AIM: To determine the distribution and frequencies of the genotypes and haplotypes of the genes encoding for the glucocorticoid receptor (GR), the tumor necrosis factor (TNF)-α and the interleukin (IL)-10 in c...AIM: To determine the distribution and frequencies of the genotypes and haplotypes of the genes encoding for the glucocorticoid receptor (GR), the tumor necrosis factor (TNF)-α and the interleukin (IL)-10 in childhood Crohn's disease (CD) and to assess the impact of the corresponding DNA variants on clinical and disease phenotypes.METHODS: Ten variants in GR, TNF-a and IL-10 were genotyped in 113 childhood CD cases and 95 healthy subjects, both of French-Canadian origin.RESULTS: For the GR polymorphisms (R23K and N363S) and IL-10 variants in the 5'flanking region (-1082 G 〉 A, -819 T 〉 C and -592 A 〉 C), no difference was observed in allele and genotype frequencies between CD patients and controls. At the haplotype level, we found three IL-10 haplotypes previously described in Caucasians (GCC, ACC and ATA) and three novel haplotypes only present in IBD patients. When we analyzed the haplotype distribution with the anatomical location of the disease, the GCC haplotype was associated with the colonic and the ACC haplotype with the terminal ileum location, respectively. The genotyping of five polymorphisms in the promoter region of the TNF-α gene (-1031 T 〉 C, -863 A 〉 C, -857 T 〉 C, -308 A 〉 G and -238 A 〉 G) revealed a significant overrepresentation of homozygous -1031 CC among CD patients (OR = 9.9) and an association with the colonic location. For TNF-α, eleven haplotypes were inferred, including two frequent ones, TCCGG and CACGG, which were significantly observed more frequently in controls and cases, respectively.CONCLUSION: This is one of the first studies investigating the association between haplotype structure and disease location in a CD pediatric cohort. Our results will help to increase our understanding of the genetic determinants of childhood CD.展开更多

Solereder has been investigated to provide insight into the evolution and systematic position of Titanotrichum. In T. oldhamii, the size differentiation of the two cotyledons begins while the hypocotyl grows. However, both of the two cotyledons develop normally and locate at the same level. Finally, the two cotyledons are almost equal in size. The aerial shoot (including stem and leaves) is produced from the permanent activity of the apical meristem in the plumular bud. Even though the seedling development in Titanotrichum basically conforms to the general growth pattern of the seedling in the Cyrtandroideae, it is remarkably different from that of other Cyrtandroideae. Based on the revealed evidence in seedling development in Titanotrichum and other comparative data, the authors have evaluated the possible evolutionary pathway of Titanotrichum and further discussed the familial placement of this genus.展开更多
Biological soil crusts(BSCs) are the important components of alpine meadow ecosystems.The extent and morphology of BSCs vary greatly with alpine meadow retrogressive succession due to grazing pressure.There is signifi...Biological soil crusts(BSCs) are the important components of alpine meadow ecosystems.The extent and morphology of BSCs vary greatly with alpine meadow retrogressive succession due to grazing pressure.There is significant interest in impacts of crust composition on plant seed germination,especial l y in(semi-) arid environments.However,little is known about the influences of BSCs,and their associations with alpine meadow succession,on germination of typical alpine meadow vascular plant species.In a full factorial common-gardenexperiment,we studied effects of:(1) crust type,(2) seed position,and(3) surface texture on seed germination.We chose three typical alpine meadow plant species(i.e.Poa pratensis,Tibetia himalaica and Potentillen nivea),which belonged to different functional groups(graminoids,legumes,and forbs) and play important roles in all alpine meadow succession stages.Crust type and seed position influenced seed germination,and the inhibitory effects of BSCs depended on the crust type and seed species tested.The major factors influencing seed germination were BSC type,seed position,soil texture,and the interactions between BSC type and seed position; species and seed position; species andsurface texture; and species,crust type,and surface texture.Cyanobacteria crust significantly inhibited germination of all seeds.Seed position also had a significant effect on seed germination(p < 0.001).Fewer seedlings germinated on the surface than below the surface,this was especially true for P.nivea.seeds within cyanobacteria and lichen crusts.Only germination rates of T.himalaica on the soil surface were significantly correlated with plant occurrence frequency within the alpine meadow community.The poor correlation for the other two species is possibly that they are perennials.Our results clearly demonstrated that BSCs can be biological filters during seed germination,depending on the BSC succession stage.Through their influences on seed germination,BSCs can strongly influence community assemblages throughout alpine meadow retrogressive succession.展开更多

Rotor position estimation of a PMSM is performed by using sliding mode ob...This work proposes a new strategy to improve the rotor position estimation of a permanent magnet synchronous motor(PMSM) over wide speed range. Rotor position estimation of a PMSM is performed by using sliding mode observer(SMO). An adaptive observer gain was designed based on Lyapunov function and applied to solve the chattering problem caused by the discontinuous function of the SMO in the wide speed range. The cascade low-pass filter(LPF) with variable cut-off frequency was proposed to reduce the chattering problem and to attenuate the filtering capability of the SMO. In addition, the phase shift caused by the filter was counterbalanced by applying the variable phase delay compensation for the whole speed area. High accuracy estimation result of the rotor position was obtained in the experiment by applying the proposed estimation strategy.展开更多

For the DNA microarray analysis of formalin-fixed samples,we developed a simple and reproducible RNA extraction and linear amplification procedure applying two polymerasebinding sites. The amplification procedure took only 8 h and yielded comparable DNA microarray data between formalin-fixed tissues and unfixed controls.RESULTS: In comparison with normal pit, adenoma/carcinoma showed 504 up-regulated and 29 down-regulated genes at the expected false significance rate 0.15%. The differential expression between adenoma and carcinoma in situ was subtle: 50 and 22 genes were up-, and down-regulated in carcinomas at the expected false significance rate of 0.61%, respectively. Differentially expressed genes were grouped according to patterns of the sequential changes for the 'tendency analysis' in the gastric mucosaadenoma-carcinoma sequence.CONCLUSION: Groups of genes are shown to reflect the sequential expression changes in the early carcinogenic steps of stomach cancer. It is suggested that molecular carcinogenic pathways could be analyzed using routinely processed biopsies.展开更多

By machine l earning,we built a model and used it to test the effect of the test data set of true and pseudo splicing sites. Results: The prediction accuracy we obtained wa s 87.53% at the true 5' end splicing site and 87.37% at the true 3' end splicing sites. The results suggested that the SVM approach could achieve higher accuracy than the previous approaches.展开更多

These parameters consisted of radius, length and position of PMT as well as surface reflective ty...In this research, different parameters of plastic scintillator detector were investigated by Geant4 simulation toolkit. These parameters consisted of radius, length and position of PMT as well as surface reflective type and finish options. Furthermore, response time distributions of two organic plastic materials were studied. The results indicated that collecting optical photons has a linear relationship with PMT radius head. Also, the vertical location of PMT has a non-linear relationship with the optical photons collection. However, the collection decreased by increasing PMT length or moving PMT head horizontal position. The response functions of two plastic scintillator materials were in good agreement with experimental published results. Also, Geant4 radiation transport code can simulate incident radiation photon and predict subsequent events to the PMT head very well. The results indicated that BC-404 has faster scintillation properties versus BC-400 organic scintillator materials. Comparison between Geant4 outputs illustrates that the best reflector material and surface finish type for optical photons is ground TiO2.展开更多
