[Objective]The aim of this study was to set up a high performance liquid chromatography for rapid determination of isoflavones from soybean and analyze the correlation between isofalvone content and protein or fat con...[Objective]The aim of this study was to set up a high performance liquid chromatography for rapid determination of isoflavones from soybean and analyze the correlation between isofalvone content and protein or fat content. [Method]The isoflavones were firstly extracted by 80% methanol and then hydrolyzed at 100 ℃. The chromatographic separation adopted a reversed-phase C18 analytical column with binary high-pressure gradient elution,while its analysis time was 25 min and column temperature was 40 ℃. The diode array detector was used for monitoring with wavelength of 260 nm. The correlation between isofalvone content and protein or fat content was analyzed by data processing system Origin 6.0. [Result]The high performance liquid chromatograph for determination of isoflavones from soybean was verified to be accurate and reliable by methodology. The isoflavones of 85 soybean lines cultivated in Jilin Province were determined,and the results primarily showed the characters and ranges of isoflavones from soybean lines cultivated in Jilin Province,while the isoflavone content of soybeans ranged from 2.29 to 4.89 mg/g,and the average content was 3.36 mg/g. The isoflavone content of 5 soybean lines exceeded 4 mg/g,while there was a remarkably negative correlation between isoflavone content and protein content,and there was no significant positive correlation between isoflavone content and fat content. [Conclusion]The isoflavone content of soybean lines cultivated in Jilin Province is higher,so it is feasible for breeding the soybean lines with high isoflavone content and fat contetnt.展开更多

[Result] According to the experimental results, the genotype variance, environmental variance and G x E interaction variance of catechin and kaempferol contents show the same trend: genotype variation 〉 environmental variation 〉 G × E interaction variation, which all reach a extremely significant level; the genotype variance, envi- ronmental variance and G × E interaction variance of quercetin and total flavonoid contents show the same trend: genetype variation 〉 G × E interaction variation 〉 environmental variation, which all reach a extremely significant level; the genotype variance and environmental variance of myricetin content both reach a extremely sig- nificant level, while the G × E interaction variance reaches a significant level, showing an order of genotype variation 〉 environmental variation 〉 G × E interaction variation; the genotype variance, environmental variance and G x E interaction vari- ance of total flavonoid content show an order of genotype variation 〉 environmental variation 〉 G × E interaction variation. Among different barley varieties, Ziguang- mangluoerling and Kuanyingdamai in Qujing, Kunming and Baoshan have relatively high content of quercetin, while other barley varieties barely contain any quercetin. The study site is located at Nihegou Watershed in the Souther...The purpose of this study was to assess the effect of long-term cultivation and water erosion on the soil organic carbon (OC) in particle-size fractions. The study site is located at Nihegou Watershed in the Southern Loess Plateau, China. The soil at this site is loess with loose and silty structure, and contains macropores. The results showed that the OC concentrations in sediments and in the particle-size fractions of sediments were higher than those in soils and in the particle-size fractions of soils. The OC concentration was highest in the clay particles and was lowest in the sand particles. Clay particles possessed higher OC enrichment ability than silt and sand particles. The proportions of OC in the silt fractions of soil and sediment were the highest (mean value of 53.87% and 58.48%, respectively), and the total proportion of OC in the clay and silt fractions accounted for 96% and 98% of the total OC in the soil and sediment, respectively. The loss of OC was highest in silt particles, with an average value of 0.16 Mg ha^-1 y^-1, and was lowest in the sand (0.003 Mg ha^-1 y^-l). This result suggests that the fine particle-size fraction in the removed sediment may be an important indicator to assess soil OC losses.展开更多
