Previously, we have purified Jerdonitin from Trimeresurusjerdonii venom. Compared with other P-Ⅱ class snake venom metalloproteinases (SVMPs), Jerdonitin has a primary structure comprising metalloproteinase and disintegrin domains. However, no hemorrhagic and fibrinogenolytic activities were detected for Jerdonitin. We thought that organic buffer of high performance liquid charamatography (HPLC) might affect its enzymatic activity. In this study, we purified Jerdonitin by another procedure excluding the HPLC. It was homogenous as judged by SDS-PAGE and had an apparent molecular weight of 36 kDa under non-reducing conditions and 38 kDa under reducing conditions, respectively. Like other typical metalloproteinases, Jerdonitin preferentially degraded alpha-chain of human fibrinogen and this fibrinogenolytic activity was completely inhibited by EDTA, but not by PMSF. It was interesting that Jerdonitin did not induce hemorrhage after intradermal injection in mice. 展开更多

Recycling human urine for farming was assessed in a peri-urban Kyanja parish, Kampala district, and in a rural Migyera parish, Nakasongola district, to demonstrate its potential and develop local use guidelines. Test crops were maize, Nakati (Solanum aethiopicum), kale, spinach, cabbage, tomatoes, egg plants. Urine-water mixtures (0, 10%, 20%, 30% urine) were applied weekly or bi-weekly. At Kyanja, 30% urine weekly gave the highest maize yields. Within 2 months, 10% urine weekly increased Nakati yield from 5,444 to 24,667 kg ha^-1. 20% Urine weekly increased kale yield (7,556 to 16,111 kg ha^-1) and spinach (4,222 to 19,022 kg ha^-1). At Migyera, 10% urine weekly increased cabbage yield (4,975 to 16,113 kg ha^-1) but 30% urine weekly decreased cabbage head-weight by 36%. Weekly applied urine produced heavier cabbage heads than bi-weekly (548 g vs. 427 g, P 〈 0.05). LeafN was higher for weekly than bi-weekly applied urine (3.3% vs. 3.0%), implying more protein in the former than the latter. From this study, the following guidelines are proposed: Kyanja area, maize: apply 30% urine weekly for 8-weeks; Nakati: apply 10% urine weekly for 8-weeks; Kale and spinach: apply 20% urine weekly; For Migyera area, cabbage and spinach: apply 10% urine weekly. Apply urine 15 cm around each plant starting 2-weeks after transplanting. So kale and spinach prolong urine application for continued harvesting. 展开更多

The juice was given heat treatment. Yoghurt was formulated and flavored with prepared beetroot (Beta vulgaris L.) juice at different concentration levels (0, 10, 20, 30, 40 and 50 mL). The most preferred flavored yoghurt samples were obtained by sensory scores (color, flavor, mouth feel, aftertaste and overall acceptability). The most preferred sample was then subjected to proximate, physico-chemical, microbiological and micro-nutrient analysis. The result obtained showed that the pH value ranged between 6.5 and 7.8. Thus, this showed that the product was quite good. However, ash and moisture content increased with addition of the beetroot juice, while micro-nutrient, (Mg, Na, K, Ca and Vitamin C) increased with the addition of the juice. There was no significant difference (P 〉 0.05) in the overall acceptability of all products. There was no coliform and mould growth in all the samples. The best concentration level in the samples was 90 mL of yoghurt and 10 mL of beetroot juice. Therefore, the beetroot flavored yoghurt could be said to be nutritious, safe and an acceptable product by the panelists. 展开更多

In order to find the optimal bacteriostatic agents and selectors and their concentrations in genetic transformation of Lespedeza bicolor with BADH as a marker gene, an resistance experiment was carried out with four chemicals, NaCl, betaine aldehyde, PEG and cefotaxime (Cef) added into the media, Different concentrations and combinations of different chemicals were added into the media in the stage of adventitious bud regeneration of cotyledonary nodes, Shoot segment multiplication, and rooting. The results showed that NaCI and betaine aldehyde could inhibit tissue differentiation of L. bicolor, and could thus be used as selectors. Furthermore, 0.8 g/L NaCI could completely inhibit the cotytedonary node differentiation and shoot segment multiplication, and 0.5 g/L NaCI was optimal for inhibition of rooting. It was also found that with the concentration increase of betaine aldehyde in media, adventitious buds regenerated from cotyledonary nodes decreased, and 1.5 g/L was confirmed to be the optimum concentration. Moreover, the mixture of 0.7 g/L NaCI and 0.3 g/L betaine aldehyde significantly inhibited shoot segment multiplication and rooting, so this mixture could be used asa selector. The Cef ex- periment showed that 200-300 mg/L C, ef in media did not inhibit significantly the regeneration of adventitious buds from cotyledonary nodes, and 100 mg/L Cef had little effect on the shoot segment multiplication and rooting. 展开更多

The researches have been carried out in 2011-2012, in the experimental fields of Yuryev Plant Production Institute Kharkiv, Ukraine. The forms of "00" type (a low content of erucic acid and glucosinolates) and self-pollinated lines--110M-10, 127M-10, 132M-10 were studied; the forms of"0+" type--selfed lines 124-10, 221-10, 305-10; the forms of"+0" type--cultivars Victor, Janus, Emerald; forms of "++" type--cultivars Fyedorovskiy, Marens, Uspikh, as well as, self-incompatible forms 5C, 15C-10, 32C-10 and the original sterile form of mutant origin were used. Viability in stigmas of the pistil lasted 3-7 days, in the forms of "+0", "++" type, 7-15 days in the cultivars and forms of "0+", "00" type, 14-21 days in the self-incompatible and sterile forms. A fast loss of sensitivity to pollen by pistil's stigma was observed in the forms of"+0" and "++" types. On the contrary, in self-incompatible and sterile forms was observed a gradual increase of sensitivity of pistil's stigma to pollination and then its gradual fall. A maximum of sensitivity of a pistil's stigma to pollen in "00" and "0+" forms was on the 3rd-6th day after castration. 展开更多
