Human X-box binding protein 1 (XBP1), an important transcription factor, participates in many signal transduction processes. To further investigate the biological function of XBP1, sequences of XBP1 promoter and its...Human X-box binding protein 1 (XBP1), an important transcription factor, participates in many signal transduction processes. To further investigate the biological function of XBP1, sequences of XBP1 promoter and its two deletion mutants were first determined using bioinformatic analysis. The report vectors containing XBP1 promoter and its deletion mutants were then constructed, namely, p1-XBPlp, p2-XBPlp, and p3-XBPlp. Each reporter vector was separately transfected into HepG2, L02, K562, SMMC-7721, HSF, and Lipocyte lto Cell line using FuGENE 6 transfection reagents. The activity of chloramphenicol acetyltransferase (CAT) in each group of transfected cells was detected by ELISA assay, which in turn reflects the transcription activity of the XBP1 gene promoter. The activity involving p3-XBPlp was the highest in HepG2, which was 12.4-fold of that of pCAT3-Basic. The activities of p3-XBPlp in K562 and SMMC-7721 were the second and the third highest, which were 10.9-fold and 10.0-fold of that of the pCAT3-Basic, respectively. The CAT activity in L02 was lower than that in the above-mentioned abnormal cell, and no reporter activity was detected in HSF and Ito Cell. The XBP1 transcription and expression in K562, HepG2 and SMMC-7721 were found to be higher than that in L02, HSF and Ito cells, based on the results of real-time RT-PCR and Western blot. The XBP1 transcription and expression in L02, HSF was lower, whereas that in Ito cells was totally lacking. The result was similar to that of CAT-ELISA. Therefore, the XBP1 gene promoter can drive its downstream gene expression and its activity is cell line-dependent. The core sequence of XBP1 promoter was found between -227bp and 66bp sequence. This sequence was closely associated with the transcriptional activity of XBP1 promoter.展开更多

The reliability and validity arouses a dispute on Internet-based CET (iB-CET) oral test in China, leading to the penetration into oral test contents in dialogue, description or comment, and question answering. Then, a probe into transformation from a spoken document into a textual document is touched on a conversion ratio of 72% adopted at the present time, AEM to be carried forward being a necessary hypothesis for Afterwards, this paper focuses on the pipeline in AEM with the content features and the language itself, which is established in two different models in marking for extraction and detection. Through experiments, this paper reveals the fact that an iB-CET oral test can be performed perfectly in the pipeline of spoken documents transformed into textual documents which can be automatically marked. According to historical records,in July of 1590 A.D.,a destructive earthquake occurred near Lintao county in Gansu Province,in which "… city walls and houses collapsed,and countless people and domestic animals w...According to historical records,in July of 1590 A.D.,a destructive earthquake occurred near Lintao county in Gansu Province,in which "… city walls and houses collapsed,and countless people and domestic animals were killed".In the same month,Binggou town in eastern Qinghai Province(now northeastern Ledu county),was also damaged by an earthquake.These two earthquakes were listed as two different cases in the published earthquake catalogues,recorded separately as the Lintao M_S5.5 earthquake with epicentral intensity Ⅶ in Gansu Province and the Ledu M_S5.0 earthquake with epicentral intensity Ⅵ in Qinghai Province.However,based on comprehensive analysis of research on historical records and field investigations,it is concluded in this paper that these two earthquakes could be the same one with magnitude 6.5 and epicentral intensity Ⅷ~Ⅸ.Its epicenter was in the Maxian Mt.,which is located in southeastern Yongjing and its seismogenic structure might be the mid-western segments of the north fringe fault zone of Maxian Mt.of Lanzhou.展开更多

Tomato has emerged as an emblematic model plants for fleshy fruit research and tomato fruit ripening is a complex and highly coordinated developmental process.The many physiology and biochemical processes associated w...Tomato has emerged as an emblematic model plants for fleshy fruit research and tomato fruit ripening is a complex and highly coordinated developmental process.The many physiology and biochemical processes associated with tomato fruit ripening require changes in the expression of hundreds to thousands of genes.Gene expression is regulated by transcriptional and post-transcriptional pathways,one of the recently discovered mechanisms in plants was small RNAs mediated gene silencing at post-transcriptional(PTGS) level.Intriguingly,several mi RNAs and endogenous si RNAs were revealed to be involved in the fruit ripening process which opened a new avenue in the field of fleshy fruit biology.This review compiled the most recent advances made in deciphering the regulation functions of mi RNAs and si RNAs in tomato fruit ripening.It also emphasized the new perspectives now possible in the small RNAs regulation research in tomato fruit ripening and senescence.展开更多

This paper deals with the values of these parameters in high-fidelity recording. METHODS: The cell lines Huh7, HepG2, SK-HEP1, SM-MC7721, Bel7402, QGY7703 and a...AIM: To investigated whether sall3 transcription was regulated by promoter CpG island hypermethylation in hepatocellular carcinoma (HCC). METHODS: The cell lines Huh7, HepG2, SK-HEP1, SM-MC7721, Bel7402, QGY7703 and a cohort of 38 HCC tissue specimens and corresponding nontumorous tissues were subjected to analysis for sall3 promoter CpG island methylation and mRNA transcription. sall3 promoter CpG island methylation levels were determined using the MassARRAY platform and mRNA transcription levels of the gene were detected by quantitative realtime polymerase chain reaction.RESULTS: The levels of sall3 mRNA were decreased by more than twofold in 33 of 38 tumor tissues compared to adjacent noncancerous tissues. Among these 33 tumor tissues with lower levels of sall3 mRNA, 24 showed higher levels of methylation. Based on these results, we hypothesized that the decrease in sall3 mRNA transcription level was likely due to promoter CpG island hypermethylation. Changes in sall3 mRNA transcription and promoter CpG island methylation were determined in the above six cell lines after treatment with 0, 0.1, 0.5 and 2.5 mmol 5-aza-2-deoxycytidine, a demethylating agent. Promoter CpG island methylation levels de- creased in a dose-dependent manner in all six cell lines, while the mRNA transcription level increased dose-dependently in Huh7, HepG2, SK-HEP1 and SMMC7721 cells and irregularly in Bel7402 and QGY7703 cells. CONCLUSION: These results indicated that promoter CpG island hypermethylation contributes to the downregulation of sall3 mRNA transcription in HCC.展开更多
From the early days of the moving image and its recordings of facts and events considered to be historical, followed by the consolidation of the classic narrative cinema grammar in the 1910s reaffirmed until today by ...From the early days of the moving image and its recordings of facts and events considered to be historical, followed by the consolidation of the classic narrative cinema grammar in the 1910s reaffirmed until today by a large part of the television production that turns to the past, without disregarding authorial aesthetic experiences produced especially since the 1920s, history has been present for over more than a century in several types of media. Movie theaters, people's homes and, nowadays, thanks to new media technology, any and every place are spaces for projecting historical narratives. They are both entertainment--by deploying strategies for constructing a "truth" about the past--and critical reflection, going against a belief in that possibility, in rendering explicit their nature as a language. Since all of these narratives presuppose an audience, a public, within different genres, styles and formats, with more realist overtones, more to the general public's taste, or anti-naturalist, in experiences for smaller audiences, it seems pertinent to discuss these issues considering that audiovisual narratives are powerful agents in constructing a memory of the past. Particularly in this text, we will examine how the most powerful communication enterprise in Brazil - Global Group - had construct a memory of the Brazilian military dictatorship (1964-1985) until 2016.展开更多
The calibration of paleoclimate proxies is one of the key problems in the study of paleoclimate at present. Historical documentary records of climate are suitable for calibration on dating and the climatic implication...The calibration of paleoclimate proxies is one of the key problems in the study of paleoclimate at present. Historical documentary records of climate are suitable for calibration on dating and the climatic implication of the proxy data in a climatological sense. A test calibration on correcting the Delingha tree ring precipitation series using Chinese historical documentary records shows that among the 44 extreme dry cases in 1401 1950 AD, 42 cases (or 95.5%) are believable. Thus the long series of Delingha rings-denoted precipitation is highly reliable. Another test to validate the monsoon intensity proxy data based on the Zhanjiang Huguangyan sediments using historical records indicates that the years of Lake Maar Ti content series-designated winter monsoon intensities are entirely opposite to historical documents- depicted years of harsh winters in 800-900 AD. As a result, serious doubt is raised about the climatic implication of this paleo-monsoon proxy series.展开更多
