Sodium butyrate (SB) can cause functional and morphological effects on Clone A cells including growth arrest at Go/G1 stage and cell differentiation as observed by morphological changes, MTT and flow cytometry assays, as well as reduced Grp94 gene expression as shown by Northern blot and Western blot assays. The possible mechanism of the correlation between Grp94 gene expression and tumor growth inhibition and cell differentiation is briefly discussed.展开更多

METHODS: We prospectively collected clinicopathological, immunohistochemical and semi-quantitative reverse t...AIM: To elucidate the role of overexpressed polo-like kinasel (PLK1)in hepatocellular carcinoma (HCC). METHODS: We prospectively collected clinicopathological, immunohistochemical and semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) data from 135 HCC patients undergoing successful hepatectomy. The correlations between PLK1 mRNA expression and clinicopathologic variables were analyzed by Mann-Whitney U test. Prognostic factors were identified by univariate and multivariate analyses. RESULTS: Immunohistochemical results showed overexpression of PLK1 was mainly found in tumor tissues compared with tumor-free tissue. A similar mRNA result was obtained by semi-quantitative RT-PCR. A total of 111 samples were positive for PLK1 mRNA expression. The positive expression was correlated with venous invasion, tumor nodules and Edmondson grade. Furthermore, 1, 3, 5-year survival rates in the positive expression group were significantly lower than the negative control group. Multivariate analysis showed that positive PLK1 expression was an independent risk factor for HCC. CONCLUSION: PLK1 could be a potential biomarker for diagnosis and therapy for HCC.展开更多

The paper aims to provide new information that may elucidate the modulatory effect of estrogen on the bone-resorbing cytokines RANKL and its anti-resorptive factor OPG secrected by HPLFs which are already force-stimulated. The expression of OPG mRNA is rising after mechanical loading either with or without stimulated by estrogen before. But HPDL cells exposured to estrogen for 24 h before loaded tend to express more OPG mRNA. Compared with the no-estrogen group, the inhibit trend of RANKL mRNA is much more apparent in with-estrogen group. Moreover, estrogen and mechanic force time-dependently increased OPG expression and attenuated the RANKL expression.展开更多
OBJECTIVE:To explore electro-acupuncture's(EA's)effect on gene expression in heart of rats with stress-induced pre-hypertension and try to reveal its biological mechanism based on gene chip technology.METHODS:...OBJECTIVE:To explore electro-acupuncture's(EA's)effect on gene expression in heart of rats with stress-induced pre-hypertension and try to reveal its biological mechanism based on gene chip technology.METHODS:Twenty-seven Wistar male rats were randomly divided into 3 groups.The stress-induced hypertensive rat model was prepared by electric foot-shocks combined with generated noise.Molding cycle lasted for 14 days and EA intervene was applied on rats in model + EA group during model preparation.Rat Gene 2.0 Sense Target Array technology was used for the determination of gene expression profiles and the screened key genes were verified by real-time quantitative polymerase chain reaction(RT-PCR) method.RESULTS:Compared with blank control group,390 genes were changed in model group;compared with model control group,330 genes were changed in model + EA group.Significance analysis of gene function showed that the differentially expressed genes are those involved in biological process,molecular function and cellular components.RT-PCR result of the screened key genes is consistent with that of gene chip test.CONCLUTION:EA could significantly lower blood pressure of stress-induced pre-hypertension rats and affect its gene expression profile in heart.Genes that related to the contraction of vascular smooth muscle may be involved in EA's anti-hypertensive mechanism.展开更多
