly reduced viable propagules to 6 and 3.4 × l03 cfu1 g soil, respectively at 5 cm depth followed by 15 and 30 cm soil depth. Natural heating of dry soil reduced an initial population of 76 × 10^3 to 46.73, 49 and 49.13 × 103 cfu'lg soil at 5, 15 and 30 cm depth, respectively. Therefore, Fusarium proliferatum reduced by 85.29 and 89.22% within 45 and 60 days compared to 38% in control aired soil. Application of mulching with T. harzianum caused a worthwhile reduction 84.37% in viable propagules, solar with low doses of Metalaxyl-Benlate further reduced Fusarium propagules at all depths 95.5%. Combining chicken manures proved its reduction of fungi cfu at 5 cm depth only, whereas NPK amendments failed in their reduction effects at various depths. However, almost 95% reduction in Fusarium propagules was achieved at various soil depths when combining Met.-Ben., with soil mulching. Combining T.h. was also proved to be significant in reducing fungus inoculum by 88.58, 96.06% at 5 and 15 cm depth. Generally, manures and NPK fertilizers amended soil found equally effective in reducing cfu of Fusarium at 5 and 15 cm depth similar to polyethylene mulching for both solar duration.展开更多

Proper parameter setting is vital to its exact use. To get guidelines for parameter setting, lowintensity ultrasound stimulation on the proliferation and reproductivity of Hep G2 and 3T3 cells in vitro was examined with a 1.06 MHz-generator by changing the parameters(including intensity, pulse repetition frequency and duty cycle)in a wide range. Cell viability and reproductivity at different time after sonication were measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)and colony formation assay to indicate timerelated proliferation. The results illustrate that ultrasound irradiation at 0.4—0.8 W/cm^2 and high pulse repetition frequency(100 Hz)can facilitate cell proliferation, while above 0.8 W/cm^2 would resist it. The extent of resistance closely correlated with duty cycle and pulse repetition frequency. Resistance effect at low pulse repetition frequency(1 Hz)is greater than that at high pulse repetition frequency(100 Hz)and not time-related. The influence of high pulse repetition frequency is time-accumulated, indicating cellular process involved. These findings would provide valuable guidelines for the application of low-intensity ultrasound in stem cell transformation and tissue engineering.展开更多
