Previously studies showed that FXR-/- mice spontaneously developed liver tumors when they aged, however, the relevance of which to human hepatocellular carcinoma (HCC) is unclear. The aim of this study is to observe whether FXR expression is also downregulated in HCC and discuss the mechanism of the reduced FXR expression in HCC. Expression of FXR and small heterodimer partner (SHP) was measured by real-time PCR and immunohistochemical technique. Effect of pro-inflammatory cytokines on expression of FXR and its promoter activity were determined in primary hepatocytes or HepG2 and Huh7 cell lines. Our results showed that expression of FXR and its target gene SHP in human HCC was strongly downregulated compared to the normal liver tissues. In addition, pro-inflammatory cytokines were able to decrease FXR expression by inhibiting the FXR promoter activity. In conclusion this work demonstrates FXR expression is strongly downregulated in human HCC, which may be caused by decreased FXR promoter activity, suggesting a potential role of FXR in human HCC development.展开更多

The new implicit numerical integration technique of the authors, which is based on a simple approximation of the linear system, has great advantages over the existing implicit integration methods. Moreover, experiment shows that the new technique is highly efficient in animating a kind of non-rigid objects, and suitable for the draping module of the 3D garment CAD system.展开更多

The software takes charge of the functions and the management of the digital system. Nowadays railway equipment has begun to be designed and implemented based on digital technology with online and real time performance to achieve high speed and high density operation. The software is designed, coded and implemented into the system, and controls hardware components in order to satisfy the functional requirements of the system. As the functions that are implemented by software are increased, it becomes very difficult to predict the soil-ware failures because of its complexity. Therefore, it has become one of the key issues how to assure that the sottware is reliable, safe and secure. In this paper, the safety criteria are suggested to secure the software safety for the field of railway system. Characteristic of railway software and review safety related standards are analyzed. The authors also suggest development methodology and certification procedure for the developer and assessor to easily make safety critical railway software with following the safety criteria.展开更多

The putative species in...Species included in the Sporothrix schenckii complex are temperature-dependent with dimorphic growth and cause sporotrichosis that is characterized by chronic and fatal lymphocutaneous lesions. The putative species included in the Sporothrix complex are S. brasiliensis, S. globosa, S. mexicana, S. pallida, S. schenckii, and S. lurei. S. globosa is the causal agent of sporotrichosis in China, and its pathogenicity appears to be closely related to the di- morphic transition, i.e. from the mycelial to the yeast phase, it adapts to changing environmental conditions. To determine the molecular mechanisms of the switching process that mediates the dimorphic transition of S. globosa, suppression subtractive hybridization (SSH) was used to prepare a complementary DNA (cDNA) subtraction library from the yeast and mycelial phases. Bioinformatics analysis was performed to profile the relationship between dif- ferently expressed genes and the dimorphic transition. Two genes that were expressed at higher levels by the yeast form were selected, and their differential expression levels were verified using a quantitative real-time reverse transcriptase polymerase chain reaction (qRT-PCR). It is believed that these differently expressed genes are involved in the pathogenesis of S. globosa infection in China.展开更多
