放养密度对大杂交鲟生长性能的影响及生理应答机制 被引量:5

为探讨放养密度对鱼类生长及生理应答机制的影响规律和作用,作者以大规格大杂交鲟(达氏鳇(Huso dauricus)♀×施氏鲟(Acipenser schrenckii)♂)为实验材料,研究了平均初始体质量约为243 g/尾,放养密度分别为6(SD1组)、9(SD2组)、12(... 为探讨放养密度对鱼类生长及生理应答机制的影响规律和作用,作者以大规格大杂交鲟(达氏鳇(Huso dauricus)♀×施氏鲟(Acipenser schrenckii)♂)为实验材料,研究了平均初始体质量约为243 g/尾,放养密度分别为6(SD1组)、9(SD2组)、12(SD3组)、15 kg/m3(SD4组)条件下,不同放养密度处理70 d后的实验鱼生长性能变化及生理应答机制。结果显示,放养密度对大杂交鲟肥满度影响不显著,SD2组鱼类具有最大的特定生长率和生长效率,随着放养密度增加,日增质量显著降低(P<0.05),特定生长率和生长效率下降。测定了血液甲状腺素(T4)、三碘甲腺原氨酸(T3)和皮质醇水平变化,发现放养密度能引起大杂交鲟3个血液生理指标发生显著改变;随着养殖时间推移,T3和皮质醇浓度显著升高,T4浓度显著下降(P<0.05)。这些结果说明神经内分泌活动的变化引起大杂交鲟血液生理指标变化,进而影响实验鱼生长性能。因此,在该养殖条件下推荐的养殖密度为9 kg/m3。 展开更多

用生物化学方法测定施氏鲟Acipenser schrenckii、大杂交鲟A.schrencki i(♂)×Huso dauricus(♀)及西伯利亚鲟A.baeri成熟卵子及体腔液的酶、维生素、微量元素及氨基酸组成和含量。结果表明:施氏鲟与大杂交鲟的卵径显著大于西伯利... 用生物化学方法测定施氏鲟Acipenser schrenckii、大杂交鲟A.schrencki i(♂)×Huso dauricus(♀)及西伯利亚鲟A.baeri成熟卵子及体腔液的酶、维生素、微量元素及氨基酸组成和含量。结果表明:施氏鲟与大杂交鲟的卵径显著大于西伯利亚鲟(P<0.05)。吸水后大杂交鲟的卵径显著大于施氏鲟和西伯利亚鲟(P<0.05)。大杂交鲟卵中谷草转氨酶(GOT,2.83U·g-1)、酸性磷酸酶(ACP,0.69 U·g-1)和碱性磷酸酶(AKP,0.81 U·mg-1)含量最高,均无显著性差异(P>0.05),仅西伯利亚鲟卵内琥珀酸脱氢酶(SDH)的活力显著升高(P<0.05)。3种鲟鱼卵与体腔液中蛋白质、微量元素及维生素组成差异显著。大杂交鲟体腔液内的ACP活力显著高于施氏鲟体腔液内的ACP活力(P<0.05)。西伯利亚鲟(0.51±0.16μmol·g-1)卵内的Fe含量显著高于施氏鲟(0.35μmol·g-1)和大杂交鲟(0.42±0.12μmol·g-1),而体腔液内均未检测到Fe和Zn,但体腔液中Vc(4.00±2.44μg·L-1)含量显著低于大杂交鲟(5.58±1.53μg·L-1)和施氏鲟(5.47±2.32μg·L-1)(P<0.05)。3种鲟鱼卵及体腔液内的酶、微量元素、维生素等组成相同,但SDH和ACP活力、Fe和Vc含量存在组织和种间差异,在亲鱼培育时应区别培育。 展开更多

采用形态学和多变量形态度量方法,对养殖施氏鲟(Acipenser schrenckii)、达氏鳇(Huso dauricus)及其杂交种(大杂交)(达氏鳇(♀)×施氏鲟(♂))的形态差异和判别进行了分析。结果表明:(1)养殖施氏鲟、达氏鳇及大杂交头部形态差异明显... 采用形态学和多变量形态度量方法,对养殖施氏鲟(Acipenser schrenckii)、达氏鳇(Huso dauricus)及其杂交种(大杂交)(达氏鳇(♀)×施氏鲟(♂))的形态差异和判别进行了分析。结果表明:(1)养殖施氏鲟、达氏鳇及大杂交头部形态差异明显,口唇形状、吻须形状及长度、鳃盖膜形态差异显著。在7项可数性状中,大杂交胸鳍、侧骨板、腹骨板和鳃耙数偏母本性状(HI>55),臀鳍条数明显地偏父本性状(HI<55),背骨板数偏离双亲性状,背鳍条数为中间性状。可数性状的平均杂交指数为74.97,总体上偏向于母本。可数性状中鳃耙数可作为判断三者所属的指标;(2)单因子方差分析显示,大杂交与施氏鲟有5个比例性状存在显著差异(P<0.05),与达氏鳇有4个比例性状存在显著差异(P<0.05);(3)聚类分析表明,大杂交与达氏鳇在外观形态上更为近似,二者聚在一枝,然后与施氏鲟聚集;(4)主成分分析构建了4个主成分,其贡献率分别为41.429%、37.809%、7.869%和5.790%,累积贡献率为92.896%,主成分分散图表明大杂交在形态差异上独立于施氏鲟和达氏鳇;(5)采用逐步判别分析获得5个比例性状构建了区分上述三个种的判别函数,判别函数预测分类总体准确率达96.67%,其中对大杂交的判断准确率达100%。研究认为,杂交鲟在形态上较接近于其母本达氏鳇,但可以通过形态判别分析将大杂交与母本达氏鳇和父本施氏鲟进行区分判别。 展开更多

以大杂交鲟大规格鱼种(达氏鳇♀×史氏鲟含,初始体重约243g/尾)为实验材料,研究在6、9、12和15kg/m^34个放养密度条件下,饲养70d后,其血液生理生化指标的变化情况。结果表明,高密度产生的拥挤胁迫能引起大杂交鲟鱼种血液生理指标和... 以大杂交鲟大规格鱼种(达氏鳇♀×史氏鲟含,初始体重约243g/尾)为实验材料,研究在6、9、12和15kg/m^34个放养密度条件下,饲养70d后,其血液生理生化指标的变化情况。结果表明,高密度产生的拥挤胁迫能引起大杂交鲟鱼种血液生理指标和主要生化指标发生明显的改变。随着放养密度的升高,血红蛋白浓度升高,白细胞数目、淋巴细胞数目、中性粒细胞数目、血栓细胞数量增加,说明鱼体自身处于免疫应激状态;红细胞数目在最高密度条件下降低,说明呼吸能力在高密度养殖备件下受到抑制;鱼体内血清中总蛋白、尿酸和尿素浓度随着放养密度增大显著升高(P<0.05),葡萄糖、甘油三酯、总胆固醇浓度随密度增大而降低(P<0.05),表明高密度养殖对大杂交鲟血液和体内消化酶活力具有一定负面影响;鱼体内血清中的谷丙转移酶、碱性磷酸酶、天门冬氨酸氨基转移酶三种酶活性也随着放养密度增大而显著增大(P<0.05),而总胆红素浓度随着放养密度的增大而显著降低(P<0.05),这均表明过高放养密度下鱼体肝脏组织受到一定损伤。本文研究结果显示该规格下大杂交鲟的适宜放养密度不超过12kg/m^3,否则对鱼类血液生理和血清生化组份均产生不良影响。 展开更多

[Objective] Taqman Quantitative PCR technique was adopted to detect the copies of exogenous nos terminator in transgenic hybrid soybean.[Method] With soybean Lectin as the endogenous reference gene,and gene complex DNA in non-GMO soybeans as the endogenous reference standard,the method of gradient dilution was used for separately calculate Ct value of endogenous reference gene and plasmid DNA and correlation standard curve equation of logarithm of copies,and then to calculate the copies of samples through substituting thus-obtained Ct into the standard curve equation.[Result] The standard curve equation of endogenous reference gene is y=-3.422x+35.201,R2=0.998;and the standard curve equation of exogenous gene is y=-3.348x+34.890,R2=0.999.Nos terminator and its lower boundary sequences in transgenic soybean is of single copy.[Conclusion] The study has provided a theoretical basis for determining exogenous gene copies in transgenic soybean. 展开更多

'Chinese Spring') X barley (Hordeum vulgare L. cv. 'Betzes') was analyzed, and six disomic alien substitution lines were screened by GISH. The chromosome configurations in pollen mother cells at meiotic metaphase I (PMCs M I) of F, from each disomic substitution line respectively crossed with double ditelocentric lines 2A, 2B and 2D of 'Chinese Spring' were observed, and a set of wheat-barley disomic alien substitution lines 2H(A), 2H(B) and 2H(D) were obtained. The RFLP analysis with the probe psr131 on the short arm of wheat homeologous group 2 combining with four restriction enzymes were carried out. The results indicated that the probe psr131 could be used as molecular marker to tag the barley chromosome 2H. The barley chromosome 2H had good genetic compensation ability for wheat chromosomes 2B and 2D in vitality and other agronomic characters. The result of testing seed was that the wheat appearance starch quality had been changed from the half-farinaceous of 'Chinese Spring' to the half-cutin of substitution lines by transferring the barley chromosome 2H to wheat. 展开更多

It has been reported that endosperm undergoes programmed cell death (PCD) during maize kernel development.Both bz1 (bronze ) and bz2 are anthocyanin biosynthetic genes,and related to development of aleuronic layer of maize seeds.Tyramide signal amplification fluorescence in situ hybridization (TSA FISH) is a novel and high sensitive FISH technique,which is suitable for routine application in plant cytogenetic research.Using this technique,we physically mapped the bz1 gene onto the short arm of chromosome 9 and the long arm of chromosome 1;the percentage distances from centromere to hybridization site were 40.2,75.4 respectively,and the bz2 onto the long arm of chromosome 1 and the short arm of chromosome 5;the percentage distances from centromere to hybridization site were 21.6,15.3 separately.The TSA FISH techniques of small low copy DNA sequences for plants are discussed. 展开更多

Fruit ripening is a complex process and is regulated by many factors. Ethylene and polygalacturonase (PG), lipoxygenase (LOX), expansin (EXP) are all critical regulating factors in fruit ripening and softening process. With antisense ACS tomato, Nr mutant tomato and cultivated tomato as materials, Northern blot hybridization showed that PG, LeEXP1 and LOXexpressed differently in different parts of cultivated tomato fruit during ripening, which was related to fruit ripening. The ripening process of columella and radial pericarp was faster than pericarp. In both Nr mutant and antisense ACS transgenic tomato fruit, expression levels ofPG, LeEXPI and LOXwere generally lower than those in cultivated fruit but still related to fruit ripening. The expression levels ofPG, LeEXP1 and LOX increased in the mature green tomato fruits after 0.5 h treatment with ethylene (100 μL/L). These results indicate that gene expression ofPG, LeEXP1 and LOXwere positively regulated by ethylene. The time and cumulative effect of the concentration exists in the expression of PG regulated by ethylene. The regulation of LOX expression mainly depended on the fruit development after great amount of ethylene was produced. PG played a major role in ripening and softening of tomato fruit, and cooperated with the regulation of EXP and LOX. 展开更多

Mutations in cardiac troponin I (cTnI) gene were assessed based on gene-chip technology.Special probes were designed to fabricate the low-density gene-chip,which could detect the mutations in exons 3,5,7,and 8 of the cTnI gene simultaneously.For each exon,two oligonucleotide sequences labeled with fluorescein at the 5'-end were designed,one (oligonucleotide Ⅰ) simulating the wild type and the other (oligonucleotide Ⅱ) simulating the mutant.Oligonucleotides Ⅰ and Ⅱ were mixed together to simulate the heterozygote.After optimizing the hybridization protocols,the fabricated gene-chip could detect the mutations in the exons of the cTnI gene with relative high sensitivity and specificity.The fully complementary probe gave a fluorescent signal almost 50% stronger than that of the one-base mismatched one,which is in accordance with the result from a theoretical estimate. An applicable special gene-chip is available to investigate and diagnose familial hypertrophic cardiomyopathy (FHCM) after further improvement. 展开更多

The dinoflagellate Alexandrium minutum is often associated with harmful algal blooms (HABs). This species consists of many strains that differ in their ability to produce toxins but have similar morphology, making identification difficult. In this study, species-specific rRNA probes were designed for whole-cell fluorescence in situ hybridization (FISH) to distinguish A. minutum from two phylogenetic clades. We acquired the complete SSU to LSU rDNA sequences (GenBank accession numbers JF906989-JF906999) of 11 Alexandrium strains and used these to design rRNA targeted oligonucleotide probes. Three ribotype-specific probes, M-GC-1, M-PC-2, and M-PC-3, were designed. The former is specific for the GC clade ("Global clade") of A. minutum, the majority of which have been found non-toxic, and the latter two are specific for the PSP (paralytic shellfish poisoning)-producing PC clade ("Pacific clade"). The specificity of these three probes was confirmed by FISH. All cells in observed fields of view were fluorescently labeled when probes and target species were incubated under optimized FISH conditions. However, the accessibility of rRNA molecules in ribosomes varied among the probe binding positions. Thus, there was variation in the distribution of positive signals in labeled cells within nucleolus and cytosol (M-GC-1, M-PC-3), or just nucleolus (M-PC-2). Our results provide a methodological basis for studying the biogeography and population dynamics of A. minutum, and providing an early warning of toxic HABs. 展开更多

To address this, we conducted a parentage analysis on a complete 3×3 diallel cross of clam Meretrix meretrix using eight microsatellite markers. The genetic diversity of the parents was higher than that of their respective offspring in most crosses (8/9). Sires or dams from the same family contributed unequally to the pool of offspring from a particular cross, and the same parent clam exhibited large variation in parental contribution among different crosses. The variance in male contribution was higher than that of the female contribution in most crosses, suggesting that male contribution was more skewed than for females. The No/N ratio for nine crosses ranged from 0.58 to 0.86. There was no linear relationship between the sex ratio and the Ne/Nratio (P〉0.05). Moreover, a sex ratio closer to one-to-one does not necessarily mean a larger effective population size. A solution to small effective population size in commercial breeding programs is increasing broodstoek numbers and attempting to maintain a balanced sex ratio. 展开更多

A lithium-assisted approach has been developed for the exfoliation of pristine graphite, which allows the large-scale preparation of few-layer graphene nanosheets. The process involves an unexpected physical insertion and exfoliafion, and the graphene nanosheets prepared by this method reveal undisturbed sp2-hybridized structures. A possible two-step mechanism, which involves the negative charge being trapped around the edges of the graphite layers and a subsequent lithiation process, is proposed to explain the insertion of lithium inside the graphite interlayers. If necessary, the present exfoliation can be repeated and thinner (single or 2-3 layer) graphene can be achieved on a large scale. This simple process provides an efficient process for the exfoliation of pristine graphite, which might promote the future applications of graphene. 展开更多

OBJECTIVES:To investigate the influence of catgut implantation at acupoints on leptin resistance(LR) and insulin resistance(IR) in the simple obesity rat.METHODS:Rats were made obese with high-fat diets,after which surgical catgut was implanted at Zusanli(ST 36) and Neiting(ST 44) acupoints once a week for 4 weeks(implantation group).Rats from the implantation group were compared with normal rats and unoperated obese rats(control group).Gene expression of the leptin receptor(OB-R) was evaluated using RT-PCR and northern blot.Serum and hypothalamus leptin and insulin(INS) levels were determined by radioimmunoassay.Body weight,Lee's index,body fat,serum and hypothalamus leptin and INS levels,and hypothalamic OB-R gene expression were determined before and after treatment.RESULTS:Body weights,Lee's index,body fat,and serum leptin and INS levels were significantly higher in obese than in normal rats.Hypothalamic leptin and INS levels and OB-R gene expression were significantly lower in obese rats.Catgut implantation at acupoint promoted weight loss and decreased serum leptin and INS levels.Hypothalamic leptin and INS levels and OB-R gene expression increased significantly.CONCLUSIONS:Catgut implantation at acupoint adjusts central and peripheral leptin and promotes hypothalamic OB-R gene expression.This may be an important method for regulation of LR,IR and abnormal endocrinology and metabolism. 展开更多

A versatile DNA recognition system using cadmium sulfide nanoparticles (CdS NPs) as labels was proposed for ultrasensitive detection of specific sequence DNA based on target recycling. This strategy utilized the magnetic particles (MNPs) for the immobilization of linker DNA and CdS NPs and the subsequent target DNA hybridization. Using the unique characteristic of nicking endonuclease for cutting one specific strand of double strand DNA (ds DNA), the linker DNA could be transected and the target DNA could be liberated for re-hybridization and hence the amount of released CdS NPs was enhanced. Due to the advantage of the MNPs and signal amplification from the target recycling, the analyte DNA could be detected by the square-wave stripping voltammetry (SWV) in a wide linear range from 0.4 fM to 100 fM with the detection limit down to 0.08 fM. The proposed DNA detection strategy possesses high sensitivity, satisfactory reproducibility and excellent stability, which might have potential in other DNA biological assays. 展开更多
