AIM:To explore the possibility of the replacement of the gag gene between human immunodeficiency virus and bovine immunodeficiency virus, to achieve chimeric virions, and thereby gain a new kind of AIDS vaccine based on BHIV chimeric viruses. METHODS: A series of chimeric BHIV proviral DNAs differing in the replacement regions in gag gene were constructed, and then were transfected into 293T cells. The expression of chimeric viral genes was detected at the RNA and protein level. The supernatant of 293T cell was ultra centrifuged to detect the probable chimeric virion. Once the chimeric virion was detected, its biological activities were also assayed by infecting HIV-sensitive MT4 cells. RESULTS: Four chimeric BHIV proviral DNAs were constructed. Genes in chimeric viruses expressed correctly in transfected 293T cells. All four constructs assembled chimeric virions with different degrees of efficiency. These virions had complete structures common to retroviruses and packaged genomic RNAs, but the cleavages of the precursor Gag proteins were abnormal to some extent. Three of these virions tested could attach and enter into MT4 cells, and one of them could complete the course of reverse transcription. Yet none of them could replicate in MT4 cells. CONCLUSION: The replacement of partial gag gene of HIV with BIV gag gene is feasible. Genes in chimeric BHIVs are accurately expressed, and virions are assembled. These chimeric BHIVs (proviral DNA together with virus particles) have the potential to become a new kind of HIV/AIDS vaccine. 展开更多

The defects of FRP pipelines would increase with the extension of service time. However, it is very difficult to detect the defects of FRP pipelines on-spot quickly. In this paper, a new method detecting defects for FRP pipes has been provided based on the NMR. The proton density distributions have been obtained at different depth of FRP components using single-side NMR. The experimental results show that there is a significant change of proton density distribution at the location of defects. And, these results would be useful for defects inspection of composite material component. 展开更多

Repairing articular cartilage defects with tissue-engineering cartilage in rabbits 被引量:15

Objective : To investigate the effect of cancellous bone matrix gelatin ( BMG ) engineered with allogeneic chondrocytes in repairing articular cartilage defects in rabbits. Methods: Chondrocytes were seeded onto threedimensional cancellous BMG and .cultured in vitro for 12 days to prepare BMG-chondrocyte complexes. Under anesthesia with 2.5% pentobarbital sodium ( lml/kg body weight), articular cartilage defects were made on the right knee joints of 38 healthy New Zealand white rabbits (regardless of sex, aged 4-5 months and weighing 2. 5-3 kg) and the defects were then treated with 2.5% trypsin. Then BMG-chondrocyte complex (Group A, n = 18), BMG (Group B, n =10), and nothing (Group C, n =10) were implanted into the cartilage defects, respectively. The repairing effects were assessed by macroscopic, histologic, transmission electron microscopic (TEM) observation, immunohistochemical examination and in situ hybridization detection, respectively, at 2, 4, 8, 12 and 24 weeks after operation. Results.. Cancellous BMG was degraded within 8 weeks after operation. In Group A, lymphocyte infiltration was observed around the graft. At 24 weeks after operation, the cartilage defects were repaired by cartilagetissues and the articular cartilage and subchondral bone were soundly healed. Proteoglycan and type II collagen were detected in the matrix of the repaired tissues by Safranin-O staining and immunohistochemical staining, respectively. In situ hybridization proved gene expression of type II collagen in the cytoplasm of chondrocytes in the repaired tissues, TEM observation showed that chondrocytes and cartilage matrix in repaired tissues were almost same as those in the normal articular cartilage. In Group B, the defects were repaired by cartilage-fibrous tissues. In Group C, the defects were repaired only by fibrous tissues. Conclusions : Cancellous BMG can be regarded as the natural cell scaffolds for cartilage tissue engineering. Articular cartilage defects can be repaired by cancellous BMG engineered with allogeneic chondrocytes. The nature of repaired tissues is closest to the normal cartilage. Local administration of trypsin can promote the adherence of repaired tissues to host tissues. Transplantation of allogeneic chondrocytes has immunogenicity, but the immune reaction is weak. 展开更多

A knock-down factor is commonly used to take into account the obvious decline of the buckling load in a cylindrical shell caused by the inevitable imperfections. In 1968, NASA guideline SP-8007 gave knock-down factors which rely on a lower-bound curve taken from experimental data. Recent research has indicated that the NASA knock-down factors are inclined to produce very conservative estimations for the buckling load of imperfect shells, due to the limitations of the computational power and the experimental skills available five decades ago. A novel knock-down factor is proposed composed of two parts for the metallic stiffened cylinders. A deterministic study is applied to achieve the first part of the knock-down factor considering the measured geometric imperfection, the other types of imperfections are considered in the second part using a stochastic analysis. A smeared model is used to achieve the implementation of the measured geometric imperfection for the stiffened cylinder. This new robust and less conservative design for the stiffened cylinders is validated by using test results. 展开更多
