Methods Ethanol extracts of the two culture strains ofAlexandrium minutum Halim were fractionated by gel filtration and ion exchange chromatography toobtain gonyautoxins. The toxin profiles were tested by HPLC method. Results Both strains containgonyautoxin-1, gonyautoxin-2, gonyautoxin-3 and gonyautoxin-4. The total gonyautoxin content ofAmtk4 strain is almost 10 times higher than that of Amtk2 strain. Conclusion Alexandrium minutumHalim Amtk4 strain is suitable for the preparation of Gonyautoxins.展开更多

Both the natural fragmentation and heavy cutting of mature forests have resulted in patchy grouse habitats. We used SPOT (XS-sensor) satellite imagery to discriminate between open land and conifer or broadleaf forests. The area analyzed is about 120000 ha in size and includes the Lianhuashan Nature Reserve and the Yeliguan Forestry Park. We identiifed 4111 ha of mature coniferous forests in 229 patches (maximum 332 ha, mean 18 ha) as the habitat used by Chinese Grouse throughout the year. We examined 31 forest islands of different sizes and degrees of isolation for the presence of Chinese Grouse. We used generalized linear models (GLM) with binomial error structure and logit link function to estimate the probability of Chinese Grouse occupancy in a forest fragment. Habitat patch size (hs) and distance to the next occupied fragment (doc) were used as predictor variables, important for occupancy. Small habitat islands were disproportionately less likely to be occupied than large, nearby habitats. There was a clear speciifc habitat size of about 40 ha, above which habitat fragments were occupied more often. Suitable habitat fragments isolated by more than 2 km appeared to be inaccessible to Chinese Grouse. The results have been used in reforestation projects to establish linking corridors in the study area.展开更多

This species consists of many strains that differ in their ability to produce toxins but have similar morphology, making identification difficult. In this study, species-specific rRNA probes were designed for whole-cell fluorescence in situ hybridization (FISH) to distinguish A. minutum from two phylogenetic clades. We acquired the complete SSU to LSU rDNA sequences (GenBank accession numbers JF906989-JF906999) of 11 Alexandrium strains and used these to design rRNA targeted oligonucleotide probes. Three ribotype-specific probes, M-GC-1, M-PC-2, and M-PC-3, were designed. The former is specific for the GC clade ("Global clade") of A. minutum, the majority of which have been found non-toxic, and the latter two are specific for the PSP (paralytic shellfish poisoning)-producing PC clade ("Pacific clade"). The specificity of these three probes was confirmed by FISH. All cells in observed fields of view were fluorescently labeled when probes and target species were incubated under optimized FISH conditions. However, the accessibility of rRNA molecules in ribosomes varied among the probe binding positions. Thus, there was variation in the distribution of positive signals in labeled cells within nucleolus and cytosol (M-GC-1, M-PC-3), or just nucleolus (M-PC-2). Our results provide a methodological basis for studying the biogeography and population dynamics of A. minutum, and providing an early warning of toxic HABs.展开更多

An evaluation of these two female characters shows and reflects that their self-damaging behavior emerges from low self esteem, which results from a number of reasons. The self-damaging behavior introduced by these women involves: self-imposed isolation, women madness, purposive accidents, physical self-abuse, and most consequently, conscious pursuit of destructive relationships with men. Although Dickens clearly means no maliciousness to women in his works, the great Victorian marital upheaval of June, 1858, is illustrative of Dickens's ambivalent attitude towards women, especially towards strong women展开更多
