目的 研究 5 0Hz正弦磁场对细胞连接蛋白Cx4 3移位的影响 ,以探索极低频磁场 (ELFMF)抑制细胞间隙连接通讯 (GJIC)的机制。方法 用 5 0Hz、0 .8mT的正弦磁场和 (或 )十四酰基咐哌醇酯 (TPA ,5ng/ml)对培养的中国仓鼠肺成纤维细胞 (CHL... 目的 研究 5 0Hz正弦磁场对细胞连接蛋白Cx4 3移位的影响 ,以探索极低频磁场 (ELFMF)抑制细胞间隙连接通讯 (GJIC)的机制。方法 用 5 0Hz、0 .8mT的正弦磁场和 (或 )十四酰基咐哌醇酯 (TPA ,5ng/ml)对培养的中国仓鼠肺成纤维细胞 (CHL)处理 2 4h(其中TPA处理 1h)。采用间接免疫荧光细胞化学法和激光共聚焦显微镜进行连接蛋白Cx4 3定位的测定 ;采用Westernblot方法检测胞浆和核内Cx4 3蛋白的含量。结果 正常组细胞间连接处有明亮的点状标记 ,连接成线 ;TPA处理组、0 .8mT正弦磁场单独作用组及 0 .8mT正弦磁场联合TPA处理组细胞的连接处标记斑点均较正常组减少 ,大量Cx4 3蛋白标记斑点出现在胞浆内 ,且在核附近聚集。各组核内Cx4 3蛋白条带很淡 ,而胞浆内Cx4 3蛋白含量ELF组 (2 .0 3± 0 .89)和TPA组 (2 .4 3± 0 .82 )明显增加 ,与正常组 (1.0 4± 0 .17)相比 ,差异有显著性 (P <0 .0 1)。结论  5 0Hz正弦磁场直接和 (或 )协同TPA抑制体外培养细胞GJIC功能与细胞连接蛋白Cx4 展开更多

Analysis on the Observation Data of Three Profiles across the Qilian Mountain-Haiyuan Fault Zone

By analyzing GPS, leveling and gravity observation data from the Minle, Huazangsi and Shagouhe profiles across the Qilian Mountain-Haiyuan fault during 2005 - 2009, the changes of these data and activity characteristics of the main faults in different time are obtained. The results show that the Changma-Erbo, Maomaoshan, Zhunglanghe and Haiyuan faults have been active in recent years. The relationship between the vertical displacement and mobile gravity data is analyzed, and the results reveal the gravity changes are mainly due to mass transfer. The results of GPS observation show that the direction of the horizontal movement velocity was almost along the fault strike in 2006 - 2007, while the horizontal velocity deviated from the fault direction in 2007 - 2008, characterized by increased eastward displacement. Further analysis upon removal of the linear trend on the time series of the N and E coordinate values of the continuous observation stations of the Crustal Movement Observation Network of China near the three profiles reveals that the Wenchuan earthquake had an effect on the observations in 2007 - 2008. Comparing the gravitational changes in 2008 - 2009 with the Bouguer gravity anomaly background, we find that the area near Minle profile is in the stage of adjustment and recovery, while the anomaly is not completely recovered around Huazangsi and Shagouhe profiles. 展开更多

The distinct characteristic of subcritical propagation requires at least two control parameters to completely express expansion of cracks in the concrete.The basic definition of crack tip opening displacement(CTOD)is corrected based on the monitored variable of crack mouth opening displacement(CMOD).The improved CTOD criterion,called the double CTODd criterion,for the abnormality diagnosis of cracks in the concrete is constructed based on the fictitious crack model and linear asymptotic superposition assumptions of the concrete.The double CTODd criterion is verified using two specimens of three-point bending notched beam.The verification results are reasonable and correct.The establishment of the double CTODd criterion lays a solid theoretical foundation for the abnormality diagnosis of cracks in concrete structures(such as concrete dams)based on CMOD. 展开更多
