A gradient HPLC method was established for the determination of harpagoside and cinnamic acid in Radix Scrophulariae (Xuanshen) and a proposition was put forward for the lowest content of the characteristic and active constituent (harpagoside) for qualified Radix Scrophulariae. The experimental conditions were as follows: Ultrasphere ODS column (250 mm 4.6 mm, 5 mm), mobile phase: acetonitrile-water (containing 1.0% acetic acid) (20:8050:50) (20 min), flow-rate 1 mLmin-1, room temperature, detection wavelength 278 nm. Twenty-eight samples of Xuan-shen (Radix Scrophulariae) from different districts of China were analyzed and the contents of harpagoside and cinnamic acid in Xuan-shen were 0.041~0.244% and 0.012~0.068% respectively. The recoveries (RSD)% were 97.13(0.80)% for harpagoside and 99.38(0.51)% for cinnamic acid. The method is simple and accurate. It can be used for the quality control of Radix Scrophulariae. We propose that the content of harpagoside in qualified Radix Scrophularia should be no less than 0.05%. 展开更多

[Objective] This study aimed to establish a method for quantitative detection of mRNA transcriptional level of SS2 adhesive related-factors of Streptococcus suis serotype 2 (SS2) by fluorescent quantitative PCR. []Vlethod] The gene fragments en- coding SS2 adhesive related-factors MRP, FBPS and CPS2J and a housekeeping gene aroA were amplified by reverse transcription PCR from the total RNA of SS2, cloned, and sequenced. The recombinant plasmids containing the target genes were constructed, and used as templates in Real-time PCR. [Result] Dynamic curves, stan- dard curves and melting curves of the adhesive related-factors and aroA were ob- tained by the optimized Real-time PCR system. The standard curves showed a good linear relationship between template copy number and circulation number, and the correlation coefficients (FF) of the standard curves were over 0.995. Also, these as- says were highly specific a^d there was single specific melting peak for every gene. Moreover, the assays were highly sensitive and had a detection limit of 1.0×102 copies in 1 μl of initial templates. Finally, it was highly repeatable and had a coeffi- cient of variation less than 2% for intra-assay. [Conclusion] This study will provide a way to reveal the adhesion mechanism of SS2 to different host cells at molecular level. 展开更多

This study describes the development and validation of a sensitive and reliable method for determination of polybrominated diphenyl ethers(PBDEs)in atmospheric particulate matter using selective pressurized liquid extraction(SPLE)and gas chromatography–mass spectrometry with a negative chemical ionization(GC-NCI-MS).Extraction and clean-up were performed using PLE with 2 g florisil and 3 g silica placed in the extraction cells.Under optimal conditions,14 PBDEs were extracted at 70℃ using hexane/dichloromethane(50:50,v/v)as solvent.Validation of SPLE returned excellent recoveries for most analytes,with relative standard deviations mostly below20%.Method detection limits ranged from 0.13 to15.38 ng·mL^-1 for the GC-MS analyses.The method was successfully applied to atmospheric particulate matter of Beijing,where analytes were detected in the range of182.79 to 468.99 pg·m^-3. 展开更多

With four packed chromatographic columns, which are Hayesep-Q pre-column, Hayesep-Q column, molecular sieve 5A column and one PLOT A1203 S capillary column, the gas compositions for direct coal liquefaction are analyzed qualitatively and quantitatively by the external standard method. This study examined levels of polycyclic aromatic hydrocarbons (PAHs) in estuarine sediments in Licun (Qingdao, China) by gas chromatography under optimized conditions for sample pretreatment via ultrasonic extraction, column chromatography, and thin layer chromatography. Methanol and dichloromethane (DCM)/methanol (2:1, v/v) were used in ultrasonic extraction, and DCM was used as eluate for column chromatography. The developing system consisted of n-hexane and DCM at a ratio of 9:1 (v/v), with DCM as the extraction solvent for PAHs-containing silica gel scraped off the plate. When the spiking level is 100 ng, total recoveries of spiked matrices for four target PAHs (phenanthrene, anthracene, pyrene and chrysene) were 83.7%, 76.4%, 85.8%, and 88.7%, respectively, with relative standard deviation (RSD) between 5.0% and 6.5% (n = 4). When the spiking level is 1000 ng, associated total recoveries were 78.6%, 72.7%, 82.7% and 85.3%, respectively, with RSD between 4.4% and 5.3% (n = 4). The opti-mized method was advantageous for determination of PAHs in complex matrix due to its effective sample purification. 展开更多

Using the reference hypernetted chain (RHNC) integral equation theory and a rigorous stability analysis method, we investigate the phase behavior of a mixture of hard-sphere dipoles and neutral hard spheres based on the correlations of the homogeneous isotropic phase. Lowering the temperature down to the points where the RHNC equations fail to have a solution, several ttuctuations strongly increase. At low densities our results indicate the onset of / chain formation, which is similar with the pure DHS system. At high densities, the results indicate the appearance of isotropic-to-ferroelectric transitions (small neutral hard spheres concentrations) and demixing transitions (large neutral hard spheres concentrations). 展开更多

Based on qualitative methods with participative experts and stakeholders, the strategic prospective allows designing different future scenarios and planning the transformation of a current situation into a desired future. To design sustainable futures, this paper proposes a four-stage methodology: understanding the context; strategy visualization; design of alternative scenarios and definition of desired future; and planning its construction. This methodology makes evident the possibility of seeking alternative scenarios for a sustainable future in different scopes, particularly, in two prospective applications: biodiversity conservation and water treatment. The outcomes of the application have allowed proposing strategies and policies for the management of environmental goods, such as environmental services and water reuse, with a long-time outlook based on a collective desired future. 展开更多

In order to study the failure process of an anchorage structure and the evolution law of the body's defor- mation field, anchor push-out tests were carried out based on digital speckle correlation methods (DSCM). The stress distribution of the anchorage interface was investigated using the particle flow numerical simulation method. The results indicate that there are three stages in the deformation and fail- ure process of an anchorage structure: elastic bonding stage, a de-bonding stage and a failure stage. The stress distribution in the interface controls the stability of the structure. In the elastic bonding stage, the shear stress peak point of the interface is close to the loading end, and the displacement field gradually develops into a "V" shape, in the de-bonding stage, there is a shear stress plateau in the center of the anchorage section, and shear strain localization begins to form in the deformation field. In the failure stage, the bonding of the interface fails rapidly and the shear stress peak point moves to the anchorage free end. The anchorage structure moves integrally along the macro-cracl~ The de-bonding stage is a research focus in the deformation and failure process of an anchorage structure, and plays an important guiding role in roadway support design and prediction of the stability of the surrounding rock. 展开更多

A new mathematical method is proposed to convert the oscillator instability parameters from Allan variance to Spectrum Density(SD)of random phase fluctuations,which is the inversion of the classic transformation formula from SD to Allan variance.Due to the fact that Allan variance does not always determine a unique SD function,power-law model of the SD of oscillator phase fluctuations is introduced to the translating algorithm and a constrained maximum likelihood solution is presented.Considering that the inversion is an ill-posed problem,a regularization method is brought forward in the process.Simulation results show that the converted SD of phase fluctuations from Allan variance parameters agrees well with the real SD function.Furthermore,the effects of the selected regularization factors and the input Allan variances are analyzed in detail. 展开更多

The modeling of volatility and correlation is important in order to calculate hedge ratios, value at risk estimates, CAPM (Capital Asset Pricing Model betas), derivate pricing and risk management in general. Recent access to intra-daily high-frequency data for two of the most liquid contracts at the Nord Pool exchange has made it possible to apply new and promising methods for analyzing volatility and correlation. The concepts of realized volatility and realized correlation are applied, and this study statistically describes the distribution (both distributional properties and temporal dependencies) of electricity forward data from 2005 to 2009. The main findings show that the logarithmic realized volatility is approximately normally distributed, while realized correlation seems not to be. Further, realized volatility and realized correlation have a long-memory feature. There also seems to be a high correlation between realized correlation and volatilities and positive relations between trading volume and realized volatility and between trading volume and realized correlation. These results are to a large extent consistent with earlier studies of stylized facts of other financial and commodity markets. 展开更多
