Objective The purpose of this experiment was to investigate the effects of exogenous hormone factors on subculture multiplication and root induction of tis-suecultured seedlings in mulberry. [Method] The aseptic seedlings of Guisangyou 12 were used as the materials and the comparative experiment on subculture multi-plication and root induction used different kinds,concentrations and combinations of exogenous hormones. [Result] The hormone combination of 0.1 mg/L IBA+1.5 mg/L 6-BA+0.03 mg/L TDZ showed the best effects to bud multiplication, and the multiple of bud multiplication was 5.28. The effects of NAA was better than IBA and IAA on root induction. The hormone combination of 2.0 mg/L NAA+1.0 mg/L PP333 was most suitable to the root induction, in which the rooting rate arrived to 100%, the root number was 7.01 and the root length was 1.38 cm on average. [Conclusion] The results wil provide some technical reference for large-scale propagation of mul-berry seedlings in vitro. 展开更多

It has been generally held in botany that Oryza sativa L. is a monocotyledon. Based on studies of rice embryo development we confirmed that rice embryo has two dimorphic cotyledons rather than just one cotyledon. In the present study we attempt to know if the morphology of embryos in other species of Oryza differs from O. sativa and if these embryos have dimorphic cotyledon. Two types of embryo structures were observed in 22 species and/or subspecies of genus Oryza under the scanning electron microscope. Type 1, the O.sativa type, which is characterized by ventral scale and lateral scales, was found in 16 species. Type 2, the O. meyeriana (Zoll. et Mor. ex Steud.) Baill. ssp. tuberculata W. C. Wu et Y. G. Lu, G. C. Wang type, with no ventral scale and lateral scales, was found in 6 species and subspecies. The embryogenic process of O.sativa and O.meyeriana sub. tuberculata showed that the scutellum primordium, coleorhiza primordium, coleoptile primordium and shoot apical meristem directly differentiate from proembryo. The former two later develop into the embryo envelope, which is the outside cotyledon; the coleoptile primordium develops into the coleoptile with the shape of inverted empty cone surrounding and covering the growth cone, which is the apical cotyledon. Both types of rice embryos have dimorphic cotyledons. The structural difference between them is that the scutellum primordium of the young embryo in type 2 does not differentiate ventral scale and lateral scales while the embryo of type 1 does. The dimorphic cotyledons of embryo of Oryza plants originate from the dorsiventrality of proembryo. 展开更多

Juss. is a species from the Brazilian Cerrado that produces edible fruits and, in common with other species from the Byrsonima genus, has pharmacological potential. Previous attempts to propagate the species through conventional methods showed difficulties. Thus, the purpose of this work was to characterize pro-embryogenic masses of Byrsonima intermedia callus, aiming for their in vitro propagation through somatic embryogenesis. Leaf segments from in vitro germinated seedlings were employed as explants for callus production. The calli were then subcultured and exposed to dyes to fulfill their embryogenic potential. Digitalizations of the cytological preparations were made in order to measure the area that was stained by both Aceto-Carmine and Evans-Blue, using image tool software. Somatic embryos were induced after treatments with l-naphthaleneacetic acid (NAA). The percentages of double-colored areas (by Aceto-Carmine and Evans-Blue) were calculated and the data were analyzed by using the Skott-Knott test (P ≤ 0.05) and, the embryogenic callus, as well as the formation of somatic embryos were analyzed by using the Krsuskal-Wallis rank test (P ≤0.05). The results show that double coloration is effective at identifying cells showing embryogenic potential. Excised zygotic embryos, cotyledons and hypocotyls of juvenile seedlings of masson pine were grown on DCR medium supplemented with several concentrations of various plant phytohormones. BA (1.0 mg/ L) in combination with NAA (0.05 mg/L)in DCR medium was found to increase the formation of adventitious buds from mature zygotic embryos, but most of them were formed at the tips of embryonic cotyledons. Adventitious buds were obtained from cotyledons and hypocotyls from juvenile seedlings when they were cultured on DCR medium containing BA 3-5 mg/L and NAA 0.1-0.2 mg/L. Elongation of buds were observed on hormone-free DCR medium with or without activated charcoal (0.5%). Root initiation was achieved with full or half strength DCR inedium supplemented with IBA 1.0 mg/L and NAA 0.25-0.5 mg/L. Approximately 11-20 axillary buds formed on each explant when juvenile seedling explants were treated (3-20h) with BA 50-100 mg/L, followed by transfer to hormone-free DCR medium. In this study, a thermophilic oil-degrading bacterial consortium KO8-2 growing within the temperature range of 45--65℃ (with 55℃ being the optimum temperature) was isolated from oil-contaminated soil of Karamay in Xinjiang, China. Denaturing gradient gel electrophoresis (DGGE) showed that there were nine strains included in KO8-2, which originated from the genera of Bacillus, Geobacillus and Clostridium. They all belonged to thermophilic bacteria, and had been previously proved as degraders of at least one petroleum fraction. The crude oil degraded by KO8-2 was analyzed by infrared spectrophotometry, hydrocarbon group type analysis and gas chromatography. The results indicated that the bacterial consortium KO8-2 was able to utilize 64.33% of saturates, 27.06% of aromatics, 13.24% of resins and the oil removal efficiency reached up to 58.73% at 55 ~C when the oil concentration was 10 g/L. Detailed analysis showed that KO8-2 was able to utilize the hydrocarbon components before C19, and the n-alkanes ranging from C20--C33 were signifi- cantly degraded. The ratios of nC17/Pr and nC18/Ph were 3.12 and 3.87, respectively, before degradation, whereas after degradation the ratios reduced to 0.21 and 0.38, respectively. Compared with the control sample, the oil removal efficiency in KO8-2 composting reactor reached 50.12% after a degradation duration of 60 days. 展开更多

The varied clinical presentations of fascioliasis still make a high index of suspicion mandatory. Besides having a wide spectrum of hepatobiliary symptoms like obstructive jaundice, cholangitis and liver cirrhosis, the parasitic infection also has extrabiliary manifestations. Until recently, extrahepatic fascioliasis has been reported in the subcutaneous tissue, brain, lungs, epididymis, inguinal lymph nodes, stomach and the cecum. In vitro Axillary Bud Multiplication of Citrus Nobilis Lour. in Indonesia

This research aimed to get the best composition ofcytokinin (BAP) and auxin (NAA) concentration on in vitro axillary bud multiplication of Tawangmangu orange (Citrus nobilis Lour.). The research was conducted in Plant Physiology and Biotechnology Laboratory of Agriculture Faculty, Sebelas Maret University Surakarta from February 2008 to September 2008, used factorial design with two factors based on Completely Random Design. The first factor was BAP concentration consisting of five levels: 0, 0.5, 1, 1.5, 2 ppm and the second factor was NAA concentration consisting of five levels: 0, 0.5, 1, 1.5, 2 ppm. So there were 25 combination treatments, each of them was repeated for three times. The result observation data of shoot initiation time and number of shoot were analyzed with analysis of variance and if significant continued with Duncan's Multiple Range Test at 5% level. Data of shoot color and amount of leaves were analyzed descriptively. The result showed that there was interaction between BAP and NAA on shoot initiation time. Combination of 0.5 ppm BAP and 1.5 ppm NAA was able to accelerate the shoot initiation time and produced the largest number of leaves that was eight leaves. BAP was able to increase the number of shoots. Average largest amount of shoot on 0.5 ppm BAP treatment was 3.93 shoots/explant. The dominant color of leaves was light green. 展开更多

The purpose of this work is to perform a detailed study of carbohydrate specificity of the new extracellular bacilli lectins which is considered to determine mechanisms of the lectins action. Sources of lectins were bacterial strains from Ukrainian collection of microorganisms. The optimized protocol of bacilli lectins isolation and purification included precipitation with ammonium sulfate with subsequent gel filtration chromatography on Sepharose CL-6B. Hemagglutinating activity of bacilli lectins and their fine carbohydrate specificity to sialic acids and their derivatives as well as sialic asid-containing and asialic glycoconjugates were studied. The ability of extracellular bacilli lectins to discriminate a- and 13-conformation of carbohydrate molecule and the type of connection between the monomers was determined. Studied lectins showed the most affinity to glycoconjugates containing both types of sialic acids (N-acetylneuraminic acid (Neu5Ac) and N-glycolylneuraminic acid (NeuGc)) and it is supposed to be a basis of their diagnostic and analytical potential. 展开更多

黄河故道地区现有的苹果,绝大部分为解放以后定植,尤以1958—1960年定植的最多,目前树龄大多为5—7年生,正值整形与定形阶段,迫切需要适合于本地区的整形修剪技术。几年来,各地都有一些园艺场以自力更生的精神,结合生产斗争,开展了科学... 黄河故道地区现有的苹果,绝大部分为解放以后定植,尤以1958—1960年定植的最多,目前树龄大多为5—7年生,正值整形与定形阶段,迫切需要适合于本地区的整形修剪技术。几年来,各地都有一些园艺场以自力更生的精神,结合生产斗争,开展了科学实验,摸出了不少可贵的经验。调查总结这些果园的经验,对于指导本地区的整形修剪有重大义意。为此,我们于1965年结合样板田工作,先后调查了江苏省丰县大沙河果园、徐州果园、安徽省肖县黄河故道园艺场、砀山果园场、河南省兰考县仪封园艺场、开封市百塔果园、西华黄泛区农场等七个国营果园及民权人和园艺场和兰考马庄园艺场等二个群营果园的整形修剪技术。 解放以后定植的苹果,绝大部分均属主干形,目前在本地推广的是主干疏层形,树体结构的要求是:干高50—60厘米,树高4—5米。第一层3个主枝,层内距20—40厘米。第二层1—2个主枝,第三、四层各1个主枝,全树总计5—7个主枝,分三—四层排列。第一—二层间距离80—100厘米(山地、瘠地60—80厘米)。第二层以上的层间距离60厘米左右。第一层主枝角度50°—60°,第二层以上45°左右。第一层每个主枝上配置侧枝2─4个,互生。第一侧枝距干60—70厘米,其余各侧枝相互距离40—50厘米。 黄泛区农场土壤深厚肥沃,树冠较大,该场制? 展开更多
