In the present study, we examined the ontogenetic pattern of behavioral responses of male giant pandas (Ailuropoda melanoleuca ) to urine odors of conspecific individuals. Our data showed that exposure to the urine of adult females induced a significant increase in sniffing and environmental sniffing/licking behaviors, but a decrease in biting behavior, in males. Males of different ages displayed specific behaviors to female urine odors. Adult males spent more time licking than juvenile and sub-adult males. Further, sub-adult and adult males displayed high levels of environmental sniffing/licking, which was absent in the juvenile males. Juvenile males displayed scent rubbing behavior significantly more frequently than sub-adult and adult males, and also spent more time showing biting behavior than sub-adult males. Finally, juvenile and sub-aduh males showed no difference in response to female and male urine odors. Together, these data suggest that chemosensory cues from conspecific urines induce age-specific responses in male giant pandas.展开更多

Therefore the objective of this study was to determine the combined effects of adenovirus-mediated wild-type (wt) p53 gene transfer and 5-FU chemotherapy on pancreatic cancer cells with different p53 gene status. METHODS:Human pancreatic cancer cell lines Capan-1^(p53mut), Capan-2^(p53wt),FAMPAC^(p53mut),PANC1^(p53mut),and rat pancreatic cancer cell lines AS^(p53wt) and DSL6A^(p53null) were used for in vitro studies.Following infection with different ratios of Ad- p53-particles (MOI) in combination with 5-FU,proliferation of tumor cells and apoptosis were quantified by cell proliferation assay (WST-1) and FACS (PI-staining).In addition,DSL6A syngeneic pancreatic tumor cells were inoculated subcutaneously in to Lewis rats for in vivo studies. Tumor size,apoptosis (TUNEL) and survival were determined. RESULTS:Ad-p53 gene transfer combined with 5-FU significantly inhibited tumor cell proliferation and substantially enhanced apoptosis in all four cell lines with an alteration in the p53 gene compared to those two cell lines containing wt-p53.In vivo experiments showed the most effective tumor regression in animals treated with Ad-p53 plus 5-FU.Both in vitro and in vivo analyses revealed that a sublethal dose of Ad-p53 augmented the apoptotic response induced by 5-FU. CONCLUSION:Our results suggest that Ad-p53 may synergistically enhance 5-FU-chemosensitivity most strikingly in pancreatic cancer cells lacking p53 function.These findings illustrate that the anticancer efficacy of this combination treatment is dependent on the p53 gene status of the target tumor cells.展开更多

Several chemotherapeutic agents have been reported to be associated with PCI, al...Pneumatosis cystoides intestinalis (PCI) is a relatively rare condition characterized by intraluminal gas in the gastrointestinal tract. Several chemotherapeutic agents have been reported to be associated with PCI, although fluorouracil-related PCI is extremely rare. We report a case of a 76-year old man who received adjuvant chemotherapy for rectal cancer with fluorouracil (FU) and leucovorin (LV). After 1 cycle of the treatment, he presented with diarrhea and abdominal pain. Abdominal radiogram revealed the presence of free air under the diaphragm and intramural gas in the intestine. Laparotomy was performed, showing a suspected diagnosis of perforation in the gastrointestinal tract. Intraoperative findings revealed penumatosis of the intestine without evidence of perforation. He was treated supportively and his symptoms improved. In conclusion, we should consider the possibility of PCI occurring in patients with malignancies during chemotherapy treatment.展开更多

METHODS: Hepatoma model was established by subcutaneous inoculation with hepatoma-22 (H22) cells into the right flank of BALB/c mice. Mice with tumor were treated by peritumoral injection of CpG ODN alone or in combination with subcutaneous injection of 5-FU. Tumor size was quantified regularly. Serum levels of IL-12 and IFN-γ in mice were assayed by enzyme-linked immunosorbent assay (ELISA). The lytic capacity of splenic NK cells was tested by lactate dehydrogenase release assay. RESULTS: Peritumoral injection of CpG ODN alone or in combination with subcutaneous injection of 5-FU, and the treatment with 5-FU alone all led to significant inhibition of hepatoma growth. The mean tumor volumes fell by 46.66% in mice injected with CpG ODN, 68.34% in the 5-FU treated mice, and 70.23% in mice treated with the combination of CpG ODN and 5-FU than in controls. There was no significant difference in tumor size between 5-FU-treated mice and mice treated with the combination of 5-FU and CpG ODN (P>0.05). The serum levels of IL-12 and IFN-γ of mice treated with CpG ODN alone (IL-12: 464.50±24.37 pg/mL; IFN-γ: 134.20?5.76 pg/mL) or with the co-administration of CpG ODN and 5-FU (IL-12: 335.83±28.74 pg/mL; IFN-γ: 111.00±5.33 pg/mL) were significantly higher than that of controls (IL-12: 237.50±45.31 pg/mL; IFN-γ: 56.75±8.22 pg/mL). The production of IL-12 and IFN-γ was suppressed moderately in 5-FU-treated mice (IL-12: 166.67±53.22 pg/mL; 53.33±16.98 pg/mL) compared to control mice (P>0.05), whereas the combination of CpG ODN and 5-FU significantly increased the serum levels of IL-12 and IFN-γ compared to 5-FU alone (P<0.05). The NK cell killing activity in CpG ODN-treated mice (44.04±1.38%) or the mice treated with CpG ODN combined with 5-FU (30.67±1.28%) was significantly potentiated compared to controls (19.22±0.95%, P<0.05). The co-administration of CpG ODN and 5-FU also significantly enhanced the lytic activity of NK cells when compared with the treatment with 5-FU alone (12.03±1.42%, P<0.05). CONCLUSION: The present data suggests that CpG ODN used as single therapeutic agent triggers anti-tumor immune response to inhibit the growth of implanted hepatoma and reverses the immunosuppression caused by the chemotherapy with 5-FU.展开更多

The effects of molar ratios of isobutyraldehyde to formaldehyde (n(I)/n(F)) and aldehyde to urea (n(A)/n(U)) on the yield, hydroxyl value (vs KOH) and softening point of the resin were investigated. The structure of the resin was characterized by FT-IR, 1H-NMR and 13C-NMR. The results show that when the molar ratio of urea to isobutyraldehyde to formaldehyde (n(U)/n(I)/n(F)) is 1.0/3.0/3.0, the yield UIF resin is 67.1%, and the softening point and hydroxyl value are 88 ℃ and 37 mg/g, respectively. The FT-IR, 1H-NMR and 13C-NMR results show that the lactam is formed by aminomethylation from urea, isobutyraldehyde, and formaldehyde.展开更多
