[Objective] Ceftiofur was as the substrate to induce the standard strain of Escherichia coli(E.coli)to be the drug-resistance one.The resistant mechanism of E.coli to ceftiofur was studied.[Method] The sub-inhibitor...[Objective] Ceftiofur was as the substrate to induce the standard strain of Escherichia coli(E.coli)to be the drug-resistance one.The resistant mechanism of E.coli to ceftiofur was studied.[Method] The sub-inhibitory concentration method was used to induce the standard strains C83907 and C83845.After they were induced for 10 generations,the double disc synergy test(DDST),NCCLS(National Committee for Clinical Laboratory Standards)confirmatory test and PCR amplification were used to detect the extend spectrum β-lactamases(ESBLs).The two fold dilution method was used to measure the minimal inhibitory concentration(MIC)of cetiofur to the strain which produced ESBLs.For the drug-resistance strain which produced ESBLs,the two fold dilution method was used to measure the minimal inhibitory concentrations of different proportions of cetiofur and tazobactam sodium.[Result] After they were induced 15 generations,MIC value of ceftiofur to the induced bacteria was during 8-10 μg/ml,and ESBLs was detected.MICs of cetiofur combining tazobactam sodium(the mass ratio was 1∶1-8∶1)to Escherichia coli produced ESBLs reduced 20-22 times than that of cetiofur.[Conclusion] The main mechanism of pathogenic Escherichia coli resistance to ceftiofur was that which produced ESBLs.展开更多

Compared with the GenBank, the identity of tTr irp2 gene sequence and the fyuA gene sequence of the strain reached 98.5% 99.2% and 98.9%-100% respectively. Compared with each other, the identity of tt- two gene sequences of irp2 was 99.3%, and that between the two fyuA gene se quences was 98.9%. The whole expression cartridge , consisting of promoter. SD sequence , target gene and transcription terminator, was tandem repeatedly engineered into a expression plasmid. Consequently, the copy number of specific gene was increased substantially, leading to the improvement of expression efficiency.Using this approach, a recombinant plasmid , designed as PLYD, was constructed and transformated into the Escherichia coli strain DH5α. Upon induction , the desired protein was synthesized in a considerable level and accumulated up to 63% of the total cell proteins. The present study revealed that tandem repeating of expression cartridge provided a convenient means to improve expression level efficiently.展开更多

A novel method to produce 3HD by recombinant bacteria was constructed in Escherichia coli HB101 and Pseudomonas putida GPpl04, respectively. Simultaneous expression of both phaG encoding (R)-3-hydroxydecanoyl-ACP:CoA transacylase and tesB encoding thioesteraseⅡin E. coli HB101 increased 3HD production approximate 1.7-folds compared with the expression of phaG gene alone under identical conditions. In addition, when the tesB gene was introduced into the strain, the polyhydroxyalkanoate synthase negative strain P. putida GPpl04 produced extracellular 3HD. Thus, a novel pathway to produce 3HD by recombinant Pseudomonas was constructed. It was also found that the ratio of carbon source to nitrogen source affected the production of 3HD by recombinant P. putida harboring tesB gene. The mean-field results suggest that for a small Hund's interaction, the flavor liquid state is stable against the solid state, but with sufficient deviation from the SU(4) limit the long-range order may be attained in 2D system. Furthermore, we employ a variational approach to calculate the phase diagram of the ground state and the temperature-dependent susceptibility by taking into account the Hund's interaction and the anisotropy in orbital wavefunctions. Finally, the implications for the experimental observations on the material, , are discussed.展开更多

Although Thomas is a creative and distinctive text for its own community, the major framework of the Logia in a textual-statistic way was based on the traditional Sophia literature that had affected the culture, custom, and authority of the Hellenised Jewish societies. Then, what kind of scrolls, books, and writings could be the religio-historical sources for Thomas? How was Thomas transformed from those sapiential manuscripts? While three data of "the Pauline Sophia concept", "the sapiential Logia of Q", and "the sapiential themes of the Jewish apocalyptic writings" are quite relevant, the Greek fragment of Papyrus Oxyrhynchus (P. Oxy.) 654 and NHC II, 2.32:10-51:28 will evince the new insight that the ancient Logia of the Thomasine community was not "the traditional Q", but could be "a new Q" coming from the common (oral, or written, or both) Sophia tradition, as an independently developed one.展开更多
Two new Mannich and Schiff base ligands namely: (1-(3'-amino-methyl-1'(H)-1',2',4'-triazole)-3-(1'(H)-l',2',4'- triazole-3'-yl) iminoisatin (Lt) and l-morpholinomethyl-3-(1'(H)-1',2',4'-tr...Two new Mannich and Schiff base ligands namely: (1-(3'-amino-methyl-1'(H)-1',2',4'-triazole)-3-(1'(H)-l',2',4'- triazole-3'-yl) iminoisatin (Lt) and l-morpholinomethyl-3-(1'(H)-1',2',4'-triazolyl)iminoisatin (LII) were prepared from condensation reaction of the new Schiff base 3-(l'(H)-l',2',4'-triazole-3'-yl)iminoisatin (SBH) (0.01 mol) with formaldehyde and 3-amino-l,2,4-triazole (3-At) or with formaldehyde and morpholine (0.01 mole each) respectively. Structures were characterized by tH NMR, CHN analyses, mass spectra and IR spectra. Metal complexes were synthesized by heating each ligand (0.02 mo with metal salts (0.01 moo in ethanol or ethanol: acetone mixture respectively. The formula of metal complexes were suggested depending on CHN and thermal analyses, metal and chloride contents, IR and UV-visible spectra, magnetic moments (laefr) and conductivity measurements. All complexes were of octahedral geometry except palladium (II) complexes which were square planar. The two ligands and some selected metal complexes showed various antibacterial and cytotoxic effects.展开更多

It is a country in the Era of Transition of nowad...According to recent hydropower inventories, the first aim of this paper is to prove that Greece is a real "Archimedean Soft Small Hydro Development Terra Incognita". It is a country in the Era of Transition of nowadays, having an important unexploited hydropower potential of several TWh and thousands of MW. The second goal is to present the results of rediscovering the old screw pumps and the always-modern Archimedean cochlear screw ideas as a series of soft hydropower turbines with inclined axis rotors and floating spiral screw devices. By following the similarity methodology, a few small-scale models of cochlear rotors were designed, developed and tested in an Armfield hydraulic channel. The conventional and unconventional, low or zero-head screw turbines, could efficiently harness the important and unexploited Greek small potential and kinetic hydraulic energy of all the natural watercourses, the man-made open urban or rural channels and the most important tidal or sea currents, producing useful green electricity. The very promising low-head hydropower inventory of Greece, the preliminary experimental results and the research studies of cochlear projects in watercourses, hydraulic works, tidal conditions of Euripus Strait and in the mysterious "Sea River Current of Cephalonia", seems to give a good answer to the question "Quo Vadis Archimedes nowadays in Greece, in the Era of Transition?".展开更多
AIM: To investigate the relationship between mutations of rearranged during transfection (RET) proto-oncogene and Chinese patients with Hirschsprung's disease (HD), and to elucidate the genetic mechanism of famili...AIM: To investigate the relationship between mutations of rearranged during transfection (RET) proto-oncogene and Chinese patients with Hirschsprung's disease (HD), and to elucidate the genetic mechanism of familial HD patient at the molecular level.METHODS: Genomic DNA was extracted from venous blood of probands and their relatives in two genealogies.Polymerase chain reaction (PCR) products, which were amplified using specific primers (RET, exons 11, 13, 15and 17), were electrophoresed to analyze the single-strand conformational polymorphism (SSCP) patterns. The positive amplified products were sequenced. Forty-eight sporadic HD patients and 30 normal children were screened for mutations of RET proto-oncogene simultaneously.RESULTS: Three cases with HD in one family were found to have a G heterozygous insertion at nucleotide 18 974 in exon 13 of RET cDNA (18 974insG), which resulted in a frameshift mutation. In another family, a heterozygosity for T to G transition at nucleotide 18 888 in the same exon which resulted in a synonymous mutation of Leu at codon 745 was detected in the proband and his father. Eight RET mutations were confirmed in 48 sporadic HD patients.CONCLUSION: Mutations of RET proto-oncogene may play an important role in the pathogenesis of Chinese patients with HD. Detection of mutated RET proto-oncogene carriers may be used for genetic counseling of potential risk for HD in the affected families.展开更多

The one dimensional and piecewise tent map with changeable parameters decided by the position index of blocked messages is chosen, and blocked massages translated into ASCII code values are employed as the iteration time of the chaotic tent map. The final 128-bit hash value is generated by logical XOR operation on intermediate hash values. Theoretical analysis and computer simulation indicate that the proposed algorithm can satisfy the performance requirements of hash function.展开更多

(Pierre Laurent Wantzel, 1837) obscurely presented a proof based on ideas from Galois field showing that, the solution of angle trisection corresponds to solution of the cubic equation; x3 - 3x - 1 = 0, which is geometrically irreducible [1]. The focus of this work is to show the possibility to solve the trisection of an angle by correcting some flawed methods meant for general construction of angles, and exemplify why the stated trisection impossible proof is not geometrically valid. The revealed proof is based on a concept from the Archimedes proposition of straightedge construction [2, 3].展开更多

Methods: The mRNA of E. Coli was enriched from the total RNA with oligo(dT)-cellulose, prior to reverse transcription using oligo(dT)18as the primer. Double-stranded cDNA was subsequently synthesized, which was subjected to digestion with Sau3A I to produce multiple gene fragments for ligation with the adapters. PCR was carried out in 10 groups according to 10 different pairs of the selective primers, and the PCR products were then cloned into T-vectors. Results: More than 100 gene fragments had been cloned, 30 of which were sequenced. Conclusion:Polyadenylation of E. Coli mRNA may not be a biochemical curiosity but a general attribute of bacterial mRNA.展开更多

The paper, as the title indicates, will be confined to a comparison and evaluation of Platonic love against love as articulated within Christianity. Both forms of love will be analyzed and I will attempt to show that although Plato, through Socrates (and Socrates through the Diotima character), tries to redeem the traditional understanding of love in the ancient Greek society that he was living in, Platonic love is still very different from the Christian concept of love.展开更多

文摘Two new Mannich and Schiff base ligands namely: (1-(3'-amino-methyl-1'(H)-1',2',4'-triazole)-3-(1'(H)-l',2',4'- triazole-3'-yl) iminoisatin (Lt) and l-morpholinomethyl-3-(1'(H)-1',2',4'-triazolyl)iminoisatin (LII) were prepared from condensation reaction of the new Schiff base 3-(l'(H)-l',2',4'-triazole-3'-yl)iminoisatin (SBH) (0.01 mol) with formaldehyde and 3-amino-l,2,4-triazole (3-At) or with formaldehyde and morpholine (0.01 mole each) respectively. Structures were characterized by tH NMR, CHN analyses, mass spectra and IR spectra. Metal complexes were synthesized by heating each ligand (0.02 mo with metal salts (0.01 moo in ethanol or ethanol: acetone mixture respectively. The formula of metal complexes were suggested depending on CHN and thermal analyses, metal and chloride contents, IR and UV-visible spectra, magnetic moments (laefr) and conductivity measurements. All complexes were of octahedral geometry except palladium (II) complexes which were square planar. The two ligands and some selected metal complexes showed various antibacterial and cytotoxic effects.
文摘According to recent hydropower inventories, the first aim of this paper is to prove that Greece is a real "Archimedean Soft Small Hydro Development Terra Incognita". It is a country in the Era of Transition of nowadays, having an important unexploited hydropower potential of several TWh and thousands of MW. The second goal is to present the results of rediscovering the old screw pumps and the always-modern Archimedean cochlear screw ideas as a series of soft hydropower turbines with inclined axis rotors and floating spiral screw devices. By following the similarity methodology, a few small-scale models of cochlear rotors were designed, developed and tested in an Armfield hydraulic channel. The conventional and unconventional, low or zero-head screw turbines, could efficiently harness the important and unexploited Greek small potential and kinetic hydraulic energy of all the natural watercourses, the man-made open urban or rural channels and the most important tidal or sea currents, producing useful green electricity. The very promising low-head hydropower inventory of Greece, the preliminary experimental results and the research studies of cochlear projects in watercourses, hydraulic works, tidal conditions of Euripus Strait and in the mysterious "Sea River Current of Cephalonia", seems to give a good answer to the question "Quo Vadis Archimedes nowadays in Greece, in the Era of Transition?".

文摘AIM: To investigate the relationship between mutations of rearranged during transfection (RET) proto-oncogene and Chinese patients with Hirschsprung's disease (HD), and to elucidate the genetic mechanism of familial HD patient at the molecular level.METHODS: Genomic DNA was extracted from venous blood of probands and their relatives in two genealogies.Polymerase chain reaction (PCR) products, which were amplified using specific primers (RET, exons 11, 13, 15and 17), were electrophoresed to analyze the single-strand conformational polymorphism (SSCP) patterns. The positive amplified products were sequenced. Forty-eight sporadic HD patients and 30 normal children were screened for mutations of RET proto-oncogene simultaneously.RESULTS: Three cases with HD in one family were found to have a G heterozygous insertion at nucleotide 18 974 in exon 13 of RET cDNA (18 974insG), which resulted in a frameshift mutation. In another family, a heterozygosity for T to G transition at nucleotide 18 888 in the same exon which resulted in a synonymous mutation of Leu at codon 745 was detected in the proband and his father. Eight RET mutations were confirmed in 48 sporadic HD patients.CONCLUSION: Mutations of RET proto-oncogene may play an important role in the pathogenesis of Chinese patients with HD. Detection of mutated RET proto-oncogene carriers may be used for genetic counseling of potential risk for HD in the affected families.

文摘A new algorithm for a novel hash function, based on chaotic tent map with changeable parameter, is proposed and analyzed. The one dimensional and piecewise tent map with changeable parameters decided by the position index of blocked messages is chosen, and blocked massages translated into ASCII code values are employed as the iteration time of the chaotic tent map. The final 128-bit hash value is generated by logical XOR operation on intermediate hash values. Theoretical analysis and computer simulation indicate that the proposed algorithm can satisfy the performance requirements of hash function.
文摘The objective of this paper is to provide a provable solution of the ancient Greek problem of trisecting an arbitrary angle employing only compass and straightedge (ruler). (Pierre Laurent Wantzel, 1837) obscurely presented a proof based on ideas from Galois field showing that, the solution of angle trisection corresponds to solution of the cubic equation; x3 - 3x - 1 = 0, which is geometrically irreducible [1]. The focus of this work is to show the possibility to solve the trisection of an angle by correcting some flawed methods meant for general construction of angles, and exemplify why the stated trisection impossible proof is not geometrically valid. The revealed proof is based on a concept from the Archimedes proposition of straightedge construction [2, 3].
文摘Objective:To investigate the polyadenylation of mRNA in E. Coli. Methods: The mRNA of E. Coli was enriched from the total RNA with oligo(dT)-cellulose, prior to reverse transcription using oligo(dT)18as the primer. Double-stranded cDNA was subsequently synthesized, which was subjected to digestion with Sau3A I to produce multiple gene fragments for ligation with the adapters. PCR was carried out in 10 groups according to 10 different pairs of the selective primers, and the PCR products were then cloned into T-vectors. Results: More than 100 gene fragments had been cloned, 30 of which were sequenced. Conclusion:Polyadenylation of E. Coli mRNA may not be a biochemical curiosity but a general attribute of bacterial mRNA.
文摘This paper will discuss Plato's view of love in The Symposium, in particular the arguments presented by the Diotima character, but not neglecting all the other views of love presented therein. The paper, as the title indicates, will be confined to a comparison and evaluation of Platonic love against love as articulated within Christianity. Both forms of love will be analyzed and I will attempt to show that although Plato, through Socrates (and Socrates through the Diotima character), tries to redeem the traditional understanding of love in the ancient Greek society that he was living in, Platonic love is still very different from the Christian concept of love.
