痕量胺相关受体(trace amine-associated receptors,TAARs)是一类G蛋白偶联受体,除TAAR1外,其余TAARs与嗅觉受体一样都在嗅觉上皮表达并发挥感知气味的作用。近年来,非嗅觉组织中同样发现TAARs高表达,提示TAARs可能存在重要的异位生理... 痕量胺相关受体(trace amine-associated receptors,TAARs)是一类G蛋白偶联受体,除TAAR1外,其余TAARs与嗅觉受体一样都在嗅觉上皮表达并发挥感知气味的作用。近年来,非嗅觉组织中同样发现TAARs高表达,提示TAARs可能存在重要的异位生理功能。事实上,相关研究证实内源性以及外源性特定痕量胺通过作用于不同的TAARs在非嗅觉组织中参与调节多种生理功能,表明TAARs有成为新的诊断和治疗靶点的潜力。本文系统介绍了异位嗅觉TAARs的表达、内源性以及外源性的配体、食物中的生物胺、异位嗅觉TAARs介导的信号通路以及生理和病理功能的现有研究,一方面为未来药物靶点开发、生理病理学研究提供新思路,另一方面也为食品中生物胺的生物活性研究提供新的研究方向。 展开更多

To investigate the effect of Yikun Neiyi Wan (益坤内异丸YKNYW) and gestrinone on the expression of aromatase P450 (P450arom), cyclo-oxygenase-2 (COX-2) and estrogen receptor (ER) in isolated ectopic and normal endometriat stroma cells in vitro. Methods: Digestion and serial filtration were used to isolate and culture the ectopic and eutopic endometrial cells from patients with chocolate cyst in virto Transformation of the cell morphology was observed in a inverted microscope. The effect of YKNYW on the expression of aromatase P450, cyclo-oxygenase-2, estrogen receptor in cultured endometriosis cells were detected by immunohistochemical method. Results: The expression levels of P450arom, COX-2 in glandular epithelium cells in vitro were decreased significantly by YKNYW compared with gestrinone (P〈0.05). ER expression in mesenchymal cells of endometriosis was increased by YKNYW in the large and medium dosage groups compared with gestrinone. Conclusion: The mechanism by which YKNYW alleviates endometriosis pain is possibly related to the decrease in ectopic endometrial P450 arom and COX-2 expression in glandular epithelium, contrary to gestrinone, and the increase in ER expression in mesenchymalis, consistent with gestrione in patients with endometriosis. 展开更多

The human influenza A (H3N2) virus dominated the 2014-2015 winter season in many countries and caused massive morbid- ity and mortality because of its antigenic variation. So far, very little is known about the antigenic patterns of the recent H3N2 virus. By systematically mapping the antigenic relationships of H3N2 strains isolated since 2010, we discovered that two groups with obvious antigenic divergence, named SW13 (A/Switzerland/9715293/2013-1ike strains) and HK14 (A/Hong Kong/5738/2014-1ike strains), co-circulated during the 2014-2015 winter season. HK14 group co-circulated with SW13 in Europe and the United States during this season, while there were few strains of HK14 in China's Mainland, where SW13 has dominated since 2012. Furthermore, we found that substitutions near the receptor-binding site on hemagglutinin played an im- portant role in the antigenic variation of both the groups. These findings provide a comprehensive understanding of the recent antigenic evolution of H3N2 virus and will aid in the selection of vaccine strains. 展开更多

Intracellular Ca2+homeostasis is essential for vascular function and blood pressure regulation.Because of their unique roles in regulating intracellular Ca2+concentration and vascular function,a novel class of non-selective cation channels,called transient receptor potential(TRP)channels,have emerged at the frontier of hypertension research.Based on their role in vasculature function regulation,TRP channels can be divided into two functional subtypes:one that participates in vasoconstriction and one that participates in vasodilatation.A functional imbalance of these two subtypes of TRP channels may disturb intracellular calcium([Ca2+]i)homeostasis,and the consequent vascular dysfunction may contribute to the development of hypertension.The potential of these TRP channels as novel pharmacological targets for the treatment of human hypertension is of great interest. 展开更多
