通过80%硫酸铵沉淀与Sephadex G-50凝胶过滤层析,从当归饮片中得到分子量为18.33 kDa的当归蛋白(命名AP)。用X射线对小鼠进行全身照射至吸收剂量6 Gy,建立急性照射损伤模型,以阿米福汀为对照,监测小鼠的体质量、外周血白细胞数、胸腺指... 通过80%硫酸铵沉淀与Sephadex G-50凝胶过滤层析,从当归饮片中得到分子量为18.33 kDa的当归蛋白(命名AP)。用X射线对小鼠进行全身照射至吸收剂量6 Gy,建立急性照射损伤模型,以阿米福汀为对照,监测小鼠的体质量、外周血白细胞数、胸腺指数、脾脏指数、脾集落形成单位(CFU-S)、脾脏表观、脾组织病理分析和脾脏抗氧化活力等指标,检验当归蛋白AP的辐射防护效应。结果表明,当归蛋白AP可使受照小鼠的外周血白细胞数量、胸腺指数、脾脏指数、CFU-S和脾脏的过氧化氢酶活力分别增加135%、103%、23%、219%和180%,改善脾白髓损伤,使其体质量下降率减少94%。结果提示,当归蛋白AP对小鼠具有辐射防护作用。 展开更多

Objective: The aim of our study was to investigate the effect of Angelica keiskei chalcone (AC) on the expression of Caspase-3 and Bax in mice hepatocarcinoma cells. Methods: Fifty mice inoculated hepatocarcinoma 22 cells were divided into five groups, 10 mice per group. Mice were given 5, 20, 40 mg/kg AC daily by mouth in low, middle and high dose groups respectively. Saline were given to the tumor control group by mouth. Twenty mg/kg cytoxan (CTX) by injection every other day were given to the positive control group. Ten days later, all mice were sacrificed. The levels of the Caspase-3 and Bax protein expression were measured by immunohistochemistry method and the proliferation activity of hepatocarcinoma cells was determined by MTT assay. Results: The expression level of Caspase-3 and Bax protein in tumor control group were 5.00% and 4.68%, respectively, and those of the high-dose group were 38.52% and 35.76%. The differences between two groups were significant (P < 0.05). The cell proliferation activity of tumor control group and high-dose group were 1.135 ± 0.032 and 0.716 ± 0.018. The difference was significant (P < 0.05). Conclusion: AC can increase the expression of Caspase-3 and Bax protein, and inhibit the proliferative activity of mice hepatocarcinoma cells. 展开更多
