为找出发根农杆菌诱导的决明毛状根与其无菌实生根的形态和解剖特征差异,采用体视显微观察、石蜡切片,番红-固绿染色及多糖-高碘酸-席夫反应染色,显微观察比较决明毛状根与实生根的形态、解剖结构及淀粉粒分布.结果发现,决明毛状根粗细... 为找出发根农杆菌诱导的决明毛状根与其无菌实生根的形态和解剖特征差异,采用体视显微观察、石蜡切片,番红-固绿染色及多糖-高碘酸-席夫反应染色,显微观察比较决明毛状根与实生根的形态、解剖结构及淀粉粒分布.结果发现,决明毛状根粗细均匀,根尖较实生根粗壮,有较多根毛;毛状根较小的表皮细胞和皮层细胞排列紧密而不规则;内皮层细胞排列紧密,细胞间隙小,凯氏带不明显;毛状根初生木质部为二原型,而实生根为四原型;毛状根形成层细胞较实生根细胞小、排列紧密、层数多;毛状根根冠几乎没有淀粉粒,而实生根有较多淀粉粒.这些结果表明,决明毛状根与实生根在外部形态和解剖结构上均存在较大差异. 展开更多

以白肉火龙果的实生根为制片材料,在去壁低渗法的基础上改进染色体制片技术,并进行核型分析。结果表明,以白肉火龙果种子在粗砂砾中萌发的实生根为材料,以等体积的0.2%秋水仙素与0.002mol/L8-羟基喹啉预处理1h,在1mol/L HCl... 以白肉火龙果的实生根为制片材料,在去壁低渗法的基础上改进染色体制片技术,并进行核型分析。结果表明,以白肉火龙果种子在粗砂砾中萌发的实生根为材料,以等体积的0.2%秋水仙素与0.002mol/L8-羟基喹啉预处理1h,在1mol/L HCl中60℃下解离13min,制片效果最好。核型分析显示白肉火龙果的染色体数目为2n=2x=22,全部染色体都为中部染色体,核型公式为2n=2x=22=22m;染色体的相对长度变化范围为6.95%~12.97%,核型为1A型,核不对称系数为52.67%。 展开更多

The nutrient solution with three pH levels, Le., 5.0, 7.0 and 8.5 were applied to culture five combination seedlings, including A (RB85-5156 × YR05-407), B (YR06-3501 ×Tolodo), C (YT94-128 × GZ75-65), D (ROC6 × YR05-733) and E (VMC87-95 ×YR05-171). The result indicated that root length, root surface, root volume and root diameter were all in the largest in B combination with solution pH= 7.0, and the root length, root surface, root volume were in the largest in combination A, C, D and E with solution pH=8.5. Root activity in combination C, D and E were in the highest with solution pH=5.0, while it was in the highest for combination B when pH=7.0. In general, the root growth and morphology in all five combinations were better in the solutions with pH of 7.0 and 8.5. 展开更多

A pot experiment was conducted to study the plant growth and fruit yields of cucumber (Cucumis sativus L.) on a greenhouse soil with or without inoculation of arbuscular mycorrhizal fungi (AMFs) and Fusarium oxysporum f. sp, cucumerinum under unsterilized conditions. Two AMF inocula were tested: only one AMF strain Glomus caledonium 90036 and an AMF consortium mainly consisting of Glornus spp. and Acaulospora spp. There were four treatments including no inoculation (control), inoculation with F. oxysporum but without mycorrhizae (FO), inoculation with F. oxysporum and G. caledonium (FO+M1), and inoculation with F. oxysporum and the AMF consortium (FO+M2). Cucumber plants were harvested at weeks 3 and 9 after transplanting. Compared with the control, the FO treatment without AMF inoculation had less biomass both at weeks 3 and 9 (P 〈 0.05) and had higher incidence of Fusarium wilt and produced no cucumber fruit at week 9. Both FO+M1 and FO+M2 treatments had higher mycorrhizal colonization than the treatments which received no AMF inoculation at week 3 (P 〈 0.05), but only the FO+M2 treatment elevated plant biomass, decreased the incidence of Fusarium wilt, and improved cucumber yields to the same level as the control at week 9. The results indicated that the AMF consortium could suppress Fusarium wilt of cucumber and, therefore, showed potential as a biological control agent in greenhouse agroecosystems. 展开更多
