It has been found that N-ras was overexpressed at the mENA and protein level in hepatoma cells. In order to explore the biological roles of N-ras in human hepatic oaroinogenesis and the potential application in control of cancer cell growth, a pseudotype retrovirus containing antisense sequence of human N-ras was constructed and packaged. A recombinant rebrovirus vector containing antisense or sense sequences of N-ras oDNA was constructed by pZIP-NeoSV(X)1. The pseudotype virus was packaged and rescued by transfeotion and infection in PA317 and ψ2 helper cells. It has been demonstrated that the pseudotype retrovirus containing antisense N-ras sequence did inhibit the growth of human PLC/PEF/5 hepatoma cells accompanied with inhibition of p21 expression, while the retrovirus containing sense sequence had none. The pseudotype virus had no effect on human diploid fibroblasts.展开更多

The approach for the phase space reconstruction of chaotic time series...This paper presents a fault-detection method based on the phase space reconstruction and data mining approaches for the complex electronic system. The approach for the phase space reconstruction of chaotic time series is a combination algorithm of multiple autocorrelation and F-test, by which the quasi-optimal embedding dimension and time delay can be obtained. The data mining algorithm, which calculates the radius of gyration of unit-mass point around the centre of mass in the phase space, can distinguish the fault parameter from the chaotic time series output by the tested system. The experimental results depict that this fault detection method can correctly detect the fault phenomena of electronic system.展开更多

Traditional architectural form as famous communication home Marshall McLuhan's "human extension", leads the order and the rules in the form of elevation. The spread of universal ethics, traditional architectural symmetry axis ceremony, and the adornment of the mythical thinking component, are traditional Chinese ethics of society to describe the core of creation through three points sequence structure constraint. The wisdom of metaphor in these ideas is the life gene of Chinese culture. β-carotene ketolase and β-carotene hydroxylase encoded by bkt and bch, respectively, are key enzymes required for astaxanthin biosynthesis in Haematococcu pluvialis 34-1n. Two expression vectors containing cDNA sequ...β-carotene ketolase and β-carotene hydroxylase encoded by bkt and bch, respectively, are key enzymes required for astaxanthin biosynthesis in Haematococcu pluvialis 34-1n. Two expression vectors containing cDNA sequences of bkt and bch were constructed and co-transformed into cell-wall-deficient Chlamydomonas reinhardtii CC-849. Transgenic algae were screened on TAP agar plates containing 10 gg mL 1 Zeomycin. PCR-Southern analysis showed that bkt and bch were integrated into the genomes of C. reinhardtii. Transcripts of bkt and bch were further confirmed by RT-PCR-Southern analysis. Compared with the wild type, transgenic algae produced 29.04% and 30.27% more carotenoids and xanthophylls, respectively. Moreover, the transgenic algae could accumulate 34% more astaxanthin than wild type. These results indicate that foreign bkt and bch genes were successfully translated into β-carotene ketolase and β-carotene hydroxylase, which were responsible for catalyzing the biosynthesis of astaxanthin in transgenic algae.展开更多
