以聚丙烯腈纤维为载体 ,共价结合制备工业生产应用的固定化青霉素酰化酶。经过测定 ,获得了制备固定化酶的基本条件 :对部分酸水解聚丙烯腈纤维进行霍夫曼重排反应溶液中的次氯酸钠浓度为 4%~ 16 % (v/v) ;用含戊二醛的溶液对经过霍夫... 以聚丙烯腈纤维为载体 ,共价结合制备工业生产应用的固定化青霉素酰化酶。经过测定 ,获得了制备固定化酶的基本条件 :对部分酸水解聚丙烯腈纤维进行霍夫曼重排反应溶液中的次氯酸钠浓度为 4%~ 16 % (v/v) ;用含戊二醛的溶液对经过霍夫曼重排反应的水解纤维进行活化反应溶液的pH为 6 5~ 8 5 ,最适 pH7 0 ;活化反应时间为 10min ;活化纤维中醛基含量为 12 5 0 μmol/g以上 ;酶与活化纤维连接反应溶液的最适pH值为 6 5~ 7 0 ;反应时间为 10min。在酶与活化纤维的连接反应溶液中加入竞争性抑制剂 苯乙酸 ,对酶活力具有保护作用 ,能提高酶活力的固定化效率。在酶与活化纤维的连接反应溶液中剩余的游离酶经吸附、洗涤和浓缩后 ,可以再用于制备固定化酶。变性固定化酶经再生活化后作为载体固定化新的酶液 ,也能制备出较好的固定化酶。 展开更多

本文研究了石油化工铝硅载体废催化剂中铂含量的电感耦合等离子体原子发射光谱测定方法。样品以硫酸溶解石油化工铝硅载体废催化剂,用氯气氧化络合铂进入溶液,在2%(v/v)硫酸条件下,用标准曲线法对废催化剂中的Pt进行测定,可精确测定0.10... 本文研究了石油化工铝硅载体废催化剂中铂含量的电感耦合等离子体原子发射光谱测定方法。样品以硫酸溶解石油化工铝硅载体废催化剂,用氯气氧化络合铂进入溶液,在2%(v/v)硫酸条件下,用标准曲线法对废催化剂中的Pt进行测定,可精确测定0.100%~0.800%含量的铂,常见的杂质元素不干扰测定。结果表明,以低浓度硫酸为介质,用标准曲线法进行样品分析,方法的检出限为0.010μg/mL,样品的加标回收率为97.2%~101.8%。通过试验结果可知,该方法准确性好、精密度高,准确快速,操作简单。 展开更多

Laccase was immobilized on the ceramic-chitosan composite support by using glutaraldehyde as the cross-linking reagent. The immobilization conditions and characterization of the immobilized enzyme were investigated. The immobilization of laccase was successfully realized when 3.0 mL of 1.25 mg/mL of laccase at a pH value of 4.0 reacted with 0.15 g of ceramic-chitosan composite support(CCCS) at 4 ℃ for 24 h. The immobilized enzyme exhibited a maximum activity at pH 3.0. The optimal temperatures for immobilized enzyme were 25 ℃ and 50 ℃. The K_m value of immobilized laccase for ABTS was 66.64 μmol/L at a pH value of 3.0 at 25 ℃. Compared with free laccase, the thermal, operating and storage stability of immobilized laccase was improved after the immobilization. 展开更多

Urbanization is the physical growth of urban areas as a result of global change. As the land cost is increasing tremendously and decreasing availability of good construction site is building up pressure on the engineers to utilize even the poorest site either by providing special type of foundation or by improving ground in urban centres. In this context literature is reviewed for use of landfill site for housing. The site exploration for old dump site was carried out to assess subsoil characteristics. The objective was to evolve strategy for economical feasible ground improvement technique to obtain permissible bearing capacity of 150 kPa and total settlement not more than 50 mm. The tests carried out are load tests with geotextile mat and stone filled wire mess matress. The analysis was attempted to evaluate the soil response and bearing capacity. The site can be used for construction of low rise housing for rehabilitation of displaced persons under TP scheme within city area utilizing old landfill sites. 展开更多
