Tm experiment showed that the compounds had interacted with RNA, and CD spectra also observed the changes of RNA conformation induced by the compounds. Here we report a highly efficient approach to induce human embryonic stem (ES) cells and induce...Human pluripotent stem cells represent a potentially unlimited source of functional pancreatic endocrine lineage cells. Here we report a highly efficient approach to induce human embryonic stem (ES) cells and induced pluripo- tent stem (iPS) cells to differentiate into mature insulin-producing cells in a chemical-defined culture system. The differentiated human ES cells obtained by this approach comprised nearly 25% insulin-positive cells as assayed by flow cytometry analysis, which released insulin/C-peptide in response to glucose stimuli in a manner comparable to that of adult human islets. Most of these insulin-producing cells co-expressed mature β cell-specific markers such as NKX6-1 and PDX1, indicating a similar gene expression pattern to adult islet β cells in vivo. In this study, we also demonstrated that EGF facilitates the expansion of PDXl-positive pancreatic progenitors. Moreover, our protocol also succeeded in efficiently inducing human iPS cells to differentiate into insuIin-producing ceils. Therefore, this work not only provides a new model to study the mechanism of human pancreatic specialization and maturation in vitro, but also enhances the possibility of utilizing patient-specific iPS cells for the treatment of diabetes.展开更多

When the...We report the oscillating propagation of kink in a nondissipative Frenkel-Kontorova (FK) chain driven by external DC force, which is different from the usual propagation of localized modes with equal speed. When the kink moves in the opposite direction of the external DC force, the kink will be accelerated and the potential of the FK chain in the external force field is transformed to be the kinetic energy of the kink. If the kink reaches the boundary of the FK chain, the kink will be bounced back and moves in the opposite direction, then the kink will be decelerated gradually and the kinetic energy of the kink & transformed to be the potential of the FK chain in the external force field. If the speed of the kink reaches zero, the kink will move in the opposite direction again driven by the external DC force, and a new oscillating cycle begins. Simulation result demonstrates exactly the transformation between the kinetic energy of the kink and the potential of the FK chain in the external force field. The interesting energy exchange is induced by the special topology of kinks, and other localized modes, such as breathers and envelope solitons, have no the interesting phenomenon.展开更多

Methods: Human ovarian cancer HO-8910 cells were cultured in vitro. The inhibitory effect of SG1-1776 combination with DDP in sub-toxic concentration on induction on viability of human ovarian cancer HO-8910 cells was evaiuated by the MTT assay. Cell apoptosis rate was analyzed by flow cytometry. The proteins expression level related to apoptosis were analyzed by Western blot. Results: SG1-1776 combination with DDP in sub-toxic concentration significantiy inhibited the proliferation of human ovarian cancer HO-8910 cells, and proliferation inhibition rate was increased drastically compared with normai saline (NS) group or DDP group in sub-toxic concentration or SG1-1776 group in sub-toxic concentration (P 〈 0.01). Apoptosis rate markedly increased after the treatment of SG1-1776 combination with DDP in sub-toxic concentration for 48 h. Western blot showed that the expression of bcl-2 protein was down-regulated and protein level of Bax and Cyto-c were depressed by SG1-1776 combination with DDP in sub-toxic concentration. Cenclusion: SG1-1776 combination with DDP in sub-toxic concentration could inhibit the cell proliferation and lead to cell apoptosis inhuman ovarian cancer HO-8910 cells, and its mechanism may be related to through mitochondrial apoptotic pathway.展开更多
