Methods Scutellarin was given to dogs by intravenous injection and determined by RP-HPLC, the mean plasma concentration-time curve was plotted and pharmacokinetic parameters were calculated by program 3p87. Resu;ts The concentration-time curve of scutellarin could be fitted to three-compartment model with T1/2 pi, T1/2 α and T1/2 β being 1.05 ± 0.80 min, 6.99 + 2.76 min and 51.61 + 28.78 min, respectively, Vc being 880.1 + 508.3 mL, CL being 189.6 + 53.8 mL@ min- 1, and AUC0-90 and AUC0-∞ being 574.43 + 133.95 μg@ min@ mL - 1 and 599.34 ± 132.00μg@ min@mL- 1, respectively. Conclusion The fact that the concentrations of scutellarin in plasma declined rapidly after the medication suggested that the T1/2 of scutellarin should be taken into account in drug administration and preparation development.展开更多

In order to investigate the effect of water content on the energy evolution of red sandstone, the axial loading–unloading experiments on dry and water-saturated sandstone samples were conducted, and the distribution ...In order to investigate the effect of water content on the energy evolution of red sandstone, the axial loading–unloading experiments on dry and water-saturated sandstone samples were conducted, and the distribution and evolution of elastic energy and dissipated energy within the rock were measured.The results show that the saturation process from dry to fully-saturated states reduces the strength, rigidity and brittleness of the rock by 30.2%, 25.5% and 16.7%, respectively. The water-saturated sample has larger irreversible deformation in the pre-peak stage and smaller stress drop in the post-peak stage.The saturation process decreases the accumulation energy limit by 38.9%, but increases the dissipated energy and residual elastic energy density, thus greatly reducing the magnitude and rate of energy release. The water-saturated sample has lower conversion efficiency to elastic energy by 3% in the prepeak region; moreover, the elastic energy ratio falls with a smaller range in the post-peak stage.Therefore, saturation process can greatly reduce the risk of dynamic disaster, and heterogeneous water content can lead to dynamic disaster possibly on the other hand.展开更多

METHODS:Establishment of continuous hypothermic machine perfusion model,detection of nucleotides in hepatocytes with HPLC,measurement of activities of LDH and AST in the perfusate,observation of histopathological changes in different experiment groups,and autoradiography were carried out to reveal the underlying mechanism of the protective effect of ATP. RESULTS:The intracellular levels of ATP and EC decreased rapidly after hypothermic preservation in control group,while a higher ATP and EC level,and a slower decreasing rate were observed when ATP-MgCl_2 was added to the perfusate (P<0.01).As compared with the control group,the activities of LDH and AST in the ATP-MgCl_2 group were lower(P<0.05). Furthermore,more severe hepatocyte damage and neutrophil infiltration were observed in the control group.Radioactive [α-^(32)P]ATP entered the hypothermically preserved rat hepatocytes. CONCLUSION:Exogenous ATP has a protective effect on rat livers during hypothermical preservation.However,Mg^(2+) is indispensable,addition of ATP alone produces no protective effect.The underlying mechanism may be that exogenous ATP enters the hypothermically preserved rat liver cells.展开更多
Yamanaka factors (Oct3/4, Sox2, Klf4, c-Myc) are highly expressed in embryonic stem (ES) cells, and their overexpression can induce pluripotency in both mouse and human somatic cells, indicating that these factors...Yamanaka factors (Oct3/4, Sox2, Klf4, c-Myc) are highly expressed in embryonic stem (ES) cells, and their overexpression can induce pluripotency in both mouse and human somatic cells, indicating that these factors regulate the developmental signaling network necessary for ES cell pluripotency. However, systemic analysis of the signaling pathways regulated by Yamanaka factors has not yet been fully described. In this study, we identified the target promoters of endogenous Yamanaka factors on a whole genome scale using ChIP (chromatin immunoprecipitation)- on-chip in El4.1 mouse ES cells, and we found that these four factors co-occupied 58 promoters. Interestingly, when Oct4 and Sox2 were analyzed as core factors, Klf4 functioned to enhance the core factors for development regulation, whereas c-Myc seemed to play a distinct role in regulating metabolism. The pathway analysis revealed that Yamanaka factors collectively regulate a developmental signaling network composed of 16 developmental signaling pathways, nine of which represent earlier unknown pathways in ES cells, including apoptosis and cellcycle pathways. We further analyzed data from a recent study examining Yamanaka factors in mouse ES ceils. Interestingly, this analysis also revealed 16 developmental signaling pathways, of which 14 pathways overlap with the ones revealed by this study, despite that the target genes and the signaling pathways regulated by each individual Yamanaka factor differ significantly between these two datasets. We suggest that Yamanaka factors critically regulate a developmental signaling network composed of approximately a dozen crucial developmental signaling pathways to maintain the pluripotency of ES cells and probably also to induce pluripotent stem cells.展开更多

To study the contaminant transport in the reclamation districts, robust and accurate estimation of dispersion coefficient is essential. In this study, the continuous sodium chloride (NaC1) solution with constant concentration was introduced into the column filled with the dredger fill to get the breakthrough curves (BTCs). Inverse error function method (IEFM) and CXTFIT program were used for estimating dispersion coefficient. Results showed that the difference between the dispersion coefficients estimated by IEFM and CXTFIT program was slight. The main reason was that the BTC was not strictly linear. IEFM performed poorly in the nonlinear area, while the CXTFIT performed well over the entire BTCs. Moreover, the dispersion coefficient of dredger fill was small. The dispersion property of dredger fill would result in the slow migration of contaminants in the dredger fill.展开更多

However, controversial results have been reported regarding the exact role of 20-HETE on blood pressure. In the present study, we used recombinant adenoassociated viral vector (rAAV) to deliver CYP 4A1 cDNA and antisense 4A1 cDNA into Sprague-Dawley (SD) rats and spontaneously hypertensive rats (SHR), respectively, to investigate the effects of long-term modifications of blood pressure and the potential for gene therapy of hyperténsion. The mean systolic pressure increased by 14.2±2.5 mm Hg in rAAV.4A 1-treated SD rats and decreased by 13.7±2.2 mm Hg in rAAV.anti4A l-treated SHR rats 5 weeks after the injection compared with controls and these changes in blood pressure were maintained until the experiments ended at 24 weeks. In 4A1 treated animals CYP4A was overexpressed in various tissues, but preferentially in the kidney at both mRNA and protein levels. In anti-4Al-treated SHR, CYP4A mRNA in various tissues was probed, especially in kidneys, but 4A l protein expression was almost completely inhibited. These results suggest that arachidonic acid CYP hydroxylases contribute not only to the maintenance of normal blood pressure but also to the development of hypertension. rAAV-mediated anti4A administration strategy has the potential to be used as targeted gene therapy in human hypertension by blocking expression of CYP 4A in kidneys.展开更多

We have prepared a polyclonal anti-mo...EKLF is an erythroid-specific, zinc finger-containing transcription factor essential for the activation of the mammalian beta globin gene in erythroid cells of definitive lineage. We have prepared a polyclonal anti-mouse EKLF antibody suitable for Western blotting and immunoprecipitation (IP) qualities, and used it to define the expression patterns of the EKLF protein during mouse erythroid development. We have also used this antibody for the chromatin-immunoprecipitation (CHIP) assay. EKLF was found to bind in vivo at both the mouse beta-major-globin promoter and the HS2 site of beta-LCR in the mouse erythroleukemia cells (MEL) in a DMSO-inducible manner. The DMSO-induced bindings of EKLF as well as three other proteins, namely, RNA polymerase Ⅱ, acetylated histone H3, and methylated histone H3, were not abolished but significantly lowered in CB3, a MEL-derived cell line with null-expression of p45/NF-E2, an erythroid-enriched factor needed for activation of the mammalian globin loci. Interestingly, binding of EKLF in vivo was also detected in the mouse alpha-like globin locus, at the adult alpha globin promoter and its far upstream regulatory element alpha-MRE (HS26). This study provides direct evidence for EKLF-binding in vivo at the major regulatory elements of the mouse beta-like globin gene clusters the data also have interesting implications with respect to the role of EKLF-chromatin interaction in mammalian globin gene regulation.展开更多

Positive clones were screened by incubating in the selective medium with 600 μg/mL G418 and named HL-7702/HBV-encoded X protein (HBx) cells. The expressions of Fas/FasL, Bax/Bcl-2, and c-myc mRNA were measured by semi-quantitative RT-PCR in HL-7702/HBx and control group, respectively.RESULTS: RT-PCR analysis confirmed that HBV X gene was transfected into HL-7702 cells successfully. By semiquantitative RT-PCR analysis, Bax and c-myc mRNA levels in HL-7702/HBx cells of transient transfection were significantly higher than those in control, FasL and c-myc mRNA levels in HL-7702/HBx cells of stable transfection were significantly higher than those in control, whereas the Bcl-2 mRNA levels in HL-7702/HBx cells of transient and stable transfection were significantly lower than thosein control.CONCLUSION: HBV X gene may promote the apoptosis of hepatocytes by regulating the expressions of Fas/FasL, Bax/Bcl-2, and c-myc gene in a dose-dependent manner.展开更多
An increase in public environmental awareness and pressures from the government’s determination to deal with environmental problems force manufacturers to implement green manufacturing(GM).However,since managers and ...An increase in public environmental awareness and pressures from the government’s determination to deal with environmental problems force manufacturers to implement green manufacturing(GM).However,since managers and stakeholders lack understanding of GM and its complexity,the manufacturing enterprise,especially the small and medium ones,are constantly facing the problems of how to make a reasonable decision for an environmental problem,and the adopted approaches have no clear payoffs.The customized design,flexible production,and diversified services in customized production make the problems more challenging.In order to solve this problem,this paper proposes a framework for the implementation of GM in customized products manufacturing enterprises(CPME).A three-layer framework,i.e.,the goal layer,the product life-cycle layer and the supporting layer,is designed to provide a methodology to help implement GM.In this model,the GM practice processes are divided into four stages from the life-cycle perspective,i.e.design,production,use,and disposal.The preliminary practice of GM in an electrical product manufacturing company is carried out,and the implementation effect shows that the system framework is helpful to make a comprehensive understanding of GM and to improve the operability of GM practices.The integrated product model is an effective way to integrate the life cycle data.展开更多

The expressions of Fas/FasL, Bax/Bcl-2, and c-myc mRNA were measured by semi-quantitative RT-PCR in HL-7702/HBx and control group, respectively.RESULTS: RT-PCR analysis confirmed that HBV X gene was transfected into HL-7702 cells successfully. By semiquantitative RT-PCR analysis, Bax and c-myc mRNA levels in HL-7702/HBx cells of transient transfection were significantly higher than those in control, FasL and c-myc mRNA levels in HL-7702/HBx cells of stable transfection were significantly higher than those in control, whereas the Bcl-2 mRNA levels in HL-7702/HBx cells of transient and stable transfection were significantly lower than thosein control.CONCLUSION: HBV X gene may promote the apoptosis of hepatocytes by regulating the expressions of Fas/FasL, Bax/Bcl-2, and c-myc gene in a dose-dependent manner.

文摘An increase in public environmental awareness and pressures from the government’s determination to deal with environmental problems force manufacturers to implement green manufacturing(GM).However,since managers and stakeholders lack understanding of GM and its complexity,the manufacturing enterprise,especially the small and medium ones,are constantly facing the problems of how to make a reasonable decision for an environmental problem,and the adopted approaches have no clear payoffs.The customized design,flexible production,and diversified services in customized production make the problems more challenging.In order to solve this problem,this paper proposes a framework for the implementation of GM in customized products manufacturing enterprises(CPME).A three-layer framework,i.e.,the goal layer,the product life-cycle layer and the supporting layer,is designed to provide a methodology to help implement GM.In this model,the GM practice processes are divided into four stages from the life-cycle perspective,i.e.design,production,use,and disposal.The preliminary practice of GM in an electrical product manufacturing company is carried out,and the implementation effect shows that the system framework is helpful to make a comprehensive understanding of GM and to improve the operability of GM practices.The integrated product model is an effective way to integrate the life cycle data.

文摘In this paper two kinds of two-boson realizations of generally deformed algebras with three generators are obtained by generalizing the Jordan-Schwlnger realizations of SU(2) and SU(1,1). For each kind, a unitary realization and a non-unitary realizatlon, together with the properties of their acting spaces, are discussed. Similarity transformations that relate the non-unitary realizations to the unitary ones are given by solving unitarization equations.
