基于生理的动力学(physiologically based kinetic,PBK)模型是一种用于模拟化学物质在有机体(如人类和其他动物物种)中的生物过程(吸收、分布、代谢和排泄,简称ADME)和推导体内、外剂量或靶器官剂量的工具。相较于PBK在药物研发的不同...基于生理的动力学(physiologically based kinetic,PBK)模型是一种用于模拟化学物质在有机体(如人类和其他动物物种)中的生物过程(吸收、分布、代谢和排泄,简称ADME)和推导体内、外剂量或靶器官剂量的工具。相较于PBK在药物研发的不同阶段的广泛应用,近几年来PBK在化学物质管理中应用的发展更为迅速。在过去十几年中,化学品安全管理领域的不同国际组织和权威机构相继发布了关于合理构建和使用PBK模型的指导文件。本文主要总结了人类/哺乳动物的PBK模型构建、化学物质健康风险评估中常用的软件和平台,以及PBK模型软件在化学物质管理中的应用和相关案例,简述了基于PBK和体外-体内外推(IVIVE)方法在下一代健康风险评估中的应用潜力,并对PBK在中国化学物质管理应用中需要解决的科学问题进行了讨论,以期为PBK在我国化学物质管理应用提供借鉴。展开更多
[Objective] The aim was to study the explants sterilization and callus induction of Aquilegia oxysepala.[Method] the seeds of Aquilegia oxysepala were sterilized by different kinds and concentrations of disinfectants,...[Objective] The aim was to study the explants sterilization and callus induction of Aquilegia oxysepala.[Method] the seeds of Aquilegia oxysepala were sterilized by different kinds and concentrations of disinfectants,and the pollution rate and pollution speed were investigated so as to find the best way to build sterile seedling setup.Taking the roots,stem segments and leaves of the sterile seedlings from Aquilegia oxysepala seeds as explants,the optimum explants and medium were screened by adding MS basic medium with different hormone proportions.[Result] The best germicidal treatment was as follows:explants were soaked in 75% alcohol for 30 s firstly,washed by sterile water for 5 times,then soaked in 0.2% mercuric chloride liquid for 2 min,finally washed by sterile water for 5 times again.The sterilization treatment could get the lowest pollution rate,the highest germinating capacity and the best sterile seedling.Roots were the optimum explants for the callus induction of Aquilegia oxysepala,meanwhile the optimal medium was MS+0.6 mg/L 2,4-D+0.5 mg/L 6-BA.[Conclusion] The research provides technical support for the large scale production of Aquilegia oxysepala and also makes a contribution to the medicinal and ornamental value of Aquilegia oxysepala.展开更多

Methods: Peripheral blood Th and Tc cell subpopu-lations were examined by three-color flow cytometry in 30 ...Objective: To understand the change of Th1/Th2 and Tc1/Tc2 balance in patients with secondary early syphilis. Methods: Peripheral blood Th and Tc cell subpopu-lations were examined by three-color flow cytometry in 30 patients with secondary early syphilis and 20 healthy subjects. Results: Percentages of CD3+ CD4+ T cells and the ratio of CD4+CD8+ and NK cells in syphilitic patients were significantly lower than in the control group (P<0.001), but no statistical difference was found in percentages of CD8+ and CD19+ cells between the two groups (P > 0.05). The percentage of Th1 cells and ratios of Th1/Th2 and Tc1/Tc2 in syphilitic patients were clearly decreased as compared to those in controls (P<0.001-0.01), whereas percentages of Tc0 and Tc2 cells in syphilitics was higher than those in controls(P<0.001). Conclusion: There could be a disturbance of cellular immunity and the relatively intact function of humoral immunity in secondary syphilis. It is postulated that the polarization toward development of Th2 and Tc2 cells and their associated cytokines might be involved in the incomplete clearance of treponemes and the development of chronic infection.展开更多
