The results showed that several parameters associated with PSⅡ functions, e.g. Fv/Fo?Fv/Fm and qP were not influenced by lower salt concentration (200 mmol/L NaCl) while CO 2 assimilation rate decreased significantly. When exposed to higher salt concentration (400 mmol/L NaCl), PSⅡ functions were significantly inhibited which led to the decrease of carbon assimilation. These results suggest that different concentrations of salt stress affected photosynthesis by different modes. Salt stress made photosynthesis more sensitive to strong light and led to more serious photoinhibition. Under lower concentration of salt stress, the Q B-non-reductive PSⅡ reaction centers formed at the beginning of photoinhibition could be effectively used to compose active PSⅡ reaction center (RC) and repair the reversible inactivated PSⅡ RC. Under higher concentration of salt stress, PSⅡ reaction centers were seriously damaged during photoinhibition, the Q B-non-reductive PSⅡ RC could only be partly effective at the early time of photoinhibition, thus led to the accumulation of Q B-non-reductive PSⅡ RC in the course of restoration under dim light.展开更多
Analysis of the gene expression differentiation in leaves of wheat (Triticum aestivum L.) cultivar Baofeng 7228, under salt stress, was carried out by Differential-Display Reverse Transcription-polymerase Chain Reac...Analysis of the gene expression differentiation in leaves of wheat (Triticum aestivum L.) cultivar Baofeng 7228, under salt stress, was carried out by Differential-Display Reverse Transcription-polymerase Chain Reaction (DDRT-PCR.) Twenty-seven differential cDNA fragments were obtained. The expression of the SR07 fragment was induced noticeably by salt treatment, and the nucleotide sequence homology of 87% between the SR07 fragment and PIPs (water channel proteins) was observed. Further research showed that a 561 bp open read frame was present in the SR07 fragment. Plant expression vector of pCAMBIA-SR07 was constructed and three transformants of tobacco (Nicotiana tobacum) mediated by Agrobacterium tumefaciens plasmid were obtained. Resistance to salt, PEG, and mannitol stresses of the three transformants were examined. No significant difference (P 〉 0.05) was observed between the control and the transformants in resistance to salt stress, but there was significant difference (P 〈 0.05) between the control and the transformants in resistance to PEG and mannitol stresses. Therefore, the expression of the SR07 fragment may play an important role in the water regulation of the plant.展开更多

It has great sig- nificance to ensure the food security by improved the production of wheat in salin- ization of soil. [Method] This research is reference to previous study to be estab- lished by germination rate and seeding plants changes under salt stress to deter- mine the ability of salt tolerance of different wheat varieties. Under 300 mM NaCI to count germination rate and measuring the height, root length and root number of seeding in 200 mM and 400 mM NaCI to evaluated wheat varieties resistant to salt. [Result] Under this method, we found 3 grade-1 salt-tolerant varieties, 3 grade-2 salt-tolerant varieties, 5 grade-3 salt-tolerant varieties. [Conclusion] This study re- duced the work of salt-tolerance of wheat germplasm identification in salt pool. What's more, this study enriches the salt-tolerance of wheat germplasm resources, and provides a theoretical and technical support of wheat salt-tolerance breeding.展开更多

The changes of respiratory pathway, contents of protein, Na+...New lines of wheat ( Triticum aestivum L.) was obtained by introducing the DNA of sorghum (Sorghum vulgare Pers.) into wheat cultivar 'Longchun 13'. The changes of respiratory pathway, contents of protein, Na+ and K+ in the leaves and roots of the new lines of wheat under salt stress were determined and compared with the control cultivar, 'Longchun 13'. The decrease of the content of K+ was observed with the increase of NaCl concentrations, but the decrease was more in the control than that in the new lines, and more in roots than in leaves. Content of proline and Na+ in both two wheats lines increased greatly, but the former increased more significantly in the new lines and the latter more significantly in control both in leaves and roots. The operation of the cyanide-resistant pathway of respiration was enhanced at different degrees after salt stress and it increased much more in roots and leaves of the control plant than that in the new lines, but the cytochrome pathway of electron transport was still the main one consistently. The possible significance of these changes was discussed.展开更多

Methods BALB/C mice in 6 groups were allowed to drink either 4% dextran sulfate sodium (DSS) solution or distilled water freely with different doses of BER (15 mg·kg^-1, 45 mg·kg^-1, 150 mg·kg^-1) or sallcylazosulfapyridine (SASP, 520 mg·kg^-1), and solvent (0. 2 mL/10 mg Wt) once a day for 7 d, respectively. The symptom of ulcerative colitis was evaluated by disease activity index (DAI). Myeloperoxidase (MPO) and superoxide dismutase (SOD) activities and malondialdehyde (MDA) content were determined by HE staining and immunohistochemistry of expressions of NF-κB p65 and intercellular adhesion molecule 1 ( ICAM-1 ) proteins to observe the damage to colon tissues and possible mechanisms. Results DAI, MPO activity, MDA content and expressions of ICAM-1 and NF-κB p65 were markedly increased, while SOD activity decreased in DSS-treated mice. Treatment of mice with different doses of BER or SASP significantly decreased DAI, MPO activity and MDA content, improved histological changes of colon tissues, blunted the expressions of NF-κB p65 and ICAM-1 proteins, and enhanced SOD activity. Conclusion Berberine chloride has excellent therapeutic effect on ulcerative colitis caused by DSS in mice. The possible mechanism may be related to its antioxidant and anti-inflammatory activities associated with inhibiting the NF-κB activation and ICAM-1 expression.展开更多
A pot experiment was conducted to examine the effects of a phosphate solubilizing bacterium(PSB),Enterobacteria sp.EnHy-401,on the availability of insoluble accumulative phosphorus(P)and growth of wheat(Triticum ...A pot experiment was conducted to examine the effects of a phosphate solubilizing bacterium(PSB),Enterobacteria sp.EnHy-401,on the availability of insoluble accumulative phosphorus(P)and growth of wheat(Triticum Ningmai No.13)plants in sterile saline soil.Our results showed that the strain EnHy-401 had the ability to activate the insoluble accumulated phosphorus in saline soil and enhanced nutrient uptake efficiency by wheat plants,then conferred resistance in wheat plants to salt stress and resulted in a significant growth increase.In saline soil inoculated with Enterobacteria sp.EnHy-401,available phosphorus and exchangeable calcium was increased from 6.4 mg/kg and 1 162 mg/kg to 10.3 mg/kg and 1 214 mg/kg,respectively.Wheat seedling grown in soil inoculated with the EnHy-401 strain increased shoot weight by 28.1% and root weight by 14.6% when compared to the control.P,Ca,K and Mg contents in shoots increased 34.4%,36.3%,31.5%,and 6.3% compared to the control,respectively.the fact that the increases in available P,biomass P,and Ca2+ concentration in saline soil treated with PSB Enterobacter sp.EnHy-401 inocula,and high relativity between the P,Ca,K,and Mg content in wheat tissue and dry matter indicated that PSB Enterobacter sp.EnHy-401 suppressed the adverse effect of salinity stress in plants through nutrient(P and Ca)supply and nutrient(K and Mg)uptake enhancement.The phosphate solubilizing activity of Enterobacteria sp.EnHy-401 and the amelioration of salt stress on wheat plants by the strain varied with the salinity levels and content of organic matter in the saline soil.展开更多

[Method] The root-tips of E. acoroides, T. hemprichii and the axillary buds of H. minor were selected as the materials in this study. The conventional crushing method was used to prepare the slice specimens of chromosomes, and the karyotypes of the three species were analyzed. [Result] The chromosome numbers of E. acoroides, T. hemprichii and H. minor were 2n=18, 18, 28, respectively. The karyotype formulas were K (2n)=18= 12m+6sm, K(2n)=18=12m+6sm and K(2n)=28=16m+8sm+4st, respectively. According to the standard of Stebbins, the karyotypes of E. acoroides and T. hemprichii were 2A, and that of H. minor was 2B. There was no B chromosome and satellite in these three species. [Conclusion] According to the comparison on the karyotypes of this three species, E. acoroides and T. hemprichii are similar in karyotypes to each other, indicating that there is close, inter-relationship between E. acoroides and T. hemprichii.展开更多

The catalase (CAT) activity and malondialdehyde (MDA) content of wheat seedlings were also measured under 0.4% salt stress. [Result] Compared with seedlings in CK group (no hypergravity or salt stress), the Gr and Gi of the seedlings in salt stress treatment decreased to different extents; while the Gr and Gi of the seedlings in treatment group (hypergravity and salt stress) increased compared with that in salt stress group. CAT in seedlings of hypergravity treatment was higher than that of CK group and 0.4% salt treatment group, meanwhile the MDA showed an opposite result. This studywas conducted to investigate its effect on FITC-PAC-1, an anti-glycoprotein IIb/IIIa monoclonalantibody binding to activated platelets, and on correlative autacoid levels in plasma or inplatelets in order to explore its potential pathway of inhibiting platelet aggregation andthrombosis. Methods Monoclonal antibody binding to activated platelets was assayed by flowcytometry; NO was assessed by colorimetric method. cAMP, TXB_2 or 6-keto-PGF_(1α) levels wereassessed by radioimmunoassay. Results Gavaged 30 mg·kg^(-1) of L-arginine·L-aspartate increasedboth concentration of NO in plasma and 6-keto-PGF_(1) in incubated supernatant of aortic segment ofrats ex vivo (P < 0.05), but it did not influence cAMP content in platelets and the level of TXB_2or 6-keto-PGF_(1) in plasma of rats, whereas ASA significantly lowered TXB_2 or 6-keto-PGF_(1α) inplasma. Both 100 μmol-L^(-1) of L-arginine ·L-aspartate and ASA inhibited FITC-PAC-1 binding toactivated platelets in vitro. Conclusion The increase in NO and PGI_2 release from endo-thelialcells and consequent inhibition of platelet activation may contribute to the inhibition of plateletaggregation and thrombosis by L-arginine· L-aspartate; whereas arachidonic acid or cAMP metabolicpathway is not closely correlative with the studied effect.展开更多
