[Method] Hep G2 cells were treated with GPS for 48 h for morphology assay by transition electron microscope. Anti-proliferative effects with the same treatment for 24 hand 48 h were assayed by MTT method.Cell cycle distribution and apoptosis assay of treated cells were performed in flow cytometry. [Result] The results showed that GPS enhanced growth inhibitory effect on Hep G2 cells in a time-and dose-dependent manner. PI(Propidium iodide)/Annexin V staining analyzed by FCM(flow cytometry) demonstrated that GPS has a cytotoxic effect on tumor cells. Cell cycle arrest of Hep G2 treated with GPS occurred in G2 phase. [Conclusion] This study suggests that GPS could exert an antitumor effect and could be used as a therapeutic agent for live cancer.展开更多

[Method] The stem apex growing points of Pteroceltis tatarinowii Maxim. were treated with different concentrations of colchicine solution for different hours to figure out a proper method and obtain poly- ploids. [Result] The most effective induction was obtained by treatment with 0.6%- 0.8% colchicine for 72 h with 34.2% mutation rate. Flow cytometry and chromosome observation of the stem apex growing point of P. tatarinowii Maxim. proved that the tetraploid plants were successfully obtained with chromosome number 2n=4x=36. [Conclusion] The result not only fills the blank of polyploid breeding of P. tatarinowii, but also provides an effective way to broaden the methods of cultivation of fast- growing, high-quality, disease-resilience, new varieties of Pteroceltis.展开更多

The diagnosis is based on the presence of both typical symptoms and pathologic findings on esophageal biopsy. Patients usually present with dysphagia, food impaction and/or reflux-like symptoms, and biopsy of the esophagus shows more than 15 eosinophils per high-power fi eld. In addition, it is essential to exclude the presence of known causes of tissue eosinophilia such as gastroesophageal reflux disease, infections, malignancy, collagen vascular diseases, hypersensitivity, and inflammatory bowel disease. There are no standardized protocols for the therapy of eosinophilic esophagitis. A variety of therapeutic approaches including acid suppression, dietary modifications, topical corticosteroids and endoscopic dilation can be used alone or in combination.展开更多
Flow cytometry(FCM)is a powerful technique for single-bacteria analysis via simultaneous light-scattering and fluorescence measurements.By offering high-throughput,quantitative,and multiparameter analysis at the singl...Flow cytometry(FCM)is a powerful technique for single-bacteria analysis via simultaneous light-scattering and fluorescence measurements.By offering high-throughput,quantitative,and multiparameter analysis at the single-cell level,FCM has gained an increased popularity in microbiological research,food safety monitoring,water quality control,and clinical diagnosis.Here we will review the recent applications of flow cytometry in areas such as(1)total bacterial cell count,(2)bacterial viability analysis,(3)specific bacterial detection and identification,(4)characterization of physiological changes under environmental perturbations,and(5)biological function studies.Nevertheless,despite these widespread applications,challenges still remain for the detection of small sizes of bacteria and biochemical features that cannot be brightly stained via fluorescence.Recent improvement in FCM instrumentation will be discussed,and particularly the development of high sensitivity flow cytometry for advanced analysis of single bacterial cells will be highlighted.展开更多
