The genomic fusions of the anaplastic lymphoma kinase(ALK)gene have been widely recognized as effective therapeutic targets for non-small cell lung carcinoma(NSCLC).The Second Xiangya Hospital of Central South Univers...The genomic fusions of the anaplastic lymphoma kinase(ALK)gene have been widely recognized as effective therapeutic targets for non-small cell lung carcinoma(NSCLC).The Second Xiangya Hospital of Central South University has treated 2 NSCLC patients with 2 distinct novel ALK gene fusions.Case 1 was a 55-year-old male with a solid nodule located in the right hilar lobe on enhanced CT scan.Case 2 was a 47-year-old female with enhanced CT showing involvement of the left upper lobe of lung.Histopathological examination of tumor tissues confirmed lung adenocarcinoma in both cases.Immunohistochemical(IHC)staining demonstrated positivity for thyroid transcription factor 1(TTF-1)and ALK-D5F3 in tumor cells,while negativity for P40.The next-generation sequencing(NGS)tests identified a PNPT1-ALK(Exon22:Exon20)fusion variant in case 1 and a TCEAL2-ALK(Exon3:Exon19)fusion variant in case 2.The TCEAL2-ALK fusion was further confirmed by amplification refractory mutation system(ARMS)-PCR at the mRNA level.Both patients were treated with oral alectinib at a dosage of 600 mg twice daily.The tumors in both patients were significantly decreased after alectinib treatment,achieving partial response.At the time of submission,there was an absence of disease progression and the progression-free survival(PFS)had surpassed 1 year.It offered compelling evidences that the individuals with NSCLC and harboring either a PNPT1-ALK(Exon22:Exon20)fusion or a TCEAL2-ALK(Exon3:Exon19)fusion,experience favorable therapeutic outcomes through the administration of alectinib.This study expands the known ALK fusion variants database and supports the precision treatment of NSCLC using ALK tyrosine kinase inhibitors(TKIs).展开更多

This study was undertaken to characterize the changes of endogenous hormone levels during floret development in three wheat ( Triticum aestivum L.) genotypes: “97J1' with the highest grain set and fertile florets per spike, “H8679' with the lowest grain set and fertile florets per spike, and a medium, “YM158'. The results showed that the peak level of ABA appeared between stamen and pistil differentiation and antherlobe formation of floret development, and the timing delayed with the size of spike (earliest in “H8679” and latest in “97J1”). From antherlobe formation to meiosis, the levels of ABA and GA 1+3 decreased sharply in the ears of “97J1”, while in the ears of “H8679” there was only a slight decrease in ABA, and even an increase in GA 1+3 . The ratio of isopentenyladenosine (iPA)/ABA and IAA/ABA in the ears of “97J1” increased sharply from antherlobe formation to meiosis, but changed only slightly in the ears of “H8679”. At antherlobe formation, IAA and GA 1+3 levels were higher in the ears of “97J1”, but lower in the ears of “H8679” than in the leaves. At meiosis, ABA, GA 1+3 and IAA levels in the “97J1” ears were much lower than in the leaves, but similar in “H8679”. These results indicated that the sharp decreases of ABA and GA 1+3 in ears from antherlobe formation to meiosis and the lowest maintenance at meiosis may be favorable for development of fertile florets and enhancement of grain set in wheat.展开更多

Two expressed cDNA fragments were isolated from wheat genome. One showed 83% homology to the Mlo gene of barley. The other contained two possible open reading frames (ORFs). NBS conservative domains 2, 3 of disease resistance gene and 13 LRR structures similar to rice Pib protein terminal were found respectively in the two ORFs. It indicated that the latter fragment belongs to NBS-LRR-like genes. The obvious difference of RT-PCR products was observed between the before challenged and the challenged for 72 h by Blumeria graminis f. sp. tritici, which, implied that this sequence could be associated with disease resistance of wheat. Using nulli-tetrasomic lines of 'Chinese Spring', the NBS-LRR-like gene had been located on chromosome 1D.展开更多
A bacterial artificial chromosome library has been constructed for Triticum boeoticum Boiss (A bA b) using the bacterial artificial chromosome (BAC) vector pECBAC1. The library consists of about 170 000 clones. A ...A bacterial artificial chromosome library has been constructed for Triticum boeoticum Boiss (A bA b) using the bacterial artificial chromosome (BAC) vector pECBAC1. The library consists of about 170 000 clones. A random sampling analysis of 200 BAC clones indicates that the average insert size is 104 kb. Based on the genome size of T. boeoticum, the library is about three times as large as T. boeoticum haploid genome (5 600 Mb). Screening the BAC library with cpDNA sequence psbA gene and mtDNA sequence atp6 gene as probe shows that contamination of the library with chloroplast and mitochondrial clones is less than 1%. The library will be a useful platform in gene clone and genomic research of wheat.展开更多
