To study the expression of cystatin C (CysC), its regulation by transforming growth factor-β1 (TGF-β1) and platelet-derived growth factor (PDGF) and the potential interference of CysC with TGF-β1 signaling in this special cell type. METHODS: We evaluated the CysC expression in cultured, profibrogenic hepatic stellate cells and transdifferentiated myofibroblasts by Northern and Western blotting and confocal laser scanning microscopy. RESULTS: CysC was increased significantly in the course of trans-differentiation. Both TGF-β1 and PDGFBB suppressed CysC expression. Furthermore, CysC secretion was induced by the treatment with TGF-β1. Although CysC induced an increased binding affinity of TGF-β receptor type Ⅲ (beta-glycan) as assessed by chemical cross-linking with [^125I]-TGF-β1, it did not modulate TGF-β1 signal transduction as shown by evaluating the Smad2/3 phosphorylation status and [CAGA]-MLP-luciferase reporter gene assay. Interestingly, the shedding of type Ⅲ TGF-β receptor beta-glycan was reduced in CysC-treated cells. Our data indicated that CysC expression was upregulated during transdifferentiation. CONCLUSION: Increased CysC levels in the serum of patients suffering from liver diseases are at least partially due to a higher expression in activated hepatic stellate cells. Furthermore, TGF-β1 influences the secretion of CysC, highlighting a potentially important role of cysteine proteases in the progression of hepatic fibrogenesis. 展开更多

Fatty acids(FAs) provide energy and also can be used to trace trophic relationships among organisms.Sea cucumber Apostichopus japonicus goes into a state of aestivation during warm summer months.We examined fatty acid profiles in aestivated and non-aestivated A.japonicus using multivariate analyses(PERMANOVA,MDS,ANOSIM,and SIMPER).The results indicate that the fatty acid profiles of aestivated and non-aestivated sea cucumbers differed significantly.The FAs that were produced by bacteria and brown kelp contributed the most to the differences in the fatty acid composition of aestivated and nonaestivated sea cucumbers.Aestivated sea cucumbers may synthesize FAs from heterotrophic bacteria during early aestivation,and long chain FAs such as eicosapentaenoic(EPA) and docosahexaenoic acid(DHA) that produced from intestinal degradation,are digested during deep aestivation.Specific changes in the fatty acid composition of A.japonicus during aestivation needs more detailed study in the future. 展开更多

We have quantitatively analyzed the tree species diversity with respect to soil nutrient status in three sites of a sacred forest ecosystem of Niyamgiri hill range, Eastern Ghats, India. Extensive field surveys and sampling were conducted in 3 sites of the hill range: Site 1 Pterocarpus dominated forest (PTF) (19°40'02.2'' N and 83°21'23.1'' E), Site 2 Mangifera dominated forest (MAF) (19°40'02.8'' N and 83°21'40.8'' E) and Site 3 Mixed forest (MIF) (19°36'47.1" N and 83°21'02.7'' E). A total of 28 families, 42 genera, 46 tree species, and 286 individual trees were recorded on an area of0.6 ha. Tree density varied between 470 and 49o individuals ha and average basal area between 3.16 and l0.04 m2 ha-1. Shannon Index (H') ranged from 2.34 to 4.53, Simpson's Index ranged from 0.07 to o.09, and equitability Index ranged from 0.7 to 1.34. The number of individuals was highest in the girth at breast height (GBH) class of 50-7o cm. The soil nutrient status of the three forest types was related to tree species diversity. The soil pH value of the three sites reflected the slightly acidic nature of the area. Species diversity was positively correlated with organic carbon and phosphorus and negatively with nitrogen, EC and pH. The results of the current study may be helpful to further develop a conservation planfor tree species in tropical sacred forest ecosystems. 展开更多

The environmental quality status of Daya Bay (22.56-22.77°N, 114.51-114.73°E), a main aquaculture area in Guangdong of China, was investigated using 16 polycyclic aromatic hydrocarbon (PAH) sediment samples of the bay. The total concentrations of 16 PAHs varied from 115 to 1 134 ng/g dry weight. The PAH composition pattern in sediments suggest dominance of 4-ring PAHs in Sites 2 and 4, and the ratio of certain related PAHs indicated important pyrolytic and petrogemc sources. The results enhance the understanding of current contamination levels and make a better assessment of likely impacts of organic contamination on ecosystems and the sustainability of local aquaculture in the area especially after the establishment of the Nuclear Power Station by the bay. 展开更多

Ten multipurpose tree species, Terminalia arjuna, Azadirechta indica, Prosopis juliflora, Pongamia pinnata, Casuarina equisetifolia, Prosopis alba, Acacia nilotica, Eucalyptus tereticornis, Pithecellobium dulce and Cassia siamea, were raised in a monoculture tree cropping system on the sodic soil of Gangetic alluvium in north India (26° 47° N: 80°46′ E) for 10 years to evaluate the biomass and bio-energy production. The soil was compact, sodic and impervious to water associated with nutrient deficiency or toxicity. Maximum plant height was recorded with E. tereticornis followed by C.equisetifolia and P. juliflora. A. nilotica performed better than the other species in terms of diameter at breast height (DBH) with a basal area of 13.04 m^2·ha^-1, followed by P. juliflora and C. equisetifolia. P. juliflora and A. nilotica produced nearly similar biomass of 56.50 and 50.75 Mg·ha^-1, respectively, at 10 years; whereas, A. indica, P. pinnata, C. siamea and P. alba did not perform well. P. juliflora scored maximum in net biomass production and nutrient demand. Nutrient (N, P, K, Ca, and Mg) concentrations were higher in leaf component of P. juliflora. However, in woody components, there was little variation between the species. N removal for production of one ton of wood was lowest in Acacia nilotica, P in T. arjuna, K in P. dulce and Ca and Mg in P. juliflora. P. juliflora gave the highest energy production of 1267.75 GJ.ha^-1 followed by A. nilotica with 1206 GJ.ha^-1 and the lowest ofA. indica (520.66 GJ.ha^-1). 展开更多

The effects of different carbon sources(sugars) on the production and molecular properties of exopolysaccharides(EPS) were evaluated in the mycelial liquid culture of a medicinal fungus Cordyceps sinensis Cs-HK1. Galactose or mannose was used(at 5 g·L^(-1)) as a secondary carbon source with glucose(35 g·L^(-1)) at the mass ratio of 1:7. Mannose was consumed notably since the first day of culture, but galactose was not even after glucose was exhausted.The volumetric yield of EPS in culture was increased slightly with the addition of galactose and decreased with mannose. The monosaccharide composition of EPS was also different, e.g., on day 8, the glucose contents of EPS were 76%with the addition of mannose, 59% with galactose, compared with 62% with glucose only. The molecular weight distribution of EPS was also affected by the secondary carbon source, being generally lower compared with that with glucose only. The results suggested that the addition of galactose improved the total yield of EPS in culture while mannose can improve the yield of glucan constituent of EPS. 展开更多

The GEF-WB Nutrient Reduction project aims to reduce the phosphorus inputs of the Black Sea from the Danube by means of different interventions. Hungary takes part of this project by enhancing the nutrient trapping capacity of the Gemenc and Beda-Karapancsa wetlands (GBK) which can be found on the premises of the Danube Drava National Park along the Danube River in Southern Hungary. Due to anthropogenic influences, less of small floods reach the oxbows of the area than some decades before which has a negative effect on the ecosystem. The objective of the study is to predict the nutrient reduction capacity of the Gemenc-B6da-Karapancsa water system and to make a proposal to increase this capacity. Although the floodplain vegetation has a huge nutrient intake capacity, this is limited by the fact that the floodplain is not always covered by the water of the river Danube. For that reason, a combination of hydrodynamic modelling, flood probability calculation and an estimation of suspended solids settling constitute the base of this study. The main course taken into account is the settling of suspended solids and the ratio of total nitrogen and total phosphorus to the mass of sediment is determined by local measurement results. 展开更多

Genetic diversity of three cultured populations of jade perch (Scortum bacoo) are studied using amplified fragment length polymorphism (AFLP) markers, which is Guangzhou(GZ) population, Foshan (FS) population and Qingdao(QD) population. Nine primer combinations were used and 385 fragments were detected. Among the 385 fragments, 80 bands (20.78%) were polymorphic. And it can be speculated that the genetic diversity of the three cultured populations of jade perch was very poor according to the gene genetic diversity among populations (Ht), the gene genetic diversity within populations (Hs) and Shannon-Weiner index (I). From gene differentiation (Gst), genetic distance (D), genetic similarity (5), and UPGMA analysis, it is found that the relationship among the three populations is very closed, and the difference in genetic diversity among the three populations is not significant. Further clustering relationships of the jade perch cultured populations also are correlated to historical-breeding observations and genetic relationships. The GZ population clustered together with the QD population first, then with FS population. 展开更多

Background The induced pluripotent stem cell (iPSC) has shown great potential in cellular therapy of myocardial infarction (MI), while its application is hampered by the low efficiency of cardiomyocyte differentiation. The present study was designed to investigate the effects of cardiotrophin-1 (CT-1) on cardiomyocyte differentiation from mouse induced pluripotent stem cells (miPSCs) and the underlying mechanisms involved. Methods The optimal treatment condition for cardiomyocyte differentiation from miPSCs was established with ideal concentration (10 ng/mL) and duration (from day 3 to day 14) of CT-1 administration. Up-regulated expression of cardiac specific genes that accounted for embryonic cardiogenesis was observed by quantitative RT-PCR. Elevated amount of a-myosin heavy chain (ct-MHC) and cardiac troponin I (cTn I) positive cells were detected by immunofluorescence staining and flow cytometry analysis in CT- 1 group. Results Transmission electron microscopic analysis revealed that cells treated with CT- 1 showed better organized sacromeric structure and more mitochondria, which are morphological characteristic of matured cardiomyocytes. Western blot demonstrated that CT-1 promotes cardiomyocyte differentiation from miPSCs partly via JAK2/STAT3/Pim-1 pathway as compared with control group. Conclusions These findings suggested that CT-1 could enhance the cardiomyocyte differentiation as well as the maturation of mouse induced pluripotent stem cell derived cardiomyocytes by regulating JAK2/STAT3/Pim-1 signaling pathway. 展开更多

Twenty-four halophilic archaeal strains were isolated from three Algerian hypersaline sites, including the solar salterns in Ichekaben and two natural salt lakes at Ouargla and Oran. Almost full-length 16S rRNA genes were amplified using archaeal-specific primers, sequenced and phylogenetically analyzed. Three isolates from the salt lake of Ouargla were affiliated to Halorubrum sp., while six isolates from Ichekaben were Natrinema sp.. The remaining strains belonged to the genus Haloarcula, which was identified in the three Algerian sites. A phylogenetic tree of Haloarcula sp. including the two copies of the 16S rRNA gene, showed that two strains group with Haloarcula hispanica, while the other 13 cluster with OHF-1/OHF-2 isolated from French commercial salt. Both Natrinema sp. and Haloarcula sp. were persistent at Ichekaben. For the six Natrinema sp. isolates, this observation correlates with their capacity to produce antimicrobial compounds, and a DNA fragment closely related to the halocin C8 gene was amplified for five strains. 展开更多

Considering the economic importance in traditional Chinese medicine(TCM) and lack of baseline information, we evaluated the proximate composition, fatty acid and amino acid profiles, trace element content and C:N ratio in the alligator pipefish, Syngnathoides biaculeatus. Amongst proximate principals, a crude protein formed the major biochemical component((58.9±2.2)% dry weight). Mean percent concentrations(dry weight) of other components such as a total lipid(TL), ash and nitrogen-free extract measured were,(1.8±0.2)%,(19.2±2.2)% and(20.1±0.45)%, respectively. The fatty acid profile revealed the presence of 27 saturated fatty acids(SFA) with 13 straight-chained and 14 branched-chained, 28 unsaturated fatty acids(UFA) with 14 monounsaturated and 14 polyunsaturated and nine other minor fatty acids. Mean percent contributions of total SFAs and UFAs to TL were found to be(55.41±0.24)% and(44.05±0.25)%, respectively. Altogether, 16 dif ferent amino acids with an equal number of essential(EAA) and non-essential(NAA) ones were identified. Percent contributions by EAA and NAA to the total amino acid content were 38.11% and 61.89%, respectively. Trace metal concentrations in S. biaculeatus were generally low and their distribution followed the order, Mg>Fe>Zn>Mn>Cu>Cr>Ni>Hg>Co. The C:N ratio was(4.37±0.04)%. The profile of major biochemical constituents in alligator pipefish, S. biaculeatus revealed its potential use in TCM as well as a nutritional diet for human consumption. The results of the study would also form the basis for formulation and optimization of diets for the culture of S. biaculeatus. 展开更多

Polyphenols are one of the most important secondary metabolites, and affect the decomposition of litter and soil organic matter. This study aims to monitor the mass loss rate of tea leaf litter and nutrient release pattern, and investigate the role of tea polyphenols played in this process. High-performance liquid chromatography(HPLC) and classical litter bag method were used to simulate the decomposition process of tea leaf litter and track the changes occurring in major polyphenols over eight months. The release patterns of nitrogen, potassium, calcium, and magnesium were also determined. The decomposition pattern of tea leaf litter could be described by a two-phase decomposition model, and the polyphenol/N ratio effectively regulated the degradation process. Most of the catechins decreased dramatically within two months; gallic acid(GA), catechin gallate(CG), and gallocatechin(GC) were faintly detected, while others were outside the detection limits by the end of the experiment. These results demonstrated that tea polyphenols transformed quickly and catechins had an effect on the individual conversion rate. The nutrient release pattern was different from other plants which might be due to the existence of tea polyphenols. 展开更多

As an ecosystem engineer cushion plant, its ability to facilitate the restoration of degraded alpine grassland was studied in a degraded alpine grassland at an elevation of 4500 m on the southern slope of the Nyainqentanglha Mountains in Damxung. The species diversity, soil nutrients and water content underneath and outside the cushion plant A. tapete were investigated. The results showed that soil nutrients underneath the A. tapete cushion were significantly increased by about 16%-48% compared to outside the cushion, of which the organic matter and total N were increased by 16.2% and 18.9% respectively, and the soil water content was increased about 12%. The index of species diversity of richness(S), Shannon-Wiener’s H and Simpson’s D all increased with the coverage of cushion plant A. tapete. Our results suggested that this cushion plant can facilitate restoration of the degraded alpine grassland by modifying the local soil environment and increasing the community diversity, so it should be conserved for the restoration of degraded alpine grasslands on the Qinghai-Tibetan Plateau. 展开更多

A multi-scale approach is essential to assess the factors that limit avian habitat use. Numerous studies have examined habitat use by the red-crowned crane, but integrated multi-scale habitat use information is lacking. We evaluated the effects of several habitat variables quantified across many spatial scales on crane use and abundance in two periods (2000 and 2009) at Yancheng National Nature Reserve, China. The natural wetlands decreased in area by 30,601 ha (-6.9%) from 2000 to 2009, pre- dominantly as a result of conversion to aquaculture ponds and farmland, and the remaining was under degradation due to expan- sion of the exotic smooth cordgrass. The cranes are focusing in on either larger patches or those that are in close proximity to each other in both years, but occupied patches had smaller size, less proximity and more regular boundaries in 2009. At landscape scales, the area percentage of common seepweed, reed ponds and paddy fields had a greater positive impact on crane presence than the area percentage of aquaculture ponds. The cranes were more abundant in patches that had a greater percent area of common seepweed and reed ponds, while the percent area of paddy fields was inversely related to crane abundance in 2009 due to changing agricultural practices. In 2009, cranes tended to use less fragmented plots in natural wetlands and more fragmented plots in anthropogenic paddy fields, which were largely associated with the huge loss and degradation of natural habitats between the two years. Management should focus on restoration of large patches of natural wetlands, and formation of a relatively stable area of large paddy field and reed pond to mitigate the loss of natural wetlands [Current Zoology 59 (5): 604-617, 2013]. 展开更多

We studied the uptake of ammonium, nitrate, and a variety of amino acids by alpine plant species in the Kobresia humil alpine meadow ecosystem in situ. We examined the extent of niche separation in uptake of N source by different plant species in alpine communities, and investigated the contribution of symbiotically fixed N to the total N in alpine meadow. The results are (1) δ15N natural abundance values of 13 plant species lie between -2.680‰ and 5.169‰, and the scope is 7.849‰. (2) Le- guminous plants, such as Trigonella ruthenica, Gueldenstaedtia diversiffolia, and Oxytyopis ochrocephala, and non-legumi- nous plant Gentiana straminea uptake low amounts of 15N labeled ammonium, nitrate, glycine or aspartate in soil. (3) As far as the plant uptake of organic N is concerned, Kobresia humilis, Poa pratensis, and Gentiuna spathutifolta can effectively uptake organic nitrogen, and about 37%-40% of the nitrogen of these species comes from soil organic nitrogen sources (such as glycine and aspartate). Stipa aliena can effectively uptake nitrate, and 60% of its nitrogen comes from soil nitrate. Potentilla anserina, Poa pratensis, and Thalictrum alpinum can effectively absorb ammonium in comparason to other plant species in the meadow, and about 25%-27% of the nitrogen in these plants comes from soil ammonium. (4) The contribution of leguminous fixed N to total N is 7.48%-9.26% in Kobresia humilis alpine meadow. (5) These data show many plant species of alpine meadow may effectively utilize dissolved organic nitrogen such as amino acids, and these plants have diverse ways to uptake soil nitrogen in alpine meadows. Based on the results we can partly explain why there are abundant biodiversities and how plants at alpine habitat utilize the limited soil N sources. 展开更多

The goal is to explore the properties of a novel model which can describe the characteristics of multistage for the population growth of microorganisms in nonlinear switch dynamic system. The improved model is developed based on the experimental data to describe the delayed, developmental and stationary stages well for the phases of batch culture. Then the existence, uniqueness and boundedness of solutions to the nonlinear multistage switch system and the Lipschitz continuity and differentiability of solutions with respect to the initial state is discussed as well.
