B...To address to what extent hypertrophy and hyperplasia contribute to liver mass restoration after major tissue loss.METHODSThe ability of the liver to regenerate is remarkable on both clinical and biological grounds. Basic mechanisms underlying this process have been intensively investigated. However, it is still debated to what extent hypertrophy and hyperplasia contribute to liver mass restoration after major tissue loss. We addressed this issue using a genetically tagged system. We were able to follow the fate of single transplanted hepatocytes during the regenerative response elicited by 2/3 partial surgical hepatectomy (PH) in rats. Clusters of transplanted cells were 3D reconstructed and their size distribution was evaluated over time after PH.RESULTSLiver size and liver DNA content were largely recovered 10 d post-PH, as expected (e.g., total DNA/liver/100 g b.w. was 6.37 ± 0.21 before PH and returned to 6.10 ± 0.36 10 d after PH). Data indicated that about 2/3 of the original residual hepatocytes entered S-phase in response to PH. Analysis of cluster size distribution at 24, 48, 96 h and 10 d after PH revealed that about half of the remnant hepatocytes completed at least 2 cell cycles. Average size of hepatocytes increased at 24 h (248.50 μm<sup>2</sup> ± 7.82 μm<sup>2</sup>, P = 0.0015), but returned to control values throughout the regenerative process (up to 10 d post-PH, 197.9 μm<sup>2</sup> ± 6.44 μm<sup>2</sup>, P = 0.11). A sizeable fraction of the remnant hepatocyte population does not participate actively in tissue mass restoration.CONCLUSIONHyperplasia stands as the major mechanism contributing to liver mass restoration after PH, with hypertrophy playing a transient role in the process.展开更多

Contrary to the healthy parental involvement, a family can also have negative effects on an athlete development. The purpose of this study was to determine gender, father involvement, and mother involvement differences in the psychological profiles of collegiate freshmen athletes as measured by perfectionism, physical self-concept, and psychological well-being. Methods: Eighty-two male and 73 female collegiate freshmen responded to self-reported measures of family involvement, perfectionism, physical self-concept, and psychological well-being. Three separate 2 x 2 x 2 (gender x mother involvement x father involvement) factorial multivariate analysis of variance (MANOVAs) were carried out on the respective subscales from the reported measures. Results: There were significant differences for gender as well as father involvement in perfectionism (p 〈 0.01). Freshmen females had lower concern over mistakes, and greater levels of organization as well as planfulness than males. Furthermore, freshmen athletes with high involved fathers showed greater levels of high standards for others as well as organization. In present study, a well-described in vitro sust...Heart remodeling is associated with the loss of cardiomyocytes and increase of fibrous tissue owing to abnormal mechanical load in a number of heart disease conditions. In present study, a well-described in vitro sustained stretch model was employed to study mechanical stretch-induced responses in both neonatal cardiomyocytes and cardiac fibroblasts. Cardiomyocytes, but not cardiac fibroblasts, underwent mitochondria-dependent apoptosis as evidenced by cytochrome c (cyto c) and Smac/DIABLO release from mitochondria into cytosol accompanied by mitochondrial membrane potential (△ψ_m) reduction, indicative of mitochondrial permeability transition pore (PTP) opening. Cyclosporin A, an inhibitor of PTP, inhibited stretch-induced cyto c release, △ψ_m reduction and apoptosis, suggesting an important role of mitochondrial PTP in stretch-induced apoptosis. The stretch also resulted in increased expression of the pro-apoptotic Bcl-2 family proteins, including Bax and Bad, in cardiomyocytes, but not in fibroblasts. Bax was accumulated in mitochondria following stretch. Cell permeable Bid-BH3 peptide could induce and facilitate stretch-induced apoptosis and △ψ_m reduction in cardiomyocytes. These results suggest that Bcl-2 family proteins play an important role in coupling stretch signaling to mitochondrial death machinery, probably by targeting to PTP. Interestingly, the levels of p53 were increased at 12 h after stretch although we observed that Bax upregulation and apoptosis occurred as early as 1 h. Adenovirus delivered dominant negative p53 blocked Bax upregulation in cardiomyocytes but showed partial effect on preventing stretch-induced apoptosis, suggesting that p53 was only partially involved in mediating stretch-induced apoptosis. Furthermore, we showed that p21 was upregulated and cyclin B1 was downregulated only in cardiac fibroblasts, which may be associated with G_2/M accumulation in response to mechanical stretch.展开更多

Methods: Cell groups with cloning capability were obtained from midbrain of new-born rats by using serum-free and floating cell culture. Immunocytochemistry was applied to detect antigens such as BrdU, Nestin and specific markers for mature neural cells. Results: Cell groups isolated from midbrain propagated in succession and shaped 'neuro-spheres'. Nestin antigen was expressed in the course of proliferation. After differentiation, epitopes of neuron , astrocyte and oligodendrocyte were all found in the neural progenitors-derived cells, the most majority were astrocyte-epitoped cells. Conclusion: Cell groups expressing Nestin isolated from midbrain of new-born rats have properties of propagating, multi-potency differentiating and self-renewal. It is demonstrated that the cell groups are stem-like neural progenitors in the central nervous system, which are qualified for cell therapy of diseases in central nervous system.展开更多

METHODS: Neonatal Sprague Dawley rats were intra- peritoneally injected with 70 mg/kg STZ. Body weight, pancreas weight and blood glucose were recorded every two days after the treatment. To identify the expression and location of transcription factors in the rat pancreas, double immunofluorescent staining was performed using antibodies to specific cell markers and transcription factors. RESULTS: Expression of Neurogenin 3 (Ngn3), a marker for endocrine precursor cells, was observed by immunofluorescence in a few β-cells and many a-cells. The expression reached a peak 12 d after treatment. Pax4, a transcription factor that lies downstream of Ngn3 and plays an important role in β-cell differentiation, was also expressed in the α-cells of STZ-treated rats. We did not observe significant changes in Nkx6.1, which is essential for β-cell maturation in the treated rats. CONCLUSION: α-cells dedifferentiated into endocrine precursor cells and acquired the ability to dedifferentiate in the neonatal rat pancreas after STZ treatment.展开更多

This paper first discussed issues and uses of literature in L2 contexts and addressed the current empirical inquiry into L2 haiku writing as a method for L2 learning. The study was designed as a qualitative research which investigated the participants' reflection on writing L2 haikus for six weeks in an EFL college writing course. The data obtained was analyzed by using the coding system and showed that writing haiku in the target language was a challenging but valuable task for L2 learning among the EFL students. AIM:To study the effects of live and dead Lactobacillus rhamnosus GG(GG) on rotavirus infection in a neonatal rat model.METHODS:At the age of 2 d,suckling Lewis rat pups were supplemented with either live or dead GG a...AIM:To study the effects of live and dead Lactobacillus rhamnosus GG(GG) on rotavirus infection in a neonatal rat model.METHODS:At the age of 2 d,suckling Lewis rat pups were supplemented with either live or dead GG and the treatment was continued daily throughout the experi-ment.At the age of 5 and 6 d the pups received oral rotavirus(RV) SA-11 strain.The pups were sacrificed at the age of 7 or 8 d by decapitation.The gastrointestinal tract was removed and macroscopic observations were done.The consistency of feces in the colon was classified using a four-tier system.RV was detected from the plasma,small intestine,colon and feces by real-time quantitative polymerase chain reaction(PCR).RESULTS:In this neonatal rat model,RV induced a mild-to-moderate diarrhea in all except one pup of the RV-inoculated rats.RV moderately reduced body weight development from day 6 onwards.On day 7,after 2 d of RV infection,live and dead GG groups gained significantly more weight than the RV group without probiotics [36%(P = 0.001) and 28%(P = 0.031),respectively].In addition,when compared with the RV control group,both live and dead GG reduced the weight ratio of colon/animal body weight to the same level as in the healthy control group,with reductions of 22%(P = 0.002) and 28%(P < 0.001),respectively.Diarrhea increased moderately in both GG groups.However,the diarrhea incidence and severity in the GG groups were not statistically significantly different as compared with the RV control group.Moreover,observed diarrhea did not provoke weight loss or death.The RV control group had the largest amount of RV PCR-positive samples among the RV-infected groups,and the live GG group had the smallest amount.Rats receiving live GG had significantly less RV in the colon(P = 0.027) when compared with the RV control group.Live GG was also more effective over dead GG in reducing the quantity of RV from plasma(P = 0.047).CONCLUSION:Both live and dead GG have beneficial effects in RV infection.GG may increase RV clearance from the body and reduce colon swelling.展开更多
The North Yellow Sea Basin ( NYSB ), which was developed on the basement of North China (Huabei) continental block, is a typical continental Mesozoic-Cenozoic sedimentary basin in the sea area. Its Mesozoic basin is a...The North Yellow Sea Basin ( NYSB ), which was developed on the basement of North China (Huabei) continental block, is a typical continental Mesozoic-Cenozoic sedimentary basin in the sea area. Its Mesozoic basin is a residual basin, below which there is probably a larger Paleozoic sedimentary basin. The North Yellow Sea Basin comprises four sags and three uplifts. Of them, the eastern sag is a Mesozoic-Cenozoic sedimentary sag in NYSB and has the biggest sediment thickness; the current Korean drilling wells are concentrated in the eastern sag. This sag is comparatively rich in oil and gas resources and thus has a relatively good petroleum prospect in the sea. The central sag has also accommodated thick Mesozoic-Cenozoic sediments. The latest research results show that there are three series of hydrocarbon source rocks in the North Yellow Sea Basin, namely, black shales of the Paleogene, Jurassic and Cretaceous. The principal hydrocarbon source rocks in NYSB are the Mesozoic black shale. According to the drilling data of Korea, the black shales of the Paleogene, Jurassic and Cretaceous have all come up to the standards of good and mature source rocks. The NYSB owns an intact system of oil generation, reservoir and capping rocks that can help hydrocarbon to form in the basin and thus it has the great potential of oil and gas. The vertical distribution of the hydrocarbon resources is mainly considered to be in the Cretaceous and then in the Jurassic.展开更多
With the evolutionary change in information and communication technologies (ICT), the definition of literacy has undergone substantial change to include the skills and strategies required to successfully adapt to th...With the evolutionary change in information and communication technologies (ICT), the definition of literacy has undergone substantial change to include the skills and strategies required to successfully adapt to the rapidly changing ICT contexts. This new definition presents an urgent need to acknowledge the gap between the types of reading experiences students come across at school and those they regularly practice outside the school. One way to bridge such gap is to expand the reading instruction to incorporate electronic reading device, like Kindle, to engage students in electronic reading. Many e-books feature multimodal functionalities, including audio, video, hyperlinks, and interactive tools. Research on this form of reading is still in its infancy in spite of some positive prospects for fostering literacy development and reading comprehension. Therefore, adopting a mixed method approach with 36 college freshmen in central Taiwan, the current study intended to fill in the void by investigating any difference in readers' reading comprehension and reading motivation, between the Kindle group and the print-based group. The results indicate that embedding aspects of technology into a traditional reading instruction boosted the participants' motivation to read. Nevertheless, the novelty-driven enthusiasm for electronic reading might not sustain students' reading motivation long enough to cultivate long-lasting reading habits. The results indicate that there was no significant difference in reading comprehension between two groups. Implications for utilizing electronic readers in EFL classes are also provided.展开更多

The schema embodying the learners background knowledge of cultural familiar materials facilitates the understanding of the text (Pritchard, 1990). Also Steffensen, Joag-Dev, and Anderson (1979) and Nelson (1987) proposed that the schemata embodying background knowledge influenced how well the text would be comprehended. However, Hudson (1982) and Carrell (1984) found there was no significant background effect in advanced level learners. From those studies, it seems that background effect is different at different language proficiency levels. Thus, the present study examines the interaction between background knowledge and language proficiency in reading comprehension. The participants were freshmen of National Chiayi University. They were divided into three language proficiency levels based on the General English Proficiency Test. Four reading comprehension tests were constructed to assess their reading comprehension: cultural familiar/unfamiliar text and topic familiar/unfamiliar text. The results of this study showed that participants had better performance on the culture/topic familiar text than the culture/topic unfamiliar text. Thus the findings suggest that the teacher can use teaching activities, such as pre-reading activities or vocabulary teaching to increase the background knowledge when teaching readings to EFL (English as a foreign language) learners展开更多

This study sought to examine mental health development measured with University Personality Inventory (UPI) among Chinese college students and its relationship with family factors (family income, family residence, and sibling status~) with a longitudinal design. Data of 390 Chinese college students were collected at their first and the third year of college period. The results showed that Chinese college students' overall mental health declined in the third year than in the first year. Urban students showed a more rapid increase in physical symptoms from the first year to the third year than rural students. The results indicated that psychological intervention should be extended to students facing graduation, especially those from urban area.展开更多

Objective To investigate the effect of graded hypothermia on neuropathologic alterations of neonatal rat brain after exposed to hypoxic-ischemic insult at 37℃, 33℃, 31℃, and 28℃, respectively, and to observe the e...Objective To investigate the effect of graded hypothermia on neuropathologic alterations of neonatal rat brain after exposed to hypoxic-ischemic insult at 37℃, 33℃, 31℃, and 28℃, respectively, and to observe the effect of hypothermia on 72-kDa heat shock protein (HSP72) expression after hypoxic-ischemic insult. Methods Seven days old Wistar rats were subjected to unilateral common carotid artery ligation followed by exposure to hypoxia in 8% oxygen for 2 hours at 37℃, 33℃, 31℃, and 28℃, respectively. The brain temperature was monitored indirectly by inserting a mini-thermocouple probe into the temporal muscle during hypoxia. After hypoxia-ischemia their mortality was assessed. Neuronal damage was assessed with HE staining 72 hours after hypoxia. HSP72 expression at 0.5, 24, and 72 hours of recovery was immunohistochemically assessed using a monoclonal antibody to HSP72. Results Hypoxia-ischemia caused 10.5% (2 / 19) of mortality in rat of 37℃ group, but no death oc- curred in 33℃, 31℃ or 28℃ groups. HE staining showed neuropathologic damage was extensive in rats exposed to hypoxia-ischemia at 37℃ (more than 80.0%). The incidence of severe brain damage was significantly decreased in 33℃ (53.3%) and 31℃ groups (44,4%), and no histologic injury was seen in the 28℃ group of rats. Expression of HSP72 was manifest and persistent in the rat brain of 37℃ group, but minimum in the rat brain of 28℃ group. Conclusion Mild and moderate hypothermia might prevent cerebral visible neuropathologic damage associated with hypoxic-ischemic injury by decreasing stress response.展开更多

Cyclosporin A, an inhibitor of PTP, inhibited stretch-induced cyto c release, △ψ_m reduction and apoptosis, suggesting an important role of mitochondrial PTP in stretch-induced apoptosis. The stretch also resulted in increased expression of the pro-apoptotic Bcl-2 family proteins, including Bax and Bad, in cardiomyocytes, but not in fibroblasts. Bax was accumulated in mitochondria following stretch. Cell permeable Bid-BH3 peptide could induce and facilitate stretch-induced apoptosis and △ψ_m reduction in cardiomyocytes. These results suggest that Bcl-2 family proteins play an important role in coupling stretch signaling to mitochondrial death machinery, probably by targeting to PTP. Interestingly, the levels of p53 were increased at 12 h after stretch although we observed that Bax upregulation and apoptosis occurred as early as 1 h. Adenovirus delivered dominant negative p53 blocked Bax upregulation in cardiomyocytes but showed partial effect on preventing stretch-induced apoptosis, suggesting that p53 was only partially involved in mediating stretch-induced apoptosis. Furthermore, we showed that p21 was upregulated and cyclin B1 was downregulated only in cardiac fibroblasts, which may be associated with G_2/M accumulation in response to mechanical stretch.

文摘Objective: To isolate neural progenitors from midbrain of new born rats. Methods: Cell groups with cloning capability were obtained from midbrain of new-born rats by using serum-free and floating cell culture. Immunocytochemistry was applied to detect antigens such as BrdU, Nestin and specific markers for mature neural cells. Results: Cell groups isolated from midbrain propagated in succession and shaped 'neuro-spheres'. Nestin antigen was expressed in the course of proliferation. After differentiation, epitopes of neuron , astrocyte and oligodendrocyte were all found in the neural progenitors-derived cells, the most majority were astrocyte-epitoped cells. Conclusion: Cell groups expressing Nestin isolated from midbrain of new-born rats have properties of propagating, multi-potency differentiating and self-renewal. It is demonstrated that the cell groups are stem-like neural progenitors in the central nervous system, which are qualified for cell therapy of diseases in central nervous system.

文摘AIM: To investigate the mechanism behind β-cell regeneration in neonatal rat pancreas treated with strep- tozotocin (STZ). METHODS: Neonatal Sprague Dawley rats were intra- peritoneally injected with 70 mg/kg STZ. Body weight, pancreas weight and blood glucose were recorded every two days after the treatment. To identify the expression and location of transcription factors in the rat pancreas, double immunofluorescent staining was performed using antibodies to specific cell markers and transcription factors. RESULTS: Expression of Neurogenin 3 (Ngn3), a marker for endocrine precursor cells, was observed by immunofluorescence in a few β-cells and many a-cells. The expression reached a peak 12 d after treatment. Pax4, a transcription factor that lies downstream of Ngn3 and plays an important role in β-cell differentiation, was also expressed in the α-cells of STZ-treated rats. We did not observe significant changes in Nkx6.1, which is essential for β-cell maturation in the treated rats. CONCLUSION: α-cells dedifferentiated into endocrine precursor cells and acquired the ability to dedifferentiate in the neonatal rat pancreas after STZ treatment.
文摘The aim of the current study is to discuss the value of poetry writing in an L2 (second language) by investigating perceptions, attitudes, and emotions of 20 EFL (English as a Foreign Language) college freshmen regarding writing Japanese poetry, haiku in English. This paper first discussed issues and uses of literature in L2 contexts and addressed the current empirical inquiry into L2 haiku writing as a method for L2 learning. The study was designed as a qualitative research which investigated the participants' reflection on writing L2 haikus for six weeks in an EFL college writing course. The data obtained was analyzed by using the coding system and showed that writing haiku in the target language was a challenging but valuable task for L2 learning among the EFL students. Especially haiku composition allowed them to develop their L2 linguistic awareness

文摘AIM:To study the effects of live and dead Lactobacillus rhamnosus GG(GG) on rotavirus infection in a neonatal rat model.METHODS:At the age of 2 d,suckling Lewis rat pups were supplemented with either live or dead GG and the treatment was continued daily throughout the experi-ment.At the age of 5 and 6 d the pups received oral rotavirus(RV) SA-11 strain.The pups were sacrificed at the age of 7 or 8 d by decapitation.The gastrointestinal tract was removed and macroscopic observations were done.The consistency of feces in the colon was classified using a four-tier system.RV was detected from the plasma,small intestine,colon and feces by real-time quantitative polymerase chain reaction(PCR).RESULTS:In this neonatal rat model,RV induced a mild-to-moderate diarrhea in all except one pup of the RV-inoculated rats.RV moderately reduced body weight development from day 6 onwards.On day 7,after 2 d of RV infection,live and dead GG groups gained significantly more weight than the RV group without probiotics [36%(P = 0.001) and 28%(P = 0.031),respectively].In addition,when compared with the RV control group,both live and dead GG reduced the weight ratio of colon/animal body weight to the same level as in the healthy control group,with reductions of 22%(P = 0.002) and 28%(P < 0.001),respectively.Diarrhea increased moderately in both GG groups.However,the diarrhea incidence and severity in the GG groups were not statistically significantly different as compared with the RV control group.Moreover,observed diarrhea did not provoke weight loss or death.The RV control group had the largest amount of RV PCR-positive samples among the RV-infected groups,and the live GG group had the smallest amount.Rats receiving live GG had significantly less RV in the colon(P = 0.027) when compared with the RV control group.Live GG was also more effective over dead GG in reducing the quantity of RV from plasma(P = 0.047).CONCLUSION:Both live and dead GG have beneficial effects in RV infection.GG may increase RV clearance from the body and reduce colon swelling.
文摘The North Yellow Sea Basin ( NYSB ), which was developed on the basement of North China (Huabei) continental block, is a typical continental Mesozoic-Cenozoic sedimentary basin in the sea area. Its Mesozoic basin is a residual basin, below which there is probably a larger Paleozoic sedimentary basin. The North Yellow Sea Basin comprises four sags and three uplifts. Of them, the eastern sag is a Mesozoic-Cenozoic sedimentary sag in NYSB and has the biggest sediment thickness; the current Korean drilling wells are concentrated in the eastern sag. This sag is comparatively rich in oil and gas resources and thus has a relatively good petroleum prospect in the sea. The central sag has also accommodated thick Mesozoic-Cenozoic sediments. The latest research results show that there are three series of hydrocarbon source rocks in the North Yellow Sea Basin, namely, black shales of the Paleogene, Jurassic and Cretaceous. The principal hydrocarbon source rocks in NYSB are the Mesozoic black shale. According to the drilling data of Korea, the black shales of the Paleogene, Jurassic and Cretaceous have all come up to the standards of good and mature source rocks. The NYSB owns an intact system of oil generation, reservoir and capping rocks that can help hydrocarbon to form in the basin and thus it has the great potential of oil and gas. The vertical distribution of the hydrocarbon resources is mainly considered to be in the Cretaceous and then in the Jurassic.
文摘With the evolutionary change in information and communication technologies (ICT), the definition of literacy has undergone substantial change to include the skills and strategies required to successfully adapt to the rapidly changing ICT contexts. This new definition presents an urgent need to acknowledge the gap between the types of reading experiences students come across at school and those they regularly practice outside the school. One way to bridge such gap is to expand the reading instruction to incorporate electronic reading device, like Kindle, to engage students in electronic reading. Many e-books feature multimodal functionalities, including audio, video, hyperlinks, and interactive tools. Research on this form of reading is still in its infancy in spite of some positive prospects for fostering literacy development and reading comprehension. Therefore, adopting a mixed method approach with 36 college freshmen in central Taiwan, the current study intended to fill in the void by investigating any difference in readers' reading comprehension and reading motivation, between the Kindle group and the print-based group. The results indicate that embedding aspects of technology into a traditional reading instruction boosted the participants' motivation to read. Nevertheless, the novelty-driven enthusiasm for electronic reading might not sustain students' reading motivation long enough to cultivate long-lasting reading habits. The results indicate that there was no significant difference in reading comprehension between two groups. Implications for utilizing electronic readers in EFL classes are also provided.
文摘Schema is known to play an important role in reading comprehension. The schema embodying the learners background knowledge of cultural familiar materials facilitates the understanding of the text (Pritchard, 1990). Also Steffensen, Joag-Dev, and Anderson (1979) and Nelson (1987) proposed that the schemata embodying background knowledge influenced how well the text would be comprehended. However, Hudson (1982) and Carrell (1984) found there was no significant background effect in advanced level learners. From those studies, it seems that background effect is different at different language proficiency levels. Thus, the present study examines the interaction between background knowledge and language proficiency in reading comprehension. The participants were freshmen of National Chiayi University. They were divided into three language proficiency levels based on the General English Proficiency Test. Four reading comprehension tests were constructed to assess their reading comprehension: cultural familiar/unfamiliar text and topic familiar/unfamiliar text. The results of this study showed that participants had better performance on the culture/topic familiar text than the culture/topic unfamiliar text. Thus the findings suggest that the teacher can use teaching activities, such as pre-reading activities or vocabulary teaching to increase the background knowledge when teaching readings to EFL (English as a foreign language) learners
文摘Autonomous learning has been attracting more and more attention in the field of second language teaching and learning since it was put forward. In order to get a better understanding about autonomous learning competence of freshmen of English major in university, this investigation was conducted in the form of questionnaire and was analyzed according to the data collected. The investigation found that autonomous learning competence of freshmen is poor and worrying. Freshmen have strong motivation for English learning, but they keep old learning habit and more rely on teachers. They are incapable of employing metacognitive strategies in their language learning and are not good at utilizing related learning resources available to them. All these deficiencies hinder improvement of them. So they are in great need of fostering the competence of automous learning.

文摘First year of college was found to be the most challenging and stressful year due to the transition from home to school and the adjustment to new life, however, others reported that pre-graduates suffered from more mental problems under the pressure of job-hunting and uncertainty of future. This study sought to examine mental health development measured with University Personality Inventory (UPI) among Chinese college students and its relationship with family factors (family income, family residence, and sibling status~) with a longitudinal design. Data of 390 Chinese college students were collected at their first and the third year of college period. The results showed that Chinese college students' overall mental health declined in the third year than in the first year. Urban students showed a more rapid increase in physical symptoms from the first year to the third year than rural students. The results indicated that psychological intervention should be extended to students facing graduation, especially those from urban area.

文摘Objective To investigate the effect of graded hypothermia on neuropathologic alterations of neonatal rat brain after exposed to hypoxic-ischemic insult at 37℃, 33℃, 31℃, and 28℃, respectively, and to observe the effect of hypothermia on 72-kDa heat shock protein (HSP72) expression after hypoxic-ischemic insult. Methods Seven days old Wistar rats were subjected to unilateral common carotid artery ligation followed by exposure to hypoxia in 8% oxygen for 2 hours at 37℃, 33℃, 31℃, and 28℃, respectively. The brain temperature was monitored indirectly by inserting a mini-thermocouple probe into the temporal muscle during hypoxia. After hypoxia-ischemia their mortality was assessed. Neuronal damage was assessed with HE staining 72 hours after hypoxia. HSP72 expression at 0.5, 24, and 72 hours of recovery was immunohistochemically assessed using a monoclonal antibody to HSP72. Results Hypoxia-ischemia caused 10.5% (2 / 19) of mortality in rat of 37℃ group, but no death oc- curred in 33℃, 31℃ or 28℃ groups. HE staining showed neuropathologic damage was extensive in rats exposed to hypoxia-ischemia at 37℃ (more than 80.0%). The incidence of severe brain damage was significantly decreased in 33℃ (53.3%) and 31℃ groups (44,4%), and no histologic injury was seen in the 28℃ group of rats. Expression of HSP72 was manifest and persistent in the rat brain of 37℃ group, but minimum in the rat brain of 28℃ group. Conclusion Mild and moderate hypothermia might prevent cerebral visible neuropathologic damage associated with hypoxic-ischemic injury by decreasing stress response.
文摘Carbon monoxide is thought to serve as a new endogenous mediator in the pathogenesis of sepsis andseptic shock. In newborn rat endotoxicosis, carbon monoxide levels in the circulation as well as liver, kid-ney and lung were found to be significantly increased (P<0. 05). Moreover, the elevations of carbonmonoxide correlated with enhanced nitric oxide production as indicated by nitrite/nitrate levels (P<0. 05).Our present data showed for the first time that endogenously produced carbon monoxide was increasedduring the course of shock-like states, which suggested that the role of carbon monoxide in sepsis and sep-tic shock might worth further study.
