本研究旨在对慢性淋巴细胞性白血病原代骨髓基质细胞(CLL-human bone marrow stromal cells,CLL-hBMSC)和正常原代骨髓基质细胞(normal hBMSC,N-hBMSC)进行比较性分析,为建立CLL-CLL-hBMSC相互作用模型,从而更好地模拟CLL体内环境提供... 本研究旨在对慢性淋巴细胞性白血病原代骨髓基质细胞(CLL-human bone marrow stromal cells,CLL-hBMSC)和正常原代骨髓基质细胞(normal hBMSC,N-hBMSC)进行比较性分析,为建立CLL-CLL-hBMSC相互作用模型,从而更好地模拟CLL体内环境提供理论依据。从CLL患者和正常供者骨髓中分离单个核细胞,进行原代骨髓基质细胞(hBMSC)培养;采用实时荧光定量PCR(real-time PCR)检测CLL-hBMSC和N-hBMSC表面粘附分子如血管细胞粘附分子1(vascular cell adhesion molecule 1,VCAM-1)和细胞间粘附分子1(intercellular adhesion molecule 1,ICAM-1)的mRNA水平;采用real-time PCR和Western blot法分别检测两类不同来源的hBMSC中淋巴毒素β受体(lymphotoxin beta receptor,LTβR)的mRNA和蛋白质表达水平;采用Western blot法检测NF-κB家族成员的表达并分析细胞因子LTα1β2对NF-κB家族成员表达的影响;建立hBMSC和CLL细胞共培养体系,利用碘化丙啶(PI)染色检测细胞死亡情况。结果表明:从CLL患者骨髓中能成功地培养出贴壁的成纤维样hBMSC。与N-hBMSC相似,表面粘附分子VCAM-1和ICAM-1在两类hBMSC中的mRNA表达一致;两类hBMSC中LTβR的mRNA和蛋白表达水平相似;各NF-κB家族成员在两类hBMSC中都有表达,且蛋白水平无显著性差异;细胞因子LTα1β2可诱导hBMSC中经典性和非经典性NF-κB信号通路的活化;体外实验表明,两类hBMSC都能够有效地支持CLL细胞的体外存活。结论:两类hBMSC培养效率以及细胞形态上无明显差异。LTβR-NF-κB信号分子在两类hBMSC的表达以及活化情况相似。 展开更多

Nowadays it is one of the most famous archaeological sites in the world. It was declared a world heritage site by UNESCO (United Nations Educational, Scientific, and Cultural Organization) in 1985. The historical and the cultural values of the city are briefly elaborated. The locality plans to the culture site protections are introduced, especially the restoration of the Great Temple, which gives us an opportunity to comprehend the protection of ancient cities in different countries. The Jordanian government has taken very seriously their commitment to preserving this valuable historical site, which is considered by many Jordanians to be a priceless national treasure. One would assume that a gradual reversal, from neglect to serious efforts of preserving the site and its beauty, would have taken place. 展开更多

Cryopreservation of Veliger Larvae of Trumpet Shell, Charonia sauliae: an Essential Preparation to Artificial Propagation 被引量:1

Trumpet shell, Charonia sauliae, is an endangered and valuable species, but its artificial propagation protocol has not been successfully established. To estimate the possibility of cryopreservation for larvae of C. sauliae, which is a potential preparation for its artificial reproduction and further research, in this study a protocoi for the cryopreservation of veliger larvae of trumpet shell was optimized. Through a two-step cryopreservation procedure, four kinds of cryoprotectants (ethylene glycol, 1, 2-propanediol, dimethyl sulfoxide and glycerol) were employed at three concentrations (1.0, 1.5 and 2.0molL^-1) respectively and survival rates of larvae were determined after a storage of lh. The larvae frozen with these four cryoprotectants after 1 h storage were cultured, and then survival rates were determined at 24, 72 and 120 h after thawing. Dimethyl sulfoxide at a concentration of 1.5 molL^-1 showed the best protective effect in all experiments (p〈0.05). And survival rates of larvae frozen with dimethyl sulfoxide were determined after 1, 7 and 15 d of storage. The survival rates of larvae frozen with 1.5 molL^-1 dimethyl sulfoxide after 1 h, 1 d, 7 d and 15 d of storage were 80.77% ±7.51%, 80.34%±11.28%, 83.10%±9.14% and 77.23%±6.22% respectively. No significant differences in survival rates of larvae frozen with dimethyl sulfoxide were observed after various storage periods (p〉0.05). 展开更多

Although fluplrtme has been used to relieve pain caused by different diseases and clinical procedures, information on the safety and efficacy of flupirtine is limited. "fhe present study was conducted to investigate the neuroprotective effects of flupirtine on U373 malignant glioma (MG) cell lines. Methods: Cellviability and cell cycle analysis was performed by MTF assay and flow cytomet-,'y, respectively. Results: Variations in the growth of U373 MG cells in $ mM N-methyl-D-aspartate (NMDA), 1 mM flupirtine, and combined treatment indicated the antagonistic effects of NMDA and flupirtine on MG cell lines. The variation in the percentage of gated cellpopulation in different cell cycle phases showed significant variations after 48 h of treatment. Conclusion: Flupirtine has neuroprotective effect of on U373 MG cells, which limits its use in the pain management of brain tumors. This property warrants further studies using animal models and large-scale clinical trials. 展开更多
