目的:观察中药祛异康对子宫内膜异位症模型大鼠肝、肾脏器形态结构的影响,为临床用药提供进一步的帮助。方法:采用手术移植法建立大鼠子宫内膜异位症模型,随机分为空白对照组、模型组、丹那唑组、祛异康组。造模成功各组给药4W,择动情... 目的:观察中药祛异康对子宫内膜异位症模型大鼠肝、肾脏器形态结构的影响,为临床用药提供进一步的帮助。方法:采用手术移植法建立大鼠子宫内膜异位症模型,随机分为空白对照组、模型组、丹那唑组、祛异康组。造模成功各组给药4W,择动情期处死,分别取肝、肾脏器称重、HE染色镜下观察。结果:丹那唑组的肝脏重量和肝脏指数明显高于空白组及祛异康组(P<0.05);祛异康组与空白组比较无明显差异(P>0.05);丹那唑组、模型组及祛异康组与空白组肾脏重量和肾脏指数比较无明显差异(P>0.05);大鼠肝、肾脏组织切片镜下观察见丹那唑组大鼠肝脏中等程度损害,有统计学意义(P<0.05);对大鼠肾脏组织形态变化影响较小,无统计学意义(P>0.05)。结论:中药祛异康与丹那唑相比,在治疗子宫内膜异位症临床用药上具有相对的安全性。 展开更多

为探讨和胃接骨胶囊对实验性大鼠骨折愈合过程中骨形态发生蛋白-2表达的影响,取SD大鼠60只,随机分为生理盐水组、麝香接骨丹组、和胃接骨胶囊组,徒手造成大鼠下肢胫腓骨标准骨折模型,造模成功后分别于1、2、4、6周每组断颈法处死5只大鼠... 为探讨和胃接骨胶囊对实验性大鼠骨折愈合过程中骨形态发生蛋白-2表达的影响,取SD大鼠60只,随机分为生理盐水组、麝香接骨丹组、和胃接骨胶囊组,徒手造成大鼠下肢胫腓骨标准骨折模型,造模成功后分别于1、2、4、6周每组断颈法处死5只大鼠,取骨痂组织制成病理切片,采用免疫组化染色技术进行染色,并用图像分析系统对骨形态发生蛋白-2的表达进行定量分析。结果显示,在骨折愈合过程中各组骨形态发生蛋白-2含量的总体变化趋势基本相同,骨形态发生蛋白-2表达的高峰都出现在骨折后第2周,但和胃接骨胶囊组骨形态发生蛋白-2阳性产物含量优于同期其他两组(P<0.05)。表明和胃接骨胶囊能诱导骨形态发生蛋白-2的合成,可能是其促进骨折愈合的机理之一。 展开更多

为观察糖尿病大鼠早期视网膜神经细胞的损害是否早于视网膜微循环改变 ,用链脲佐菌素 (STZ ,6 0mg/kg)建立糖尿病动物模型 ,分别于模型建立后 1周、2周、1月处死大鼠 ,电镜观察大鼠视网膜及其微血管的变化。结果 :与正常对照组比较 ,糖... 为观察糖尿病大鼠早期视网膜神经细胞的损害是否早于视网膜微循环改变 ,用链脲佐菌素 (STZ ,6 0mg/kg)建立糖尿病动物模型 ,分别于模型建立后 1周、2周、1月处死大鼠 ,电镜观察大鼠视网膜及其微血管的变化。结果 :与正常对照组比较 ,糖尿病大鼠于模型建立后 1周、2周 ,其神经网膜神经细胞和视网膜微血管无明显改变 ;模型建立后 1月 ,其视网膜色素上皮层、神经网膜微血管基底膜和周细胞无明显改变 ,但其神经网膜从外丛状层至神经纤维层有明显变化 ,表现为线粒体数目减少 ,线粒体肿胀 ,嵴变短、消失和髓样变性。神经纤维层和内、外网状层细胞突起水肿 ,在神经纤维层尤其明显 ,在神经节细胞层可见暗细胞形成。由于神经纤维层细胞突起水肿明显 ,压迫其周围微血管 ,导致微血管管腔狭窄 ,甚至闭锁 ,但微血管内皮细胞及基底膜未见异常。结果提示 :糖尿病早期大鼠在视网膜神经细胞损害已发生时 ,视网膜微血管内皮细胞和基底膜超微结构未见异常 ,神经网膜神经细胞的损害可能是糖尿病视网膜病变 (DR)的早期表现。 展开更多

目的 :观察耳尖放血疗法对高血压肝阳上亢证模型大鼠血清NO浓度的影响 ,以探讨耳尖放血疗法对高血压肝阳上亢证的实验疗效与作用机理。方法 :采用自发性高血压大鼠 (SHR)加灌附子汤制造高血压肝阳上亢证大鼠模型 ,40只SHR大鼠随机分成... 目的 :观察耳尖放血疗法对高血压肝阳上亢证模型大鼠血清NO浓度的影响 ,以探讨耳尖放血疗法对高血压肝阳上亢证的实验疗效与作用机理。方法 :采用自发性高血压大鼠 (SHR)加灌附子汤制造高血压肝阳上亢证大鼠模型 ,40只SHR大鼠随机分成空白对照组、模型对照组、氨氯地平治疗组、耳尖放血治疗组、耳尖放血合氨氯地平治疗组等 5组 ,在观察高血压肝阳上亢证候的变化同时 ,用硝酸还原酶法测定大鼠血清NO浓度。结果 :三个治疗处理组的外观、易激惹程度 ,饮水量 ,血压等肝阳上亢症候较对照组有明显好转 ,血清NO浓度显著高于对照组 (P<0 .0 1) 。结论 :耳尖放血疗法对高血压肝阳上亢证有确切的治疗作用 ,其作用可能与血清NO浓度的变化有关。 展开更多

为研究红曲对体外培养大鼠成骨细胞BMP-2表达的影响,从而为治疗骨质疏松症开发出中药新药提供细胞学依据。取成年SD大鼠48只,随机将大鼠分为A组(空白组)、B组(倍美力组)、C组(普拉固组)、D、E、F组(红曲高、中、低剂量组)6组,按10ml.kg-... 为研究红曲对体外培养大鼠成骨细胞BMP-2表达的影响,从而为治疗骨质疏松症开发出中药新药提供细胞学依据。取成年SD大鼠48只,随机将大鼠分为A组(空白组)、B组(倍美力组)、C组(普拉固组)、D、E、F组(红曲高、中、低剂量组)6组,按10ml.kg-1分别予以生理盐水,倍美力水溶液(0.00625mg.ml-1),普拉固水溶液0.2mg.ml-1)及红曲水提液(1.25g.ml-1、0.625g.ml-1、0.125g.ml-1)灌胃。灌胃10天后,腹腔静脉取血制备含药血清;将从胎鼠颅骨取材的成骨细胞培养于含药血清培养液中,然后应用BMP-2免疫组化染色方法观察红曲对体外培养成骨细胞BMP-2表达的影响。结果显示,通过免疫组化染色发现BMP-2棕色深染位于细胞浆,红曲高、中剂量组成骨细胞棕色深染数量明显高于空白组,低剂量组与空白组无明显差异,阳性细胞比例呈剂量依赖性增长。表明红曲含药血清可以呈剂量依赖性地促进成骨细胞BMP-2表达;在细胞和分子水平为中药红曲治疗骨质疏松症提供了客观化的依据。 展开更多

目的:观察延胡索碱预处理对大鼠心肌是否有抑制心肌细胞凋亡和下调fas基因的表达以保护心肌细胞的作用。方法:将清洁级成年SD大鼠60只随机分为6组,采用TUNEL法和S-P免疫组化法分别检测细胞凋亡和fas基因的表达。结果:假手术组心肌仅偶... 目的:观察延胡索碱预处理对大鼠心肌是否有抑制心肌细胞凋亡和下调fas基因的表达以保护心肌细胞的作用。方法:将清洁级成年SD大鼠60只随机分为6组,采用TUNEL法和S-P免疫组化法分别检测细胞凋亡和fas基因的表达。结果:假手术组心肌仅偶见细胞凋亡发生,模型组可见大量胞核呈深浅不一棕褐色的凋亡细胞,心肌细胞凋亡指数明显高于假手术组(P<0.01);缺血预处理组和延胡索碱高、中、低剂量预处理组凋亡心肌细胞和心肌细胞凋亡指数均较模型组明显减少(P<0.01,P<0.05);而缺血预处理组和延胡索碱高剂量组预处理组间差异无显著性(P>0.05)。假手术组极少数心肌细胞Fas基因蛋白表达阳性;与假手术组比较,缺血再灌注后Fas基因蛋白表达明显加强,心肌细胞Fas蛋白阳性表达指数(PEI)明显高于假手术组(P<0.01);缺血预处理组和延胡索碱高剂量组预处理组Fas蛋白阳性表达指数(PEI)均较模型组明显减少(P<0.05),组间差异无显著性(P>0.05)。结论:高剂量组延胡索碱预处理和缺血预处理效应相似,均能明显减少缺血再灌注时凋亡心肌细胞数量和Fas蛋白阳性表达指数(PEI),降低心肌细胞凋亡和拮抗心肌细胞凋亡的发生。 展开更多

AIM: To observe the therapeutic efficacy of Baicalin in rats with severe acute pancreatitis (SAP) and explore its therapeutic mechanisms. METHODS: The SAP rat models were randomly divided into the model control group, Baicalin treatment group, octreotide treatment group and sham operation group. All groups were randomly subdivided into 3 h, 6 h and 12 h groups with 15 rats in each group. The survival, ascites volume and pathological changes of pancreas in all rats were observed at different time points after operation. The plasma amylase content and serum TNF-α, IL-6, malonaldehyde (MDA) and PLA2 contents were also determined. RESULTS: The survival was not obviously different between the treated groups, and was significantly higher in treated groups at 12 h compared to the model control group (P 〈 0.05, 15 vs 10). The ascites/body weight ratio at 3 h and 6 h was significantly lower in Baicalin treatment group compared to the model control group and octreotide treatment group (P 〈 0.05, 1.00 vs 2.02 and 1.43 and P 〈 0.001, 2.29 (1.21) vs 2.70 (0.80) and 2.08 (2.21), respectively). The contents of amylase, TNF-α, IL-6, MDA and PLA2 were significantly lower in the treated groups than in the model control group (P 〈 0.05, 4342 vs 5303, 5058 vs 6272 in amylase, P 〈 0.01, 21.90 vs 36.30, 23.80 vs 39.70, 36 vs 54.35 in MDA and 56.25 vs 76.10 in PIA2, or P 〈 0.001, 65.10 and 47.60 vs 92.15 in TNF-α, 3.03 vs 5.44, 2.88 vs 6.82, 2.83 vs 5.36 in IL-6, respectively). The pathological scores of pancreas in the treated groups were significantly lower than that in the model control group (P 〈 0.05, 9.00 vs 10.05, 6.00 vs 9.00, 8.00 vs 10.05), but no marked difference was found between the treated groups. CONCLUSION: The Baicalin injection has significant therapeutic effects on SAP rats, its effects are similar to those of octreotide. The Baicalin injection is also cheap and has a big application range, quite hopefully to be used in clinical treatment of SAP. 展开更多

AIM: To investigate the effects of garlicin on apoptosis and expression of bcl-2 and bax in lymphocytes in rat model of ulcerative colitis (UC). METHODS: Healthy adult Sprague-Dawley rats of both sexes, weighing 180±30 g, were employed in the present study. The rat model of UC was induced by 2,4,6-trinitrobenzene sulfonic acid (TNBS) enema. The experimental animals were randomly divided into garlicin treatment group (including high and low concentration), model control group, and normal control group. Rats in garlicin treatment group and model control group received intracolic garlicin daily at doses of 10.0 and 30.0 mg/kg and equal amount of saline respectively 24 h after colitis model was induced by alcohol and TNBS co-enema. Rats in normal control group received neither alcohol nor only TNBS but only saline enema in this study. On the 28^th d of the experiment, rats were executed, the expression of bd-2 and bax protein was determined immunohistochemically and the apoptotic cells were detected by the terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate fluorescence nick end labeling (TUNEL) method. At the same time, the rat colon mucosal damage index (CMDI) was calculated. RESULTS: In garlicin treatment group, the positive expression of bcl-2 in lymphocytes decreased and the number of apoptotic cells was more than that in model control group, CMDI was lower than that in model control group. The positive expression of bax in lymphocytes had no significant difference. CONCLUSION: Garlicin can protect colonic mucosa against damage in rat model of UC induced by TNBS enema. 展开更多

TE (zinc and copper) are constituents of antioxidant enzymes. Iron is involved in the pathogenesis of chronic inflammation. The aim was to evaluate zinc and copper status and the effects of iron manipulation in experimental colitis.METHODS: Twenty-four male Sprague-Dawley rats were divided into four groups: standard diet, iron-deprived diet,iron-supplemented diet, and sham-treated controls.Macroscopic damage was scored. DNA adducts were measured in the colon. Liver and colonic concentration of TE were measured.RESULTS: Macroscopic damage was reduced in irondeprived groups and increased in iron-supplemented rats.Damage to the DNA was reduced in iron-deprived groups and increased in iron-supplemented groups. Liver and colonic iron concentrations were reduced in iron-deprived and increased in iron-supplemented rats. Liver zinc concentration was reduced after supplementation whereas colonic levels were similar in controls and treated rats. Liver copper concentration was reduced in all the colitic groups except in the iron-supplemented group whereas colonic concentration was increased in iron-deprived rats.CONCLUSION: Iron deprivation diminishes the severity of DNBS colitis while supplementation worsens colitis. Zinc and copper status are modified by iron manipulation. 展开更多
