Both toluidine blue and Alcian blue were the excellent stains generally, but Alcian blue stained more porcine mast cells than did toluidine blue( P <0 01). Staining with toluidine blue of a wide pH range(from 0 1 to 7 0)showed that porcine mast cells were not very pH dependent, but the dye at pH 0 5 seemed to have the strongest affinity for all mast cells in pigs and it was also suitable for bovine and ovine mast cell staining. In the three species, unlike in rodents, the Alcian blue method did not distinguish between mast cells in the intestinal mucosa(MMC)and those in the connective tissue of the intestinal submucosa, tongue and skin(CTMC). Porcine CTMC,but not MMC,fluoresced strongly when stained with berberine sulphate or with a mixture of berberine sulphate and acridine orange. It suggested that porcine CTMC contained heparin proteoglycan.展开更多

In the present study, the authors have obtained cDNAs to all cistrons of a Chinese SMV isolate, SMV_ZK, by RT_PCR. By analysing the nucleotide and amino acid sequence of the HC_PRO, NIb and CP cistrons, it was found that SMV_ZK was highly homologous to the G2 strain of SMV, thus confirming the existence of G2_like isolates in soybean crop in China. The amplified cDNAs were directly cloned into a bacterial expression vector. With the exception of the P3 cistron, expression of the cDNAs of all other cistrons in bacteria gave rise to polypeptides of expected molecular weight. The expressed viral proteins were subsequently purified by gel elution. The preparation of viral_specific cDNAs and gene products will be useful in future functional study of the SMV genome.展开更多

The mean group size was 6.2 in August 2003 and 6.4 in August 2005. Two subadult males disappeared from their natal home range and three newborns were given birth in Group 3 (G3) and G4 during this study. The two adult females in G1, G2 and G3 gave births and/or carried babies but at different times. There was no aggressive or dominating behaviour observed between the two adult females. One floating female was first seen in G3's territory on April 15, 2005. The two resident females interrupted her duet with adult male and chased her. We did not observe adult male chased this floating female and she left G3's territory 10 days later. Sub-adult males often kept distance with the family, and they often sang solo bouts in their natal territory before they dispersed. The sub-adult males and females dispersed from natal territory and two adult resident females rejected the third one, which might were the reasons why the black gibbon groups were polygyny in Dazhaizi.展开更多
